The authors describe a method of treating scintigraphic data with which they can correctly compare images of a structure that were obtained in different conditions. This method comprises two steps which are fully computerized. During the first one known as the "registration" step and intended to make a comparison possible they maximise a new similarity criterion by a series of random trials in a five parameter space. During the second, which is the actual comparison, they use an appropriate statistical test to recognise those homologous pixels with a significantly different contents which will be the only ones retained to build up the final image. They give then two examples of what the method brings in the medical field which are the detection of adenomatous parathyroid glands and the follow-up of lung perfusion in case of embolism.  相似文献

The use of synthetic trisaccharides as acceptors led to the definition of five main (1----3)-alpha-L-fucosyltransferase activity patterns in human adult tissues: (I). Myeliod cells, granulocytes, monocytes, and lymphoblasts, transfer an alpha-L-fucopyranosyl group to O-3 of a 2-acetamido-2-deoxy-D-glucosyl residue of H blood-group Type 2 oligosaccharide [alpha-L-Fucp-(1----2)-beta-D-Galp-(1----4)-beta-D-GlcpNAc----R] with Mn2+ as activator. (II) Brain has the same acceptor specificity pattern as myeloid cells, but can also use Co2+ as activator. (III) Plasma and liver transfer an alpha-L-furopyranosyl group to H blood-group Type 2 and to sialyl-N-acetyllactosamine [alpha-NeuAc-(2----3)-beta-D-Galp-(1----4)-beta-D-GlcpNAc----R]. (IV) Intestine, gall bladder, kidney, and milk have the same activity as (III), but also transfer an alpha-L-fucopyranosyl group to O-4 of a 2-acetamido-2-deoxy-D-glucose residue of H blood-group Type 1 [alpha-L-Fucp-(1----2)-beta-D-Galp-(1----3)-beta-D-GlcpNAc----R] and sialyl Type 1 [alpha-NeuAc-(1----3)-beta-D-Galp-(1----3)-beta-D-GlcpNAc----R]. (V) Stomach mucosa is not able to use sialyl-N-acetyllactosamine, but can transfer an alpha-L-fucopyranosyl group to the other Type 1 and Type 2 acceptors. Unlike in adult tissue, a single myeloid-like pattern of (1----3)-alpha-L-fucosyltransferase activity was found at early stages of development in all tissues tested. This embryonic enzyme is later progressively replaced by enzymes or mixtures of enzymes having the corresponding adult patterns of enzyme expression. All lymphoblastoid cell lines and half of the tumor epithelial cell lines tested expressed the myeloid-like pattern of enzyme found in normal embryonic tissues. The remaining tumor epithelial cell lines expressed different forms of (1----3/4)-alpha-L-fucosyltransferase acceptor specificity patterns.  相似文献

A range of N-acetyllactosamine derivatives, which are modified by a wide range of functionalities at C-2(') and C-6, have been synthesised and the kinetic parameters of transfer catalysed by recombinant alpha-2,6-sialyltransferase and alpha-1,3-fucoyltransferase VI determined. Several of the chemical modifications led to selective modulate the activity the enzymes and offer promising lead compounds for the development of oligosaccharide primers for selective metabolic inhibition of oligosaccharide biosynthesis.  相似文献

Thyroglobulin (Tg), the thyroid hormone precursor, is sulfated both on tyrosines and on carbohydrates. We showed recently that sulfated tyrosines were involved in thyroid hormone synthesis. Moreover, we also reported that Tg sulfation is downregulated by thyrotropin (TSH), especially on tyrosines. This control may occur at each step in the sulfation process. In this paper, we studied the regulation of the concentration of cytosolic inorganic sulfate, the first substrate, in porcine thyroid cells stimulated by TSH with or without iodide. The amounts of cytosolic sulfate and the cytosolic volumes measured showed that the sulfate concentration depends only on cytosolic volume changes in response to TSH and iodide treatment. After the cells were labelled with [35S]-sulfate, the specific radioactivity (SRA) of cytosolic sulfate was determined. When cells were treated with only TSH, the concentration and SRA of cytosolic sulfate decreased by 30%, and by about 15% when cells were incubated with both TSH and iodide. TSH decreased more conspicuously the rate of [35S]-sulfate incorporation into Tg (by 57% without iodide, by 43% with iodide) than the concentration and SRA of cytosolic sulfate, while iodide altered these parameters to the same extent (15%). These findings suggest that TSH regulates other steps in the sulfation process, such as specific substrate and enzyme levels, while iodide controls mainly the sulfate concentration.  相似文献

IntroductionWhether diabetes mellitus increases the risk of acute kidney injury (AKI) during sepsis is controversial.ResultsFirst, we compared 451 patients with severe sepsis or septic shock and diabetes to 3,277 controls with severe sepsis or septic shock and without diabetes. Then, we compared 318 cases (with diabetes) to 746 matched controls (without diabetes). Diabetic patients did not have a higher frequency of AKI (hazard ratio [HR], 1.18; P = 0.05]) or RRT (HR, 1.09; P = 0.6). However, at discharge, diabetic patients with severe sepsis or septic shock who experienced acute kidney injury during the ICU stay and were discharged alive more often required RRT (9.5% vs. 4.8%; P = 0.02), had higher serum creatinine values (134 vs. 103 µmoL/L; P<0.001) and had less often recovered a creatinine level less than 1.25 fold the basal creatinine (41.1% vs. 60.5%; P<0.001).ConclusionsIn patients with severe sepsis or septic shock, diabetes is not associated with occurrence of AKI or need for RRT but is an independent risk factor for persistent renal dysfunction in patients who experience AKI during their ICU stay.  相似文献

Heparan sulfate (HS) glucosaminyl 3-O-sulfotranferases sulfate the C3-hydroxyl group of certain glucosamine residues on heparan sulfate. Six different 3-OST isoforms exist, each of which can sulfate very distinct glucosamine residues within the HS chain. Among these isoforms, 3-OST1 has been shown to play a role in generating ATIII-binding HS anticoagulants whereas 3-OST2, 3-OST3, 3-OST4 and 3OST-6 have been shown to play a vital role in generating gD-binding HS chains that permit the entry of herpes simplex virus type 1 into cells. 3-OST5 has been found to generate both ATIII- and gD-binding HS motifs. Previous studies have examined the substrate specificities of all the 3-OST isoforms using HS polysaccharides. However, very few studies have examined the contribution of the epimer configuration of neighboring uronic acid residues next to the target site to 3-OST action. In this study, we utilized a well-defined synthetic oligosaccharide library to examine the substrate specificity of 3-OST3a and compared it to 3-OST1. We found that both 3-OST1 and 3-OST3a preferentially sulfate the 6-O-sulfated, N-sulfoglucosamine when an adjacent iduronyl residue is located to its reducing side. On the other hand, 2-O-sulfation of this uronyl residue can inhibit the action of 3-OST3a on the target residue. The results reveal novel substrate sites for the enzyme actions of 3-OST3a. It is also evident that both these enzymes have promiscuous and overlapping actions that are differentially regulated by iduronyl 2-O-sulfation.  相似文献

ABCB4/MDR3, a member of the ABC superfamily, is an ATP-dependent phosphatidylcholine translocator expressed at the canalicular membrane of hepatocytes. Defects in the ABCB4 gene are associated with rare biliary diseases. It is essential to understand the mechanisms of its canalicular membrane expression in particular for the development of new therapies. The stability of several ABC transporters is regulated through their binding to PDZ (PSD95/DglA/ZO-1) domain-containing proteins. ABCB4 protein ends by the sequence glutamine-asparagine-leucine (QNL), which shows some similarity to PDZ-binding motifs. The aim of our study was to assess the potential role of the QNL motif on the surface expression of ABCB4 and to determine if PDZ domain-containing proteins are involved. We found that truncation of the QNL motif decreased the stability of ABCB4 in HepG2-transfected cells. The deleted mutant ABCB4-ΔQNL also displayed accelerated endocytosis. EBP50, a PDZ protein highly expressed in the liver, strongly colocalized and coimmunoprecipitated with ABCB4, and this interaction required the QNL motif. Down-regulation of EBP50 by siRNA or by expression of an EBP50 dominant-negative mutant caused a significant decrease in the level of ABCB4 protein expression, and in the amount of ABCB4 localized at the canalicular membrane. Interaction of ABCB4 with EBP50 through its PDZ-like motif plays a critical role in the regulation of ABCB4 expression and stability at the canalicular plasma membrane.  相似文献

Electron microscopy was performed on Candida albicans yeast after application of ribonuclease to clear the cytoplasmic background. In conjunction with Thiery's method of highlighting polysaccharide components, this clearing technique, which has not been used since 1959, enabled visualization of the nucleus, the mitochondria, the vacuolar system and another structure which seemed to be the Golgi apparatus.The invaginations of the plasmalemma membrane (or lomasomes) are highly developed and may be partially responsible for transporting material required for development of the cell wall, especially during budding.
Résumé L'aspect ultrastructural des levures Candida albicans est observé grace à l'action d'une ribonuclease dans le but d'éclaircir le fond cytoplasmique. Cette méthode non utilisée depuis 1959, associée au test de Thiery pour la mise en évidence des polysaccharides nous permet de déceler le noyau, les mitochondries, l'appareil vacuolaire et semble-t-il l'appareil de Golgi.Les invaginations du plasmalemme (ou lomasomes) particulièrement développées dans ces cellules pourraient jouer un rôle dans l'apport du matériel nécessaire à l'élaboration des parois notamment lors du bourgeonnement des levures.
