The structure of the pore is critical to understanding the molecular mechanisms underlying selective permeation and voltage-dependent gating of channels formed by the connexin gene family. Here, we describe a portion of the pore structure of unapposed hemichannels formed by a Cx32 chimera, Cx32*Cx43E1, in which the first extracellular loop (E1) of Cx32 is replaced with the E1 of Cx43. Cysteine substitutions of two residues, V38 and G45, located in the vicinity of the border of the first transmembrane (TM) domain (TM1) and E1 are shown to react with the thiol modification reagent, MTSEA–biotin-X, when the channel resides in the open state. Cysteine substitutions of flanking residues A40 and A43 do not react with MTSEA–biotin-X when the channel resides in the open state, but they react with dibromobimane when the unapposed hemichannels are closed by the voltage-dependent “loop-gating” mechanism. Cysteine substitutions of residues V37 and A39 do not appear to be modified in either state. Furthermore, we demonstrate that A43C channels form a high affinity Cd2+ site that locks the channel in the loop-gated closed state. Biochemical assays demonstrate that A43C can also form disulfide bonds when oocytes are cultured under conditions that favor channel closure. A40C channels are also sensitive to micromolar Cd2+ concentrations when closed by loop gating, but with substantially lower affinity than A43C. We propose that the voltage-dependent loop-gating mechanism for Cx32*Cx43E1 unapposed hemichannels involves a conformational change in the TM1/E1 region that involves a rotation of TM1 and an inward tilt of either each of the six connexin subunits or TM1 domains.  相似文献

【背景】主要组织相容性复合体(major histocompatibility complex,MHC)的多态性在很大程度上会影响生物医学实验的结果,而且特定的MHC-B等位基因与多种疾病的发展进程密切相关。食蟹猴(Macaca fascicularis,Mafa)是一种开展生物医学研究的重要实验动物,与人类相比,目前尚缺乏对食蟹猴Mafa-B等位基因的综合表征。【目的】获得全面的食蟹猴Mafa-B等位基因信息,鉴定Mafa-B等位基因共表达与进化关系。【方法】基于三代测序获得的食蟹猴MHC-B基因组信息,设计特异性引物扩增33只越南食蟹猴群体中的Mafa-B序列,并结合多种生物信息学方法进行分析。【结果】基于92个Mafa-B等位基因信息,鉴定了65个新的Mafa-B等位基因。其中,8个Mafa-B等位基因与其他地理来源的食蟹猴群体中已报道的序列相同,32个Mafa-B等位基因与其他猕猴物种中已报道的序列相同。此外,鉴定了7个高频Mafa-B谱系和7对共表达的Mafa-B等位基因,并检测到了一个潜在的重组事件。进化分析表明不同地理来源的食蟹猴群体Mafa-B序列具有很高的相似性。【结论】越南食蟹猴群体中共表达的Mafa-B等位基因经历了某些抗原的选择,不同地理来源的食蟹猴群体可能微调其Mafa-B序列以适应病原体的选择压力,本文为食蟹猴MHC遗传背景研究奠定了基础。  相似文献

A hexavalent chromium [Cr(VI)] reducing bacterial strain was isolated from chromium-containing slag. It was identified as Pannonibacter phragmitetus based on physiological, biochemical characteristics and 16S rRNA gene sequence analysis. This bacterium displayed great Cr(VI) reduction capability. The Cr(VI) could be completely removed in 24 h under anaerobic condition when the initial concentration was 1,917 mg L−1, with the maximum reduction rate of 562.8 mg L−1 h−1. The Cr(VI) reduction rate increased with the increase of Cr(VI) concentration. P. phragmitetus was able to use many carbon sources such as lactose, fructose, glucose, pyruvate, citrate, formate, lactate, NADPH and NADH as electron donors, among which the lactate had the greatest power to promote the reduction process. Zn2+, Cd2+ and Ni2+ inhibited, while Cu2+, Pb2+, Mn2+ and Co2+ stimulated the reduction. The optimum pH and temperature for reduction were 9.0 and 30 °C, respectively. The results indicated that this strain had great potential for application in the bioremediation of chromate-polluted soil and water systems.  相似文献

勐养保护区亚洲象微卫星位点筛选及种群遗传多样性分析   总被引:1,自引:1,他引:1  
本文以亚洲象肌肉样品提出的DNA为模板,从非洲象31个微卫星位点和5个已知亚洲象微卫星位点中筛选勐养亚洲象的微卫星位点,进而对在西双版纳勐养保护区3年采集到的191 份亚洲象粪便样品中提出的DNA进行特异性PCR扩增、基因分型、检测位点信息和遗传多样性分析.结果表明:36个位点中有14个位点能在亚洲象肌肉样品提出的DNA中成功扩增,且经测序证实为微卫星位点.其中9个多态位点能在185份粪便样品DNA中稳定扩增.勐养种群中,3个位点可能偏离Hardy Weubberg平衡,至少8个位点间无明显连锁存在,平均等位基因数3.78±1.72,平均期望杂和度0.32 ± 0.06,平均观察杂和度0.36±0.02,平均多态信息含量0.28 ,说明这9个位点适用于勐养亚洲象的遗传学研究.根据微卫星位点的杂合度和等位基因频率,相比于其他亚洲象种群,勐养亚洲象种群遗传多样性较低且等位基因频率具有特异性.  相似文献

以亚硝酸盐水溶液为研究对象,研究了辐照剂量、亚硝酸盐初始浓度、Vc和NaCl含量对亚硝酸盐降解效果的影响.结果表明,辐照能有效降低水溶液中亚硝酸盐含量,亚硝酸盐降解率与辐照剂量呈正相关,与初始浓度呈负相关,Vc对辐照降解亚硝酸盐具有协同作用,NaCl对辐照降解亚硝酸盐具有抑制作用.亚硝酸盐水溶液经辐照后,硝态氮和铵态氮均有一定的增加,当辐照剂量为10.55 kGy,初始浓度为100 mg· L-1时,硝态氮含量由14.857 mg·L-1增加到17.270 mg·L-1,铵态氮含量由0.013 mg·L-1增加到0.041 mg·L-1.  相似文献

Objective: This study explored the effect of circadian disorder on structure and function of neurons in hippocampus of mice. Methods: Forty male ICR mice were randomly divided into rhythm disorder group (RDG) and normal rhythm group (NRG). The RDG was treated with 3 h light/5 h dark and 5 h light/3 h dark light–dark (LD) cycle alternately. The normal rhythm group was treated with 12/12 h LD cycle. Electron microscope was used to detect the mouse hippocampal cell ultrastructure. Morris water maze was used to test mice cognitive function. The brain slice electric physiological techniques were used to detect synapses in the hippocampal Long-term potentiation (LTP) effect. Results: The time of through central area of RDG was less than that of NRG. The axoplasm in anterior-posterior membranes of synapses of RDG was dissolved and synaptic vesicles of RDG were decreased. Conclusion: Circadian rhythm disorder induced by irregular light–dark circle can lead to the structural damage of hippocampal neurons and damage the cognitive function in mice.  相似文献

The cyclin-dependent kinase (CDK) inhibitor p27(Kip1) (p27) is an important regulator of cell cycle progression controlling the transition from G to S-phase. Low p27 levels or accelerated p27 degradation correlate with excessive cell proliferation and poor prognosis in several forms of cancer. Phosphorylation of p27 at Thr187 by cyclin E-CDK2 is required to initiate the ubiquitination-proteasomal degradation of p27. Protecting p27 from ubiquitin-mediated proteasomal degradation may increase its potential in cancer gene therapy. Here we constructed a non-phosphorylatable, proteolysis-resistant p27 mutant containing a Thr187-to-Ala substitution (T187A) which is not degraded by ubiquitin-mediated proteasome pathway, and compared its effects on cell growth, cell-cycle control, and apoptosis with those of wild-type p27. In muristerone A-inducible cell lines overexpressing wild-type or mutant p27, the p27 mutant was more resistant to proteolysis in vivo and more potent in inducing cell-cycle arrest and other growth-inhibitory effects such as apoptosis. Transduction of p27(T187A) in breast cancer cells with a doxycycline-regulated adenovirus led to greater inhibition of proliferation, more extensive apoptosis, with a markedly reduced protein levels of cyclin E and increased accumulation of cyclin D1, compared with wild-type p27. These findings support the potential effectiveness of a degradation-resistant form of p27 in breast cancer gene therapy.  相似文献
