A novel endophytic actinomycete, designated strain NEAU-J3T, was isolated from soybean root (Glycine max (L.) Merr) and characterized using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences suggested that strain NEAU-J3T fell within the family Micromonosporaceae. The strain was observed to form an extensively branched substrate mycelium, which carried non-motile oval spores with a smooth surface. The cell walls of strain NEAU-J3T were determined to contain meso-diaminopimelic acid and galactose, ribose and glucose were detected as whole-cell sugars. The major menaquinones were determined to be MK-9(H4) and MK-9(H6). The phospholipids detected were phosphatidylcholine and phosphatidylethanolamine. The major cellular fatty acids were determined to be C16:0, C18:1 ω9c, C18:0, C17:0, C17:1 ω7c, anteiso-C17:0, C16:1 ω7c and C15:0. The DNA G + C content was 62.5 mol%. On the basis of the morphological and chemotaxonomic characteristics, phylogenetic analysis and characteristic patterns of 16S rRNA gene signature nucleotides, strain NEAU-J3T is considered to represent a novel species of a new genus within the family Micromonosporaceae, for which the name Wangella harbinensis gen. nov., sp. nov. is proposed. This is the first report on the determination of proteins with 3-hydroxy-4-(2-hydroxy-4-sulfo-1-naphthalenyl)azo (Cal-Red) by Rayleigh light scattering (RLS). At pH 4.07, the weak RLS of Cal-Red can be enhanced greatly by the addition of proteins. On this basis, the reaction of Cal-Red and proteins was studied. A new quantitative determination method for proteins has been developed. This method is very sensitive (0.45-36.9 microgml(-1) for bovine serum albumen (BSA)), rapid (<2min), simple (one step), and tolerant of most interfering substances. The maximum binding number of Cal-Red to BSA was 143 and the binding constant was 4.1x10(6)mol(-1)L. Four samples of total protein in human serum were determined and the maximum relative error is no more than 3%. 相似文献

The p75(NTR) neurotrophin receptor has been implicated in multiple biological and pathological processes. While significant advances have recently been made in understanding the physiologic role of p75(NTR) , many details and aspects remain to be determined. This is in part because the two existing knockout mouse models (Exons 3 or 4 deleted, respectively), both display features that defy definitive conclusions. Here we describe the generation of mice that carry a conditional p75(NTR) (p75(NTR-FX) ) allele made by flanking Exons 4-6, which encode the transmembrane and all cytoplasmic domains, by loxP sites. To validate this novel conditional allele, both neural crest-specific p75(NTR) /Wnt1-Cre mutants and conventional p75(NTR) null mutants were generated. Both mutants displayed abnormal hind limb reflexes, implying that loss of p75(NTR) in neural crest-derived cells causes a peripheral neuropathy similar to that seen in conventional p75(NTR) mutants. This novel conditional p75(NTR) allele will offer new opportunities to investigate the role of p75(NTR) in specific tissues and cells. 相似文献

重度退化的喀斯特天坑草地物种多样性及群落稳定性——以云南沾益退化天坑为例 总被引:3,自引:0,他引:3
喀斯特天坑是21世纪初命名的宏大地表负地形,其隐域生境为动植物的生长和繁衍提供了良好的条件,近几年来,随着天坑价值的深入挖掘,天坑独特生境特征及其植物群落逐渐得到重视,但现有研究较多关注原生天坑及轻度退化天坑,对退化程度较高且可进入性较强的退化天坑的研究仍相对薄弱。云南大竹菁天坑属于典型的重度退化天坑,可进入性较强,坑底以草地植物群落为主,将其作为退化天坑的代表性区域开展天坑内外植物群落的特征差异及植物群落的稳定程度研究,有利于补充系列天坑现有的理论知识体系,亦能为广大喀斯特区域的生态恢复提供一定的借鉴意义。采用野外采样及数据分析相结合的方法,从定性及定量的角度研究大竹菁退化天坑植被群落特征以及植物群落的稳定程度,研究结果表明:1)退化天坑内草地群落现有14科35属35种,坑外14科30属30种,坑内外物种组成相对多样,两者优势种差异较大,仅一种共优种;2)退化天坑生态系统较为脆弱,坑内外植物群落多样性总体偏低,坑外植物多样性高于坑内植物多样性,但差异不显著,而坑外均匀度却显著高于坑内均匀度;3)坑内外植物群落处于极不相似水平,结构分异明显,群落之间具有较大的生境差异性;4)坑内植物群落种间总体呈正联结性,种间联结较松散,具有一定的独立性,处于正向演替阶段,而坑外呈负关联性,群落较不稳定;5)采用稳定性分析进一步阐释得出,坑内植物群落离稳定状态仍有一定差距,说明受到人为干扰的重度退化天坑生境仍较脆弱。 相似文献

基于Landsat系列数据的盐分指数和植被指数对土壤盐度变异性的响应分析——以新疆天山南北典型绿洲为例 总被引:3,自引:0,他引:3
基于不同地理区域,借助目前已有或者构建新的盐分和植被指数定量评估研究区的土壤盐度状况。但多数指数并未在盐渍化较为严重的中国新疆地区进行系统性对比分析。因此,以新疆阜北地区(采样数=37),玛纳斯河绿洲(采样数=68)和渭干河-库车河绿洲(采样数=38)为研究区,以灌区农田和盐渍地采样数据和Landsat TM/ETM+/OLI为数据源,利用线性模型和多个非线性模型(10个)测试上述指数(14个指数)和原始波段对于研究区土壤盐度的敏感性。结果显示,阜北地区基于遥感获取的扩展的增强型植被指数Extented Enhanced Vegetation Index(EEVI)在全样本和部分样本(盐渍化样本,土壤盐度0.3%)两种模式下(0—10cm),较其他指数和波段而言较为敏感。在全样本和部分样本(土壤饱和溶液电导率2ds/m)两种模式下,与玛纳斯流域各层土壤盐度最为敏感的为band 2,部分样本模式下土壤盐度变异性显著性探测最大下探深度为30cm。渭干河-库车河绿洲全样本模式下,最大土壤盐度变异性显著性探测深度为40cm,0—10cm和10—20cm深度表现最为敏感的是土壤盐分指数SI-T,20—40cm深度则为植被指数TGDVI。部分样本下(土壤饱和溶液电导率2ds/m),0—10cm深度最为敏感的为band5,10—20cm深度最为敏感的为TGDVI,20—40cm深度则为EEVI。其他指数因地理环境的差异性(气候,土壤盐分类型,土壤类型,采样时间),与土壤盐度之间并未达到显著性(sig=0.05或者0.01)的水平。以上结果只是初步结论,但也暗示其中的某些指数在本区具有一定土壤盐度的识别潜力。此外,由于土壤本身的复杂性,需要采集更多的样本以深入分析不同盐度等级下上述指数的具体表现。 相似文献

LIN9 functions to regulate cell mitotic process.Dysregulation of LIN9 expression is associated with development of human cancers.In this study we assessed the association of LIN9 expression with paclitaxel resistance and clarified the underlying mechanisms for the first time.LIN9 expression in breast cancer tissues was retrieved from publicly available online databases and statistically analyzed.Human TNBC cell lines MDA-MB-231 and MDA-MB-468 and their corresponding paclitaxelresistant sublines 231PTX and 468PTX were used to assess the expression of LIN9 by qRT-PCR and Western blot,cell growth by cell counting,cell viability by MTS assay,and cell apoptosis by flow cytometry.The data showed that high LIN9 expression in breast cancer patients receiving chemotherapy was related to poor overall survival (OS).LIN9 expression was upregulated in paclitaxel-resistant TNBC cells compared to their parental cells.Knockdown of LIN9 or treatment of paclitaxel-resistant TNBC cells with a bromo-and extra-terminal domain inhibitor (BETi) JQ1 which also decreased LIN9 expression enhanced the sensitivity of paclitaxel-resistant TNBC cells to paclitaxel.Mechanistically,decreased LIN9 in resistant cell lines reduced tumor cell viability,promoted multinucleated cells formation and induced tumor cell apoptosis,potentially by directly regulating microtubule-binding protein CCSAP.In conclusion,high LIN9 expression contributed to poor clinical outcomes and paclitaxel resistance in TNBC and BETi,targeting LIN9 expression,could be a reversible drug for PTX-resistant TNBC patients. 相似文献
