Tonic activity of sympathetic nerve fibers of the tibial and peroneal nerves was investigated in rabbits anesthetized with urethane by the multichannel coherent recording technique. Activity of stochastic character was shown to predominate in the tonic activity of these fibers. A component of activity with the frequency of the heart beat also is frequently observed, but the power of this component is never more than half the total power of activity. Activity with a frequency of 10 Hz, observed by other workers, and also modulation of tonic activity in the rhythm of respiration were not recorded in these experiments. Slow changes in the power of activity from zero to a level several times above average were observed when the blood pressure was stable.A. A. Effects of d-sparteine (d-SP), a ganglionic blocking agent, on membrane currents evoked by iontophoretic applications of acetylcholine to rat superior cervical ganglion neurons, were studied using a whole-cell patch-clamp recording technique. Blocking effects of d-SP were enhanced by membrane hyperpolarization to potentials more negative than –50 mV. Analysis of the d-SP effect on the dose—response relationship suggests that d-SP at concentrations of 0.5–5.0 µM exerts both voltage-independent and voltage-dependent competitive actions on nicotinic acetylcholine receptors. No use-dependence of the d-SP-induced blockade was found using paired ACh applications at interpulse intervals longer than 0.5 sec. Inhibitory constantK i estimated by the Dixon method was equal to 0.62±0.15 and 0.28±0.08 µM at membrane potential levels –30 and –90 mV, respectively. These characteristics of the d-SP blocking effects are compatible with a voltage-dependent competitive blocking mechanism. On neurons of the superior cervical ganglion of 3-week-old rats, we studied the mechanism underlying the blocking effect of mecamylamine on transmembrane currents evoked by iontophoretic application of acetylcholine (ACh currents); these currents were recorded with the use of a patch-clamp technique in the whole-cell configuration. The IC50 of the above agent equaled (2.7 ± 0.3) · 10-10 M. The blocking effect of mecamylamine on ACh current did not depend on the membrane potential and decreased with rise in the concentration of the drug. Thus, a competitive blocking mechanism mostly underlies the above phenomenon.  相似文献

In experiments on rabbits anesthetized with urethane, we recorded spontaneous sympathetic activity of single nerve fibers. The activity was recorded from fine filaments dissected from the cervical sympathetic nerve trunk (CSNT). Conduction velocities in CSNT fibers and action-potential amplitude of single B-fibers were measured during recording of whole nerve trunk. Spectral analysis of the activity recorded was carried out. It was shown that unmyelinated CSNT fibers are under stronger baroreceptor control than myelinated fibers. Also, a periodic component of 2–3 Hz, noted by a number of authors after denervation of baroreceptors, was detected in intact animals along with a rhythm at the pulse rate. This component was found equally often in the activity of B- and C-fibers. Cross-correlation analysis of the simultaneously recorded activities of 24 pairs of CSNT single fibers was carried out. A correlation due to synchronization of the activities of pairs of fibers with a physiological rhythm (respiration, pulse) was found in five pairs. The correlation in one pair suggests the presence of a common excitatory synaptic input into two distinct preganglionic neurons.A. A. In experiments on rabbits anesthetized with urethane, we made multichannel coherent recordings of spontaneous activity and evoked activity of preganglionic sympathetic fibers in peripheral nerves of a leg and unmyelinated preganglionic fibers of the cervical sympathetic nerve trunk (CSNT). The results of a spectral analysis of the activity recorded and a correlational analysis of the activity and changes in arterial pressure allow us to conclude that preganglionic unmyelinated fibers of the CSNT are primarily vasoconstrictive and are under stronger baroreceptor control than vasoconstrictive fibers going to the skin and limb muscles. This is possibly due to differences in the functions of the innvervated organs. The latent period of the change in arterial pressure evoked by a change in the activity of vasoconstrictor fibers equals 4–10 sec.A. A. Using intracellular recording, we studied the effects of lead ions on the synaptic responses of neurons of the superior cervical ganglion (SCG) of the rat. Three groups of SCG neurons were found; they differed from each other in directions of the changes evoked by Pb2+ applications and in their sensitivity to this agent. In two groups of the phasic SCG neurons, Pb2+ decreased the amplitude of EPSP evoked by stimulation of the cervical sympathetic nerve with EC50 of (2.2 ± 0.2) · 10-6 M and (3.55 ± 0.29) · 10-5 M, respectively. In the tonic neurons, 2 · 10-6 M Pb2+ increased the EPSP amplitude by 27 ± 2%, on average, as compared with the control values. We postulate that the specificity of responses of these three groups of SCG neurons to the action of Pb2+ is determined by the different subunit composition of the nicotinic cholinoreceptors in the membrane of these cells.  相似文献
