The occurrence of functional chloroplasts in internal stem tissues and their distribution profiles in 20 woody species have been investigated. Chloroplasts were identified from the red chlorophyll auto-fluorescence using epi-fluorescence microscopy. Chloroplasts were detected in the cortex of all species examined, in the xylem rays of 19 and in the perimedullar and the pith cells of 16 out of the 20 investigated species. Chloroplast containing cell clusters in the pith were identified in some species. In addition, we report on the semi-quantitative distribution of chlorophylls in various internal stem tissues. Chlorophyll level was estimated by reflectance measurements at specific wave bands. Although decreasing chlorophyll gradients from cortex to pith were observed in half of the species, chlorophyll distribution in the remaining species was irregular with occasionally high levels in the pith. According to our data, chlorophyll occurrence in stem internal tissues is quite widespread, even in the light remote, deeply shaded central compartments like pith, provided that corresponding cells are viable. The species-specific tissue distribution of chlorophyll levels may be used to select suitable plants to investigate further this neglected area of photosynthesis research.  相似文献

The fine structure and photosynthetic cost of structural leaf variegation   总被引:1,自引:0,他引:1  
The leaves of some plants display an optical patchiness on their upper side, displaying light- and dark-green areas with high and low reflectance, respectively. In this investigation, we studied the fine structure of the corresponding sectors and we asked whether the lost reflected light entails a photosynthetic cost to these leaves. Four species, i.e. Arum italicum, Ranunculus ficaria, Cyclamen hederifolium and Cyclamen persicum were investigated. Scanning electron microscope examination revealed that epidermal cells of light-green sectors of all species are more bulgy than corresponding cells of neighboring dark-green leaf sectors. The comparative anatomical study revealed that (i) epidermis thickness of the light-green areas and the number of mesophyll cell layers does not differ from those of the adjacent dark-green leaf sectors and (ii) palisade cells of light-green sectors are slightly larger and more loosely arranged, allowing a much higher percentage of intercellular air spaces. The latter histological feature seems to provide the structural basis for the different optical properties between the two leaf sectors. Contrary to expectations, net photosynthetic rates (expressed on a leaf area basis) were similar in the light-green and the dark-green areas of the two cyclamen species. Yet, in C. persicum net photosynthesis was higher in the light-green areas, if expressed on a dry mass basis. The small size of the light-green spots in the rest of the test plants precluded CO2 assimilation measurements, yet maximum linear photosynthetic electron transport rates displayed no differences between the two sectors in all plants. Thus, the assumption of a photosynthetic cost in the light-green areas was not confirmed. On the contrary, a higher construction cost was evident in the dark-green areas of three species, displaying a significantly higher specific leaf mass, without any photosynthetic benefit. The results on net photosynthesis were compatible with leaf optical properties and pigment levels. Thus, in spite of the considerably higher reflectance of the light-green areas and their lower (yet normal for a green leaf) chlorophyll levels, corresponding differences in absorptance were slight. In addition, dry mass-based pigment contents in dark-green areas were higher, while chlorophyll a/b (in two species) and carotenoid/chlorophyll ratios (in three species) were lower, pointing to a shade adaptation in these sectors. We conclude that in variegated leaves of this kind, dark-green areas are more costly to build and probably less photosynthetically active. We argue that the high pigment contents of dark-green areas establish steep light gradients in the corresponding mesophyll, rendering deeper chloroplast layers more shade adapted.  相似文献

The possible mechanism(s) by which supplemental UV-B radiation alleviates the adverse effects of summer drought in Mediterranean pines (Petropoulou et al. 1995) were investigated with seedlings of Pinus pinea. Plants received ambient or ambient plus supplemental UV-B radiation (biologically equivalent to a 15% ozone depletion over Patras, 38.3° N, 29.1° E) and natural precipitation or additional irrigation. Treatments started on 1 February, 1994 and lasted up to the end of the dry period (29 September). In well-watered plants, UV-B radiation had no influence on photosystem II photochemical efficiency and biomass accumulation. Water stressed plants suffered from needle loss and reduced photosystem II photochemical efficiency during the summer. These symptoms, however, were less pronounced in plants receiving supplemental UV-B radiation, resulting in higher total biomass at plant harvest. Laboratory tests showed that enhanced UV-B radiation did not improve the tolerance of photosystem II against drought, high light, high temperature and oxidative stress. Enhanced UV-B radiation, however, improved the water economy of water stressed plants, as judged by measurements of needle relative water content. In addition, it caused an almost two-fold increase of cuticle thickness. No such UV-B radiation effects were observed in well-watered pines. The results indicate that the combination of water stress and UV-B radiation may trigger specific responses, enabling the plants to avoid excessive water loss and, thereby, maintain a more efficient photosynthetic apparatus during the summer. The extent of this apparently positive UV-B radiation effect would depend on the amount of summer precipitation. The phytochrome-mediated spore germination of the fern Dryopteris filix-mas was studied under reduced water content. Spores were sown on solutions containing polyethylene glycol (PEG), sorbitol, or mannitol as osmotically active substances, yielding water potential values (ψ) between ? 500 and ? 3500 kPa for most of the experiments. Our main results can be summarized as follows:
  • 1 The germination of the spores is completely blocked by ψ ≤ ?1200 kPa (≥ 300 g/l PEG).
  • 2 Reduced water content of the spores, at least up to 300 g/l PEG, does not impair the phototransformation of phytochrome from its inactive form Pr to its active form Pfr.
  • 3 The inhibiting effect of the osmoticum is phase-specific: the reduced water content of the spores impairs the coupling of the phytochrome system to the transduction chain. Under these conditions, Pfr is found to be stable for several days.
  • 4 In addition, the reduced water potential acts on the terminal response, although differently on different parameters of germination: Whereas formation of the rhizoid is completely blocked by 180 g/1 PEG, chlorophyll formation requires 300 g/1 PEG to become slightly retarded.

Alkaline phosphatase (ALP) is expressed in 3T3-L1 preadipocytes, and its activity increases during adipogenesis. The purpose of this study was to determine whether ALP activity could be used as a measure of intracellular lipid accumulation in human preadipocytes and 3T3-L1 cells and which of the factors that induce adipogenesis are responsible for stimulating ALP activity. Adipogenesis was initiated in 3T3-L1 cells by incubation with differentiation medium containing insulin, dexamethasone, and 3-isobutyl-1-methylxanthine. The effect of leaving out each of the differentiation medium components was studied. Adipogenesis was also assessed in human preadipocytes and 3T3-L1 cells in the presence of the ALP inhibitor histidine. ALP activity was measured using an automated colorimetric assay and intracellular lipid accumulation was measured using the lipid-specific dye oil red O. Removal of insulin or dexamethasone from the differentiation medium had little effect on either ALP activity or lipid accumulation in 3T3-L1 cells, while removal of IBMX blocked both. Histidine inhibited ALP activity and adipogenesis in human preadipocytes and 3T3-L1 cells. Pearson univariate correlation analysis demonstrated strong correlations between ALP activity and lipid accumulation in human preadipocytes (r=0.78, n=69) and in 3T3-L1 cells (r=0.92, n=27). These data suggest that ALP and fat storage are tightly linked during preadipocyte maturation and that the measurement of ALP activity may be a novel technique for the quantification of intracellular lipid accumulation that is more sensitive and rapid than currently used methods.  相似文献

Reflectance indices are frequently used for the nondestructive assessment of leaf chemistry, especially pigment content, in environmental or developmental studies. Since reflectance spectra are influenced by trichome density, and trichome density displays a considerable phenotypic plasticity, we asked whether this structural parameter could be a source of variation in the values of the most commonly used indices. Trichome density was manipulated in detached leaves of three species having either peltate (Olea europaea and Elaeagnus angustifolius) or tubular (Populus alba) trichomes by successive removal of hairs. After each dehairing step, trichome density was determined by light or scanning electron microscopy and reflectance spectra were obtained with a diode-array spectrometer. Although species-specific differences were evident, most of the indices were considerably affected even at low trichome densities. In general, the less-affected indices were those using wavebands within the visible spectral region. The index that could be safely used even at very high hair densities in all species was the red edge index (lambda(RE)) for chlorophyll. The results indicate that changes in reflectance indices should be interpreted cautiously when concurrent changes in trichome density are suspected. In this case, the red edge for chlorophyll content may be the index of choice.  相似文献

The structure and development of Myrtus communis L. secretory cavities has been studied in young and expanded leaves, using light and scanning electron microscope. Secretory cavities are continuously formed during leaf development, but in mature leaves the rhythm of their appearance shows steep decrease. Each secretory cavity is developed from a single epidermal cell, which undergoes a periclinal division followed by anticlinal and several oblique cell divisions. The lumen of the secretory cavity is initiated by cell wall separation, i.e., schizogenously. The secretory cells line the cavity, where the secreted material is collected. Secretory cavities are covered by modified epidermal cells, which do not seem to form any special aperture. Essential oils seem to be discharged after mechanical treatment of the leaf.  相似文献

Herein, we report the case of a 28-year old woman clinically presenting with unclear weight gain over the last years. The patient displayed facial and neck edema in combination with unobtrusive striae distensae. Endocrinological examinations led to the diagnosis of Cushing's disease. Neuroradiological examination revealed an intrasellar tumor mass of 7 mm in diameter. Subsequently, transsphenoidal tumor resection was performed. Histological and immunohistochemical investigations revealed a pituitary gland adenoma showing a biphasic tumor growth pattern with two morphologically different tumor areas producing ACTH and prolactin respectively. Co-expression of ACTH and prolactin is exceedingly rare in pituitary adenoma. To our surprise, both tumor areas exhibited features of atypia consisting in elevated MIB-1 proliferation index in the ACTH-producing portion as well as p53 expression selectively in the prolactin-producing tumor parts. To our knowledge, this is the first case of an ACTH- and prolactin-producing pituitary gland adenoma exhibiting biphasic features of atypia.  相似文献
