Lentil root statoliths reach a stable state in microgravity   总被引:3,自引:0,他引:3  
 The kinetics of the movement of statoliths in gravity-perceiving root cap cells of Lens culinaris L. and the force responsible for it have been analysed under 1 g and under microgravity conditions (S/MM-03 mission of Spacehab 1996). At the beginning of the experiment in space, the amyloplasts were grouped at the distal pole of the statocytes by a root-tip-directed 1-g centrifugal acceleration. The seedlings were then placed in microgravity for increasing periods of time (13, 29, 46 or 122 min) and chemically fixed. During the first 29 min of microgravity there were local displacements (mean velocity: 0.154 μm min−1) of some amyloplasts (first at the front of the group and then at the rear). Nevertheless, the group of amyloplasts tended to reconstitute. After 122 min in microgravity the bulk of amyloplasts had almost reached the proximal pole where further movement was blocked by the nucleus. After a longer period in microgravity (4 h; experiment carried out 1994 during the IML 2 mission) the statoliths reached a stable position due to the fact that they were stopped by the nucleus. The position was similar to that observed in roots grown continuously in microgravity. Treatment with cytochalasin D (CD) did not stop the movement of the amyloplasts but slowed down the velocity of their displacement (0.019 μm min−1). Initial movement patterns were the same as in control roots in water. Comparisons of mean velocities of amyloplast movements in roots in space and in inverted roots on earth showed that the force responsible for the movement in microgravity (Fc) was about 86% less (Fc = 0.016 pN) than the gravity force (Fg = 0.11 pN). Treatment with CD reduced Fc by two-thirds. The apparent viscosity of the statocyte cytoplasm was found to be 1 Pa s or 3.3 Pa s for control roots or CD treated roots, respectively. Brownian motion or elastic forces due to endoplasmic reticulum membranes do not cause the movement of the amyloplasts in microgravity. Up to 70% regenerating hypocotyls provided with 5 to 20 buds were obtained on MS medium containing 0.01 or 1 mg l-1 NAA and 0.2 mg l-1 BA or 0.2 mg l-1 BA and 0.2 mg l-1 TDZ. The ability to regenerate buds was correlated with the presence of oil glands at a stage in germination when oil secretion was not yet occurring. The regeneration of shoot meristems took place from the cells involved in the differentiation of these glands. Such glands could also appear during callus redifferentiation, giving rise to indirect regeneration. Rooting of the regenerants was efficient using a two-step procedure of induction under darkness in the presence of 3 mg l-1 IBA, followed by root development on medium devoid of growth regulators under a 16-h photoperiod, the medium being solidified with Gelrite. Regenerated plants showed no phenotypic alterations. Nuclear DNA contents in mother-plant material and regenerants were analysed using flow cytometry. There was no evidence of polyploidy in any of the samples, indicating the absence of polyploidisation during cell differentiation and under in vitro conditions. No regeneration was obtained from leaf or stem explants from micropropagated plantlets. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Plant life strategies differ radically from those of most animals. Plants are not motile, and can only face stress by developing appropriate physiological responses. In addition, many developmental decisions take place during post-embryonic life in plants, whereas vertebrate and invertebrate development is nearly complete by the time of birth. For instance, while the germ line is typically set aside early during embryogenesis in animals, plants produce gametes from stem cell populations that were previously used for the vegetative growth of shoots. Nevertheless, plants and animals have similar nuclear organization, chromatin constitution and gene content, which raises the question as to whether or not fundamental differences in the use of genetic information underlie their distinct life strategies. More specifically, we would like to know if chromatin and the epigenetically defined, heritable cell fates that it can confer play comparable roles in plants and animals. Here we review our current knowledge on chromatin-mediated epigenetic processes in plants. Based on available evidence, we argue that epigenetic regulation of gene expression plays a relatively minor role in plants compared to mammals. Conversely, plants appear to be more prone than other multicellular organisms to the induction of chromatin-based, epigenetically modified gene activity states that can be transmitted over many generations. These so-called "epimutations" may therefore represent a significant proportion of the natural genetic variation seen in plants. In humans, epimutations are frequently observed in cancers, and given their metastable nature, they could also play an important role in familial disorders that do not demonstrate clear Mendelian inheritance.  相似文献

Eukaryotic and archaeal DRG factors are highly conserved proteins with characteristic GTPase motifs. This suggests their implication in a central biological process, which has so far escaped detection. We show here that the two Saccharomyces cerevisiae DRGs form distinct complexes, RBG1 and RBG2, and that the former co-fractionate with translating ribosomes. A genetic screen for triple synthetic interaction demonstrates that yeast DRGs have redundant function with Slh1, a putative RNA helicase also associating with translating ribosomes. Translation and cell growth are severely impaired in a triple mutant lacking both yeast DRGs and Slh1, but not in double mutants. This new genetic assay allowed us to characterize the roles of conserved motifs present in these proteins for efficient translation and/or association with ribosomes. Altogether, our results demonstrate for the first time a direct role of the highly conserved DRG factors in translation and indicate that this function is redundantly shared by three factors. Furthermore, our data suggest that important cellular processes are highly buffered against external perturbation and, consequently, that redundantly acting factors may escape detection in current high-throughput binary genetic interaction screens.  相似文献

 Hairy root clones were established from carrot root discs inoculated with an agropine-type strain of Agrobacterium rhizogenes A4 harbouring the gus reporter gene on the TL-DNA. The clones were periodically examined for their phenotypic characteristics and for their ability to express the gus gene, to produce opines and to grow in the presence of NAM. The presence of the gus gene in the roots was confirmed by Southern blot hybridisation. The clones displayed various morphologies which were generally not correlated with the transformation events, and they were highly unstable throughout the successive subcultures, both for their phenotype and for their ability to express the transgenes. Reversible inactivation of the gus gene expression was associated with a high gus copy number. This could have some consequences for fundamental studies and practical uses of hairy roots. Lentil root statocytes show a strict structural polarity of their organelles with respect to the g vector. These cells are involved in the perception of gravity and are responsible for the orientation of the root. Actin filaments take part in the positioning of their organelles and could also be involved in the transduction of the gravitropic signal. A pre-embedding immunogold silver technique was carried out with a monoclonal antibody in order to study the distribution of actin cytoskeleton in the statocytes at the electron microscopic level. Some areas were never labelled (cell wall, vacuole, nucleoplasm, mitochondria, starch grains of the amyloplasts) or very slightly labelled (stroma of the amyloplasts). The labelling was scattered in the cytoplasm always close to, or on the nuclear and amyloplast envelopes and the tonoplast. Associations of 2 to 6 dots in file were observed, but these short files were not oriented in one preferential direction. They corresponded to a maximum distance of 0.9 micron. This work demonstrated that each statocyte organelle was enmeshed in an actin web of short filaments arranged in different ways. The images obtained by rhodaminephalloidin staining were in accordance with those of immunogold labelling. The diffuse fluorescence of the cytoplasm could be explained by the fact that the meshes of the web should be narrow. The vicinity of actin and of the amyloplasts envelope could account for the movement of these organelles that was observed in spatial microgravity.  相似文献

Using the distribution patterns of benthic insects (198 species) and fishes ( 29 species) from 11 tributaries and the main channel of the Doubs River drainage basin (French Jura), the authors have tried to establish whether there is an organization of species into discrete, identifiable communities. Principal Component Analysis (PCA) was used to identify whether a continuum existed, and also used to select 50 least-disturbed sites, which were used to define a theoretical watercourse. The density classes of each species were projected on this longitudinal gradient and each species response was characterised by two typological traits: its typological preferendum (tp) and its typological amplitude (ta),thus creating a synthesis of ecological characteristics. In the typological index given in the appendix, the 210 species which form a biological templet are listed in alphabetical order with their tp and ta values. These typological species traits are useful contributions to a database for running waters biomonitoring.  相似文献

All‐trans‐retinal (atRAL) is a highly reactive carbonyl specie, known for its reactivity on cellular phosphatidylethanolamine in photoreceptor. It is generated by photoisomerization of 11‐cis‐retinal chromophore linked to opsin by the Schiff's base reaction. In ABCA4‐associated autosomal recessive Stargardt macular dystrophy, atRAL results in carbonyl and oxidative stress, which leads to bisretinoid A2E, accumulation in the retinal pigment epithelium (RPE). This A2E‐accumulation presents as lipofuscin fluorescent pigment, and its photooxidation causes subsequent damage. Here we describe protection against a lethal dose of atRAL in both photoreceptors and RPE in primary cultures by a lipidic polyphenol derivative, an isopropyl‐phloroglucinol linked to DHA, referred to as IP‐DHA. Next, we addressed the cellular and molecular defence mechanisms in commonly used human ARPE‐19 cells. We determined that both polyunsaturated fatty acid and isopropyl substituents bond to phloroglucinol are essential to confer the highest protection. IP‐DHA responds rapidly against the toxicity of atRAL and its protective effect persists. This healthy effect of IP‐DHA applies to the mitochondrial respiration. IP‐DHA also rescues RPE cells subjected to the toxic effects of A2E after blue light exposure. Together, our findings suggest that the beneficial role of IP‐DHA in retinal cells involves both anti‐carbonyl and anti‐oxidative capacities.  相似文献
