A lack of oxygen is classically described as a major cause of impaired wound healing in diabetic patients. Even if the role of oxygen in the wound healing process is well recognized, measurement of oxygen levels in a wound remains challenging. The purpose of the present study was to assess the value of electron paramagnetic resonance (EPR) oximetry to monitor pO2 in wounds during the healing process in diabetic mouse models. Kinetics of wound closure were carried out in streptozotocin (STZ)-treated and db/db mice. The pO2 was followed repeatedly during the healing process by 1 GHz EPR spectroscopy with lithium phthalocyanine (LiPc) crystals used as oxygen sensor in two different wound models: a full-thickness excisional skin wound and a pedicled skin flap. Wound closure kinetics were dramatically slower in 12-week-old db/db compared to control (db/+) mice, whereas kinetics were not statistically different in STZ-treated compared to control mice. At the center of excisional wounds, measurements were highly influenced by atmospheric oxygen early in the healing process. In pedicled flaps, hypoxia was observed early after wounding. While reoxygenation occurred over time in db/+ mice, hypoxia was prolonged in the diabetic db/db model. This observation was consistent with impaired healing and microangiopathies observed using intravital microscopy. In conclusion, EPR oximetry using LiPc crystals as the oxygen sensor is an appropriate technique to follow wound oxygenation in acute and chronic wounds, in normal and diabetic animals. Nevertheless, the technique is limited for measurements in pedicled skin flaps and cannot be applied to excisional wounds in which diffusion of atmospheric oxygen significantly affects the measurements. 相似文献

We introduce an analytical model, the Wald analytical long-distance dispersal (WALD) model, for estimating dispersal kernels of wind-dispersed seeds and their escape probability from the canopy. The model is based on simplifications to well-established three-dimensional Lagrangian stochastic approaches for turbulent scalar transport resulting in a two-parameter Wald (or inverse Gaussian) distribution. Unlike commonly used phenomenological models, WALD's parameters can be estimated from the key factors affecting wind dispersal--wind statistics, seed release height, and seed terminal velocity--determined independently of dispersal data. WALD's asymptotic power-law tail has an exponent of -3/2, a limiting value verified by a meta-analysis for a wide variety of measured dispersal kernels and larger than the exponent of the bivariate Student t-test (2Dt). We tested WALD using three dispersal data sets on forest trees, heathland shrubs, and grassland forbs and compared WALD's performance with that of other analytical mechanistic models (revised versions of the tilted Gaussian Plume model and the advection-diffusion equation), revealing fairest agreement between WALD predictions and measurements. Analytical mechanistic models, such as WALD, combine the advantages of simplicity and mechanistic understanding and are valuable tools for modeling large-scale, long-term plant population dynamics. 相似文献

Environmental and stoichiometric controls on microbial carbon-use efficiency in soils 总被引:8,自引:0,他引:8
Carbon (C) metabolism is at the core of ecosystem function. Decomposers play a critical role in this metabolism as they drive soil C cycle by mineralizing organic matter to CO(2) . Their growth depends on the carbon-use efficiency (CUE), defined as the ratio of growth over C uptake. By definition, high CUE promotes growth and possibly C stabilization in soils, while low CUE favors respiration. Despite the importance of this variable, flexibility in CUE for terrestrial decomposers is still poorly characterized and is not represented in most biogeochemical models. Here, we synthesize the theoretical and empirical basis of changes in CUE across aquatic and terrestrial ecosystems, highlighting common patterns and hypothesizing changes in CUE under future climates. Both theoretical considerations and empirical evidence from aquatic organisms indicate that CUE decreases as temperature increases and nutrient availability decreases. More limited evidence shows a similar sensitivity of CUE to temperature and nutrient availability in terrestrial decomposers. Increasing CUE with improved nutrient availability might explain observed declines in respiration from fertilized stands, while decreased CUE with increasing temperature and plant C?:?N ratios might decrease soil C storage. Current biogeochemical models could be improved by accounting for these CUE responses along environmental and stoichiometric gradients. 相似文献

Aim
The introduction of non‐indigenous species (NIS) via man‐made corridors connecting previously disparate oceanic regions is increasing globally. However, the environmental and anthropogenic factors facilitating invasion dynamics and their interactions are still largely unknown. This study compiles and inputs available data for the NIS bivalve Brachidontes pharaonis across the invaded biogeographic range in the Mediterranean basin into a species distribution model to predict future spread under a range of marine scenarios.Location

A systematic review produced the largest presence database ever assembled to inform the selection of biological, chemical and physical factors linked to the spread of B. pharaonis through the Suez Canal. We carried out a sensitivity analysis to simulate current and future trophic and salinity scenarios. A species distribution model was run to determine key drivers of invasion, quantify interactive impacts arising from a range of trophic states, salinity conditions and climatic scenarios and forecast future trajectories for the spread of NIS into new regions under multiple‐parameter scenarios (based on the main factors identified from the systematic review).Results
Impacts on invasion trajectory arising from climate change and interactions with increasing salinity from the new opening of the canal were the primary drivers of expansion across the basin, the effects of which were further enhanced by eutrophication. Predictions of the current distribution were most accurate when multiple stressors were used to drive the model. A habitat suitability index developed at a subcontinental scale from model outputs identified novel favourable conditions for future colonization at specific locations under 2030 and 2050 climatic scenarios.Main conclusions
Future expansion of B. pharaonis will be enhanced by climate‐facilitated increased sea temperature, interacting with increasing pressures from salinity and eutrophication. The spatially explicit risk output maps of invasions represent a powerful visual product for use in communication of the spread of NIS and decision‐support tools for scientists and policymakers. The suggested approach, the observed distribution pattern and driving processes can be applied to other NIS species and regions by providing novel forecasts of species occurrences under future multiple stressor scenarios and the location of suitable recipient habitats with respect to anthropogenic and environmental parameters. 相似文献6.

Profiles of subsurface soil CO2 concentration, soil temperature, and soil moisture, and throughfall were measured continuously during the years 2005 and
2006 in 16 locations at the free air CO2 enrichment facility situated within a temperate loblolly pine (Pinus taeda L.) stand. Sampling at these locations followed a 4 by 4 replicated experimental design comprised of two atmospheric CO2 concentration levels (ambient [CO2]a, ambient + 200 ppmv, [CO2]e) and two soil nitrogen (N) deposition levels (ambient, ambient + fertilization at 11.2 gN m−2 year−1). The combination of these measurements permitted indirect estimation of belowground CO2 production and flux profiles in the mineral soil. Adjacent to the soil CO2 profiles, direct (chamber-based) measurements of CO2 fluxes from the soil–litter complex were simultaneously conducted using the automated carbon efflux system. Based on the
measured soil CO2 profiles, neither [CO2]e nor N fertilization had a statistically significant effect on seasonal soil CO2, CO2 production, and effluxes from the mineral soil over the study period. Soil moisture and temperature had different effects
on CO2 concentration depending on the depth. Variations in CO2 were mostly explained by soil temperature at deeper soil layers, while water content was an important driver at the surface
(within the first 10 cm), where CO2 pulses were induced by rainfall events. The soil effluxes were equal to the CO2 production for most of the time, suggesting that the site reached near steady-state conditions. The fluxes estimated from
the CO2 profiles were highly correlated to the direct measurements when the soil was neither very dry nor very wet. This suggests
that a better parameterization of the soil CO2 diffusivity is required for these soil moisture extremes. 相似文献

Ghrelin is an acylated peptidyl gastric hormone acting on the pituitary and hypothalamus to stimulate appetite, adiposity, and growth hormone release, through activation of growth hormone secretagogue receptor (GHSR)-1a receptor. Moreover, ghrelin features several activities such as inhibition of apoptosis, regulation of differentiation, and stimulation or inhibition of proliferation of several cell types. Ghrelin acylation is absolutely required for both GHSR-1a binding and its central endocrine activities. However, the unacylated ghrelin form, des-acyl ghrelin, which does not bind GHSR-1a and is devoid of any endocrine activity, is far more abundant than ghrelin in plasma, and it shares with ghrelin some of its cellular activities. In here we show that both ghrelin and des-acyl ghrelin stimulate proliferating C2C12 skeletal myoblasts to differentiate and to fuse into multinucleated myotubes in vitro through activation of p38. Consistently, both ghrelin and des-acyl ghrelin inhibit C2C12 proliferation in growth medium. Moreover, the ectopic expression of ghrelin in C2C12 enhances differentiation and fusion of these myoblasts in differentiation medium. Finally, we show that C2C12 cells do not express GHSR-1a, but they do contain a common high-affinity binding site recognized by both acylated and des-acylated ghrelin, suggesting that the described activities on C2C12 are likely mediated by this novel, yet unidentified receptor for both ghrelin forms. 相似文献

The yellow‐legged Asian hornet is an invasive species of wasps, indigenous to the Southeast Asia but recently spreading in Southern Europe. Because of its exponential diffusion and its serious threat to the local honeybee colonies (and to humans as well), restraint measures are currently under investigation. We developed and tested an harmonic radar capable of tracking the flying trajectory of these insects, once equipped with a small transponder, in their natural environment. Several hornets were captured close to a small cluster of honeybee hives, tagged with different transponders and then released in order to follow the flight toward their nest. On‐field testing proved an initial maximum detection range of about 125 m in a hilly and woody area. A number of detections were clearly recorded, and preferential directions of flight were identified. The system herein described is intended as a first low‐cost harmonic radar; it proved the capability to track the hornets while flying and it permitted to test the tagging techniques. Several upgrades of the system have been identified during this work and are extensively described in the last chapter. The designed system has three major advantages over conventional harmonic radars. First and most importantly, it adopts advanced processing techniques to suppress clutter and to improve target detection. Second, it allows radar operations in complex environments, generally hilly and rich in vegetation. Finally, it can continuously track tagged insects (24/7) and in any meteorological condition, providing an effective tool in order to locate the nests of the yellow‐legged Asian hornet. 相似文献

Although Bornean tropical rainforests are among the moistest biomes in the world, they sporadically experience periods of water stress. The observations indicate that these ecosystems tend to have little regulation of water use, despite episodes of relatively severe drought. This water-use behaviour is often referred to as anisohydric behaviour, as opposed to isohydric plants that regulate stomatal movement to prevent hydraulic failure. Although it is generally thought that anisohydric behaviour is an adaptation to more drought-prone habitats, we show that anisohydric plants may also be more favoured than isohydric plants under very moist environments where there is little risk of hydraulic failure. To explore this subject, we examined the advantages of isohydric and anisohydric species as a function of the hydroclimatic environment using a stochastic model of soil moisture and carbon assimilation dynamics parameterized by field observations. The results showed that under very moist conditions, anisohydric species tend to have higher productivity than isohydric plants, despite the fact that the two plant types show almost the same drought-induced mortality. As precipitation decreases, the mortality of anisohydric plants drastically increases whereas that of isohydric plants remains relatively constant and low; in these conditions, isohydric plants surpass anisohydric plants in their productivity. 相似文献
