Pulmonary insufficiency secondary to outflow tract reconstruction remains an unsolved problem. Although a number of surgical techniques have been designed to avoid or palliate this condition, none so far has been satisfactory. This report describes a new composite stented porcine monocusp patch, which, because of its desirable hemodynamic characteristics, promises to yield stable and reproducible results. After activation, Bacillus thuringiensis (Bt) insecticidal toxin forms pores in larval midgut epithelial cell membranes, leading to host death. Although the crystal
structure of the soluble form of Cry1Aa has been determined, the conformation of the pores and the mechanism of toxin interaction
with and insertion into membranes are still not clear. Here we show that Cry1Aa spontaneously inserts into lipid mono- and
bilayer membranes of appropriate compositions. Fourier Transform InfraRed spectroscopy (FTIR) indicates that insertion is
accompanied by conformational changes characterized mainly by an unfolding of the β-sheet domains. Moreover, Atomic Force
Microscopy (AFM) imaging strongly suggests that the pores are composed of four subunits surrounding a 1.5 nm diameter central
depression.

A field experiment was conducted under non-limiting water and nutritional
conditions with three near-isogenic lines of spring wheat (dwarf, DD;
semi-dwarf, SD and standard height, SH) to study the impact of the
GA-insensitive alleles Rht1 and
Rht2, at the cellular level, on the growth of
different vegetative organs and of the pericarp of grains. Cell length and
width of blades of different leaves (3, 7 and flag leaf), the flag-leaf
sheath and the penultimate internode as well as the pericarp of basal
grains from central spikelets of the spike were evaluated. With the
exception of the flag leaf, dwarfing genes produced a significant reduction
in cell length in all the different vegetative organs analysed. There was
no effect on the number of cells nor their width. Therefore, in vegetative
organs, the effects of these alleles appeared to be exclusively due to a
reduction in cell length. It would appear that dwarfing genes act on cell
elongation without affecting cell division.The Rht
alleles did not modify cell length nor width in the pericarp. Grain weight
was different between the lines and these differences were associated with
grain volume at the beginning of linear grain growth. Thus, they reduced
the size of individual grains by reducing the total number of cells in the
pericarp.It appears that Rht alleles reduced the final
sizes of vegetative organs (such as internodes and leaves) and of tissues
(pericarp) associated with reproductive structures (grains), but the modes
of action in these different organs were different.Keywords:

* The most distinctive variation in the monomer composition of lignins in vascular land plants is that found between the two main groups of seed plants. Thus, while gymnosperm lignins are typically composed of guaiacyl (G) units, angiosperm lignins are largely composed of similar levels of G and syringyl (S) units. * However, and contrary to what might be expected, peroxidases isolated from basal (Cycadales and Ginkgoales) and differentially evolved (Coniferales and Gnetales) gymnosperms are also able to oxidize S moieties, and this ability is independent of the presence or absence of S-type units in their lignins. * The results obtained led us to look at the protein database to search for homologies between gymnosperm peroxidases and true eudicot S-peroxidases, such as the Zinnia elegans peroxidase. * The findings showed that certain structural motifs characteristic of eudicot S-peroxidases (certain amino acid sequences and beta-sheet secondary structures) predate the gymnosperm-angiosperm divergence and the radiation of tracheophytes, since they are found not only in peroxidases from basal gymnosperms, ferns and lycopods, but also in peroxidases from the moss Physcomitrella patens (Bryopsida) and the liverwort Marchantia polymorpha (Marchantiopsida), which, as typical of bryophytes, do not have xylem tissue nor lignins. 相似文献

The last step of lignin biosynthesis in Zinnia elegans suspension cell cultures (SCCs) catalyzed by peroxidase (ZePrx) has been characterized. The k(3) values shown by ZePrx for the three monolignols revealed that sinapyl alcohol was the best substrate, and were proportional to their oxido/reduction potentials, signifying that these reactions are driven exclusively by redox thermodynamic forces. Feeding experiments demonstrate that cell wall lignification in SCCs is controlled by the rate of supply of H(2)O(2). The results also showed that sites for monolignol beta-O-4 cross-coupling in cell walls may be saturated, suggesting that the growth of the lineal lignin macromolecule is not infinite. 相似文献

The objective of this study was to investigate the feasibility of using psychrophilic anaerobic digestion in sequencing batch reactors (PADSBRs) to co-digest grinded swine carcasses and swine manure slurry at 20 degrees C and 25 degrees C. The PADSBRs were operated on two-week and four-week treatment cycle lengths, which included the fill, react, and draw phases. Two carcass loading rates (CLRs) were tested, that is 20 and 40g of carcass per litre of manure, which were equivalent to 4 and 8 times, respectively, the normal mortality rate on commercial farms. The PADSBR performance was compared to that of PADSBRs operated at 25 degrees C and fed manure only. The addition of swine carcass to PADSBR feed did not affect the stability of the bioreactors at both CLRs. The performance of the PADSBRs co-digesting swine carcasses was not statistically different from the control in terms of biogas production and quality. There was no accumulation of volatile fatty acids in the bioreactors at the end of the treatment cycle. The mixed-liquor pH and alkalinity remained within acceptable ranges for the anaerobic microflora. Also, there was no operational problem caused by the formation of foam and scum in the system. 相似文献

The successful exploitation of germplasm banks, harbouring plant genetic resources indispensable for plant breeding, will depend on our ability to characterize their genetic diversity. The Vegetable Germplasm Bank of Zaragoza (BGHZ) (Spain) holds an important Capsicum annuum collection, where most of the Spanish pepper variability is represented, as well as several accessions of other domesticated and non-domesticated Capsicum spp from all over the five continents. In the present work, a total of 51 C. annuum landraces (mainly from Spain) and 51 accessions from nine Capsicum species maintained at the BGHZ were evaluated using 39 microsatellite (SSR) markers spanning the whole genome. The 39 polymorphic markers allowed the detection of 381 alleles, with an average of 9.8 alleles per locus. A sizeable proportion of alleles (41.2%) were recorded as specific alleles and the majority of these were present at very low frequencies (rare alleles). Multivariate and model-based analyses partitioned the collection in seven clusters comprising the ten different Capsicum spp analysed: C. annuum, C. chinense, C. frutescens, C. pubescens, C. bacatum, C. chacoense and C. eximium. The data clearly showed the close relationships between C. chinense and C. frutescens. C. cardenasii and C. eximium were indistinguishable as a single, morphologically variable species. Moreover, C. chacoense was placed between C. baccatum and C. pubescens complexes. The C. annuum group was structured into three main clusters, mostly according to the pepper fruit shape, size and potential pungency. Results suggest that the diversification of C. annuum in Spain may occur from a rather limited gene pool, still represented by few landraces with ancestral traits. This ancient population would suffer from local selection at the distinct geographical regions of Spain, giving way to pungent and elongated fruited peppers in the South and Center, while sweet blocky and triangular types in Northern Spain. 相似文献

The Tortonian carbonate ramp of Menorca was previously studied on the basis of outcrops along sea-cliff outcrops. These sea
cliffs, in combination with inland water wells, are the basis for a facies model for the reconstruction of the internal architecture
and for characterizing the internal heterogeneities of this carbonate platform. However, any such three-dimensional reconstruction
is generally limited by the given geometrical arrangement of the two-dimensional outcrops and the uncertainties of correlation
with the one-dimensional wells. Here, ground-penetrating radar (GPR) has been employed in order to test and refine the depositional
model. Although GPR is well known for being an excellent tool for high-resolution underground studies of sedimentary systems,
the application for studying carbonate rocks is still far from routine. The reason for this discrepancy is two-fold: the minor
mineralogical contrast between lithologies in carbonate rocks results in subtle reflections, and, even more important, the
porosity structure in carbonates is thoroughly and repeatedly changed during diagenesis, commonly across the different facies,
leading to problems in predictability of the petrophysical properties. The study of the Menorcan carbonate ramp with large
distance–deep penetration GPR sections demonstrates that in spite of these difficulties, GPR is a valuable tool for extrapolating
information from outcrops and wells. It is useful for characterizing heterogeneities larger than outcrop scale. 相似文献

Pollution relative to phosphorus excretion in poultry manure as well as the soaring prices of phosphate, a non-renewable resource, remain of major importance. Thus, a good understanding of bird response regarding dietary phosphorus (P) is a prerequisite to optimise the utilisation of this essential element in broiler diets. A database built from 15 experiments with 203 treatments was used to predict the response of 21-day-old broilers to dietary non-phytate P (NPP), taking into account the main factors of variation, calcium (Ca) and microbial phytase derived from Aspergillus niger, in terms of average daily feed intake (ADFI), average daily gain (ADG), gain to feed (G:F) and tibia ash concentration. All criteria evolve linearly (P < 0.001) and quadratically (P < 0.001) with dietary NPP concentration. Dietary Ca affected the intercept and linear component for ADG (P < 0.01), G:F (P < 0.05) and tibia ash concentration (P < 0.001), whereas for ADFI, it affected only the intercept (P < 0.01). Microbial phytase addition impacted on the intercept, the linear and the quadratic coefficient for ADFI (P < 0.01), ADG (P < 0.001) and G:F (P < 0.05), and on the intercept and the linear component (P < 0.001) for tibia ash concentration. An evaluation of these models was then performed on a database built from 28 experiments and 255 treatments that were not used to perform the models. Results showed that ADFI, ADG and Tibia ash concentration were predicted fairly well (slope and intercept did not deviate from 0 to 1, respectively), whereas this was not the case for G:F. The increase in dietary Ca concentration aggravated P deficiency for all criteria while phytase addition had a positive effect. The more P deficiency was marked, the more the bird response to ADFI, ADG, G:F and tibia ash concentration was exacerbated. It must also be considered that even if the decrease in dietary Ca may improve P utilisation, it could in turn become limiting for bone mineralisation. In conclusion, this meta-analysis provides ways to reduce dietary P in broiler diets without impairing performance, taking into account dietary Ca and microbial phytase. 相似文献
