Experiments are reported in which the uptake of 86Rb+, used as an analog of K+, into cultured cells of Arabidopsis thaliana is investigated. A single transport system is found with Km = 0.34 millimolar and Vmax = 14 nmoles per milligram of protein per hour. This system is blocked by the metabolic inhibitor carbonyl cyanide m-chlorophenyl hydrazone (CCCP) and by cold. At high concentrations of external K+ (above 1 millimolar), a significant fraction of total uptake is energy-independent. No evidence is found for more than one energy-dependent uptake system or for concentration-dependent modifications of a carrier as postulated in multiphasic transport models.  相似文献

Mouse leukocytes were studied for membrane receptors for the third C component by rosette formation with C coated erythrocytes (EAC). Methods were devised for the preparation of EAC complexes containing either mouse C3b or mouse C3d. EAC 1-3dmo were prepared from EA treated with whole mouse serum while EAC 1-3bmo were produced from EAC 142hu treated with whole mouse serum containing sodium suramin. The specificity of the EAC complexes for mouse leukocytes was confirmed by inhibition experiments using fluid phase human C3d. Low concentrations of fluid phase human C3d inhibited EAC1-3dmo rosettes but failed to inhibit EAC 1-3bmo rosettes. Eight-fold higher concentrations of fluid phase C3d caused partial inhibition of EAC1-3bmo rosette formation with lymphocytes, but not with other types of murine leukocytes. Thus mouse leukocytes apparently contain the same two types of C receptors as do human and guinea pig leukocytes. Mouse CR1 is specific for a non-C3d region of C3b, (possibly analogous to human C3c) whereas mouse CR2 is specific for both C3d and the C3d region of C3b.  相似文献

Between 1971 and 1975, Fascioloides magna was found in 46 of 67 (69%) feral swine (Sus scrofa) in southern Texas. Flukes were recovered from swine in areas where F. magna commonly has been recovered from white-tailed deer and cattle. One to 12 flukes were recovered from each infected animal. Their presence was indicated by black hematin pigment on the liver and various other internal organs. Eggs were not detected in the gallbladder or feces of infected animals although mature flukes and eggs were recovered in the livers suggesting that, like cattle, feral swine can be infected but are aberrant hosts for the parasite and do not disseminate eggs.  相似文献

Stomatal density, stomatal aperture length, area/leaf, and number of stomata/leaf were measured after the annual C3 agronomic grasses oats (Avena sativa) and wheat (Triticum aestivum), the C, woody legume honey mesquite (Prosopis glandulosa), and the perennial C4 grass little bluestem (Schizachyrium scoparium) were grown across a subambient carbon dioxide concentration ([CO2]) gradient from near 200 to 350 μmol/mol in a growth chamber. The purpose was to determine if the size and density of stomata vary in response to atmospheric [CO2] during growth, across a subambient [CO2] range representative of the doubling that has occurred since the last ice age. Changes in stomatal density and aperture length with increasing [CO2] were small when detected. Stomatal density decreased on adaxial flag leaf surfaces of wheat, and aperture length increased slightly with [CO2], Leaf area and number of stomata/flag leaf increased by similar proportions with [CO2] in two wheat cultivars. No consistent relationship between [CO2] and stomatal density or size was detected in mesquite, oats, or little bluestem. We conclude that individual plants of these species lack the plasticity to significantly alter stomatal density and aperture length in response to increasing atmospheric [CO2] in a single generation (annuals) or growing season (perennials).  相似文献

Identifying the plant traits and patterns of trait distribution in communities that are responsible for biotic regulation of CO2 uptake–climate responses remains a priority for modeling terrestrial C dynamics. We used remotely sensed estimates of gross primary productivity (GPP) from plots planted to different combinations of perennial grassland species in order to determine links between traits and GPP–climate relationships. Climatic variables explained about 50% of the variance in temporal trends in GPP despite large variation in CO2 uptake among seasons, years, and plots of differing composition. GPP was highly correlated with contemporary changes in net radiation (Rn) and precipitation deficit (potential evapotranspiration minus precipitation) but was negatively correlated with precipitation summed over 210 days prior to flux measurements. Plots differed in GPP–Rn and GPP–water (deficit, precipitation) relationships. Accounting for differences in GPP–climate relationships explained an additional 11% of variance in GPP. Plot differences in GPP–Rn and GPP–precipitation slopes were linked to differences in community-level light-use efficiency (GEE*). Plot differences in GPP–deficit slopes were linked to differences in a species abundance-weighted index of specific leaf area (SLA). GEE* and weighted SLA represent vegetation properties that may regulate how CO2 uptake responds to climatic variation in grasslands.  相似文献

The surface-accessible ectodomain region of the Plasmodium falciparum apical membrane antigen 1 (AMA1) is a malaria vaccine candidate. The amino acid sequence may be under selection from naturally acquired immune responses, and previous analyses with a small number of allele sequences indicate a non-neutral pattern of nucleotide variation. To investigate whether there is selection to maintain polymorphism within a population, and to identify the parts of the ectodomain under strongest selection, a sample of 51 alleles from a single endemic population was studied. Analyses using Fu and Li's D and F tests, Tajima's D test, and the McDonald-Kreitman test (with the chimpanzee parasite P. reichenowi as outgroup) show significant departure from neutrality and indicate the selective maintenance of alleles within the population. There is also evidence of a very high recombination rate throughout the sequence, as estimated by the recombination parameter, C, and by the rapid decline in linkage disequilibrium with increasing nucleotide distance. Of the three domains (I-III) encoding structures determined by disulfide bonds, the evidence of selection is strongest for Domains I and III. We predict that these domains in particular are targets of naturally acquired protective immune responses in humans.  相似文献

Damage or deprivation of a localized region of the skin surface has been shown to induce a selective expansion of adjacent skin surface representations in the adult somatosensory cortex. Here, we use repeated optical imaging in conjunction with single unit recordings to assess the plasticity of a single whisker's functional representation in the adult rat. We observed a large-scale expansion of a single whisker's functional representation following innocuous removal of all neighboring whiskers. Surprisingly, the same manipulation can also induce a large-scale contraction of the representation if the animal is removed from its home cage and given a brief opportunity to use its whiskers for active exploration of a different environment. Both the expansion and contraction reverse upon regrowth of the deprived whiskers. Thus, allowing the animal to use its deprived receptor organ in active exploration can determine the direction of plasticity in the adult cortex.  相似文献

Biodiversity survey and inventory have resulted in new information on the distribution of Protostrongylidae in Dall's sheep (Ovis dalli dalli) from the Northwest Territories (NT, Canada) and from Alaska (AK, USA). In 1998, Parelaphostrongylus odocoilei adults were found for the first time in the skeletal muscles of Dall's sheep in the Mackenzie Mountains (NT). Adult P. odocoilei were associated with petechial and ecchymotic hemorrhages and localized myositis; eggs and larvae in the lungs were associated with diffuse granulomatous pneumonia. Experimental infections of the slugs Deroceras laeve and Deroceras reticulatum with dorsal-spined first-stage larvae assumed to be P. odocoilei, from ground-collected feces from Dall's sheep in the Mackenzie Mountains, yielded third-stage larvae by at least 28 (in D. laeve) and 48 (in D. reticulatum) days post-infection. Third-stage larvae emerged from D. laeve between days 19 and 46 post-infection and emergence occurred both at room temperature and at 10 to 12 C. Protostrongylus stilesi were definitively identified from the lungs of Dall's sheep collected in the Mackenzie Mountains, NT in 1998. Specimens collected from sheep in the Mackenzie Mountains, NT in 1971-72, and the Alaska Range, AK in 1972 were also confirmed as P. stilesi. Lung pathology associated with adults, eggs, and larvae of P. stilesi was similar to that described in bighorn sheep (Ovis canadensis). Concurrent infections with P. odocoilei and P. stilesi in a single host have not been previously reported.  相似文献

Important drivers for emergence of infectious disease in wildlifeinclude changes in the environment, shrinking habitats or concentrationof wildlife, and movement of people, animals, pathogens, orvectors. In this paper we present three case-studies of emergingparasitic infections and diseases in ungulates in the Canadiannorth. First we discuss climate warming as an important driverfor the emergence of disease associated with Umingmakstrongyluspallikuukensis, a nematode lungworm of muskoxen. Then we examinehow Protostrongylus stilesi, the sheep lungworm, emerged (orre-emerged) in muskoxen after re-introduction of this host intoits historical range made it sympatric with Dall's sheep. Finally,we consider Teladorsagia boreoarcticus, a newly described andcommon abomasal nematode of muskoxen that is emerging as a disease-causingparasite and may be an important regulator for muskox populationson Banks Island, Northwest Territories. These and other arctichost-parasite systems are exquisitely tuned and constrainedby a harsh and highly seasonal environment. The dynamics ofthese systems will be impacted by climate change and other ecologicaldisruptions. Baseline knowledge of parasite biodiversity andparasite and host ecology, together with predictive models andlong-term monitoring programs, are essential for anticipatingand detecting altered patterns of host range, geographic distribution,and the emergence of parasitic infections and diseases.  相似文献
