Glioblastoma multiforme (GBM) is the most aggressive type of brain tumor, and the prognosis remains poor. Rearrangement of ROS1 gene, which was shown to have an oncogenic potential, was previously discovered in GBM cell lines. In this pilot study, we aimed to identify the incidence of ROS1 rearrangement in GBM patient tissues to explore novel biomarkers for therapeutic strategy. Formalin-fixed and paraffin-embedded (FFPE) tissue sections from 109 patients with GBM were screened for ROS1 rearrangement by anti-ROS immunohistochemistry (IHC) and ROS1 break-apart fluorescent in situ hybridization (FISH) assays. O6-methylguanine-DNA methyltransferase (MGMT) gene promoter methylation and Isocitrate dehydrogenase 1 (IDH1) mutation status were also assessed. All samples were interpreted by two experienced pathologists who were blinded to the clinical data. A total of 109 samples were collected and all samples were examined for ROS1 rearrangement by IHC and FISH assays, and none was found to harbor ROS1 rearrangement. MGMT gene methylation was found in 42 (39.2%) cases, and IDH1 mutation was found in 6 (5.5%) cases. In this study, ROS1 rearrangement was not identified in GBM patients, and thus it is difficult to classify ROS1 rearrangement as a novel molecular subset in GBM patients for now.  相似文献

The recently discovered ammonia-oxidizing archaea (AOA) have been suggested as contributors to the first step of nitrification in terrestrial ecosystems, a role that was previously assigned exclusively to ammonia-oxidizing bacteria (AOB). The current study assessed the effects of agricultural management, specifically amendment of soil with biosolids or synthetic fertilizer, on nitrification rates and copy numbers of archaeal and bacterial ammonia monooxygenase (amoA) genes. Anaerobically digested biosolids or synthetic fertilizer was applied annually for three consecutive years to field plots used for corn production. Biosolids were applied at two loading rates, a typical agronomic rate (27 Mg hectare(-1) year(-1)) and double the agronomic rate (54 Mg hectare(-1) year(-1)), while synthetic fertilizer was applied at an agronomic rate typical for the region (291 kg N hectare(-1) year(-1)). Both biosolids amendments and synthetic fertilizer increased soil N and corn yield, but only the biosolids amendments resulted in significant increases in nitrification rates and increases in the copy numbers of archaeal and bacterial amoA genes. In addition, only archaeal amoA gene copy numbers increased in response to biosolids applied at the typical agronomic rate and showed a significant correlation with nitrification rates. Finally, copy numbers of archaeal amoA genes were significantly higher than copy numbers of bacterial amoA genes for all treatments. These results implicate AOA as being primarily responsible for the increased nitrification observed in an agricultural soil amended with biosolids. These results also support the hypothesis that physiological differences between AOA and AOB may enable them to occupy distinct ecological niches.  相似文献

Mother–offspring interactions soon after parturition play a key role in the survival of mammals. We investigated the suckling behavior of semi-captive Western Derby eland (Taurotragus derbianus derbianus) in a 60-ha enclosure covered by dense savanna vegetation in Senegal and farmed Common eland (T. oryx) on an open 2-ha pasture in the Czech Republic. We hypothesized that the basic pattern of suckling bout duration and mother–offspring interactions would be similar between species, but would vary in response to the environmental conditions and breeding system. During three calving periods, we observed the suckling of 27 and 23 calves of Derby and Common elands, respectively, between the ages of 1–5 months, and the interactions between mother and calf before and during suckling. Suckling bout duration increased with the age of the calves for both elands. However, in Derby elands we recorded longer suckling bouts in male than female calves and shorter suckling bouts in primiparous mothers than multiparous ones; no differences were found in farmed Common elands. The animals’ active approach to mother–offspring contact, for example naso–anal contact, and initiation and termination of suckling, resulted in longer suckling bouts in Derby elands. The results suggest that Derby elands that range over a large enclosure with dense vegetation cover adjust their maternal behavior in compliance with potential predator risk, facing a trade-off between nursing and vigilant behavior in the wild. The suckling behavior of farmed elands, on the other hand, reflects the conditions of captivity without predators and with the small available area enabling permanent visual contact of animals.  相似文献
