Summary During embryogenesis and planula development of the colonial hydroidHydractinia echinata cell proliferation decreases in a distinct spatio-temporal pattern. Arrest in S-phase activity appears first in cells localized at the posterior and then subsequently at the anterior pole of the elongating embryo. These areas do not resume S-phase activity, even during the metamorphosis of the planula larva into the primary polyp. Tissue containing the quiescent cells gives rise to the terminal structures of the polyp. The posterior area of the larva becomes the hypostome and tentacles, while the anterior part of the larva develops into the basal plate and stolon tips. In mature planulae only a very few cells continue to proliferate. These cells are found in the middle part of the larva. Labelling experiments indicate that the prospective material of the postmetamorphic tentacles and stolon tips originates from cells which have exited from the cell cycle in embryogenesis or early in planula development. Precursor cells of the nematocytes which appear in the tentacles of the polyp following metamorphosis appear to have ceased cycling before the 38th hour of embryonic development. The vast majority of the cells that constitute the stolon tips of the primary polyp leave the cell cycle not later than 58 h after the beginning of development. We also report the identification of a cell type which differentiates in the polyp without passing through a post-metamorphic S-phase. The cell type appears to be neural in origin, based upon the identification of a neuropeptide of the FMRFamide type.  相似文献

Summary Polyps of mature colonies of Hydractinia echinata obey the rule of distal transformation by regenerating heads but not stolons. However, this rule is not valid for young polyps as these regenerate stolons from proximal cut ends. Also, small cell aggregates and even small fragments excised from full-grown polyps are capable of stolon formation. Aggregates produced from dissociated cells undergo either distal or proximal transformation depending on their size, speed of head regeneration in the donor used for dissociation and the positional derivation of the cells. The latent capability of stolon formation is released under conditions that cause loss of morphogens and depletion of their sources. However, internal regulative processes can also lead to gradual proximal transformation: regenerating segments of polyps sometimes form heads at both ends and the distal pattern is duplicated. Subsequently, all sets of proximal structures, including stolons, are intercalated. In contrast to distal transformation, proximal transformation is a process the velocity of which declines with the age and size of the cell community.  相似文献

Background

Campylobacter jejuni is a leading cause of foodborne bacterial enterocolitis worldwide. Investigation of immunopathology is hampered by a lack of suitable vertebrate models. We have recently shown that gnotobiotic mice as well as conventional IL-10−/− animals are susceptible to C. jejuni infection and develop intestinal immune responses. However, clinical symptoms of C. jejuni infection were rather subtle and did not reflect acute bloody diarrhea seen in human campylobacteriosis.

Methodology/Principal Findings

In order to overcome these limitations we generated gnotobiotic IL-10−/− mice by quintuple antibiotic treatment starting right after weaning. The early treatment was essential to prevent these animals from chronic colitis. Following oral infection C. jejuni colonized the gastrointestinal tract at high levels and induced acute enterocolitis within 7 days as indicated by bloody diarrhea and pronounced histopathological changes of the colonic mucosa. Immunopathology was further characterized by increased numbers of apoptotic cells, regulatory T-cells, T- and B-lymphocytes as well as elevated TNF-α, IFN-γ, and MCP-1 concentrations in the inflamed colon. The induction of enterocolitis was specific for C. jejuni given that control animals infected with a commensal E. coli strain did not display any signs of disease. Most strikingly, intestinal immunopathology was ameliorated in mice lacking Toll-like-receptors-2 or -4 indicating that C. jejuni lipoproteins and lipooligosaccharide are essential for induction and progression of immunopathology.

Conclusion/Significance

Gnotobiotic IL-10−/− mice develop acute enterocolitis following C. jejuni infection mimicking severe episodes of human campylobacteriosis and are thus well suited to further dissect mechanisms underlying Campylobacter infections in vivo.  相似文献

Colonies of hydroids exhibit periodic biological patterns. Polyps form on stolons at fixed distances, obeying distinct rules. The spacing mechanism is based on inhibition emanating from existing polyps, predominantly from the head of the polyp. Removal of polyps from young colonies reduces the distance between initiating polyps and newly formed polyps to 50% of the normal values. Head removal suffices to obtain an almost identical reduction. Polyp enlargement which increases the distance between the inhibition-emitting head and the stolon tissue reduces the intrastolonal range of inhibition. In the stolon tissue, decrease of inhibitory activity occurs. An increase in the stolon/polyp ratio of a colony reduces bud distances. The decay is, in part, due to loss of inhibitory activity into the external medium: if colonies are incubated in conditioned culture medium derived from crowded colonies having normally large interpolyp distances, bud distances increase in test colonies. The effectiveness of transfer of inhibitory activity from tissue into the medium depends on culture conditions. If convection is increased by agitation of the culture medium, the distances between polyp and bud decreases; viscosity enhancement of the culture medium reduces convection and bud distances become larger. This effect is compensated for by additional agitation of the viscosity enhanced culture medium. Our results support the idea that a lateral inhibition mechanism controls polyp spacing in the stolon and that inhibition is based on diffusible inhibitory compounds.  相似文献

Growth pulsations (GP) in hydroid polyps are associated with changes in vacuolar patterns which can be imitated by altering external osmolarity. With the use of X-ray spectroscopy we measured the elemental contents in the vacuoles and cytoplasm of the growing tips of a hydroid polyp, Podocoryne carnea, under various tonicity conditions. Under hypertonic condition which arrested the samples at the retraction phase of normal GP, the elemental content within the vacuolar compartment appeared to be similar to that of the external medium, confirming our previous conclusion about the dehermetization of the vacuolar compartment under these conditions. Under hypotonical condition which arrested samples at the extension GP phase (vacuoles isolated) element ratio data displayed an obvious bimodality. At least one of the data groups could be characterized by a significant increase in the concentrations of sodium and potassium, as related to Cl, Ca and Mg, and in comparison to the same ratios in hypotonical samples and those in the external medium. We suggest that under hypotonical conditions the isolated vacuolar compartment is formed by influx of sodium and potassium ions. These cations are accompanied by anions other than chloride. Potassium appears to be transferred into the vacuoles from the cytoplasm while the sodium derives from the external environment.  相似文献
