East African runners are continually successful in international distance running. The extent to which genetic factors influence this phenomenon is unknown. The insertion (I) rather than deletion (D) of a 287 bp fragment in the human angiotensin converting enzyme (ACE) gene is associated with lower circulating and tissue ACE activity and with endurance performance amongst Caucasians. To assess the association between ACE gene variation and elite endurance athlete status in an African population successful in distance running, DNA samples were obtained from 221 national Kenyan athletes (N), 70 international Kenyan athletes (I), and 85 members of the general Kenyan population (C). Blood samples were obtained from C and assayed for circulating ACE activity. ACE I/D (rs????--from NCBI SNPdb first time poly mentioned) genotype was determined, as was genotype at A22982GD (rs????--from NCBI SNPdb first time poly mentioned) which has been shown to associate more closely with ACE levels in African subjects than the I/D polymorphism. ACE I/D and A22982G genotypes explained 13 and 24% of variation in circulating ACE activity levels (P = 0.034 and <0.001 respectively). I/D genotype was not associated with elite endurance athlete status (df = 4, chi(2) = 4.1, P=0.39). In addition, genotype at 22982 was not associated with elite endurance athlete status (df = 4, chi(2) = 5.7, P = 0.23). Nor was the A allele at 22982, which is associated with lower ACE activity, more prevalent in N (0.52) or I (0.41) relative to C (0.53). We conclude that ACE I/D and A22982G polymorphisms are not strongly associated with elite endurance athlete status amongst Kenyans.  相似文献

Genetic variation in the human angiotensin I‐converting enzyme (ACE) gene has been associated with many heritable traits, including obesity. Herein, we report the results of a study of obesity‐related phenotypes and lifestyle in 1016 teen‐aged Greeks. We show that there is a strong association (p = 0.001) between subcutaneous fat and the ACE insertion/deletion (I/D) polymorphism in females, possession of genotypes containing the D allele being associated with increased fat thickness. This association is strongest in females who participate in no extra exercise and accounts for 6.5% of the phenotypic variance in fat thickness by ANOVA. The association is additive, with the mean phenotypic values in heterozygotes intermediate between the means of the two homozygotes, and the association acts at both extremes of the fat thickness distribution in a classical polygenic manner. Other ACE polymorphisms (rs4424958, rs4311) that define major haplotypes in European populations fail to provide stronger associations with the subcutaneous fat phenotype. Because ACE I/D is the polymorphism most strongly associated with circulating ACE levels in European populations, we propose that the functional allelic differences that influence circulating ACE levels also mediate the associations with the obesity‐related phenotypes studied here.  相似文献

Peroxisome proliferator-activated receptor gamma (PPAR gamma) and peroxisome proliferator-activated receptor delta (PPAR delta) are promising candidate genes for obesity. Associations between adiposity-related phenotypes and genetic variation in PPAR gamma (Pro12Ala and C1431T), as well as PPAR delta (T+294C) were assessed in 2,102 Greek children aged 1-6 years, as part of a large-scale epidemiological study (Growth, Exercise and Nutrition Epidemiological Study In preSchoolers). In girls aged 3-4 years, the Ala12 allele was associated with higher mid-upper arm (P = 0.010) and hip (P = 0.005) circumferences, as well as subscapular (P = 0.008) and total skinfolds (P = 0.011) that explained 2.0, 3.7, 2.1, and 1.9% of the phenotypic variance, respectively, while the T1431 allele was associated with higher mean values for waist circumference (P = 0.018) and suprailiac skinfold (P = 0.017), genotype accounting for 1.6% of the variance in both phenotypes. No significant effects of PPAR delta T+294C polymorphism or the interaction of the PPAR delta and PPAR gamma variants on adiposity-related phenotypes were observed in any age group or gender. Haplotype-based analysis including both PPAR gamma polymorphisms revealed that in girls aged 3-4 years, the Ala-T haplotype was associated with higher waist (P = 0.014) and hip (P = 0.007) circumferences compared to the common Pro-C haplotype. The PPAR gamma Pro12Ala and C1431T polymorphisms are associated with increased adiposity during early childhood in a gender- and age-specific manner and independently of the PPAR delta T+294C polymorphism.  相似文献

This study was designed to examine the effects of alterations in dietary carbohydrate (CHO) intake on the performance of high-intensity exercise lasting approximately 10 min (EXP 1) and 30 min (EXP 2). Trained subjects exercised to exhaustion on four occasions on a cycle ergometer at 90% of maximal oxygen consumption (VO2max; EXP 1, n = 5) and 80% of VO2max (EXP 2, n = 7). The first two tests were familiarisation trials and were carried out following the subjects' normal diet. Normal training was continued but standardised during the periods of dietary control. The subsequent two tests were performed 2 weeks apart after 7 days of dietary manipulation. The two diets were a 70% and a 40% CHO diet, isoenergetic with each subject's normal diet and administered in a randomised order. At both exercise intensities, time to exhaustion following the high CHO and low CHO diets was not different [mean (SD) EXP 1: 11.56 (3.78) min and 8.95 (2.35) min, P = 0.22; EXP 2: 26.9 (7.4) min and 26.5 (6.5) min, P = 0.90]. No differences in resting blood metabolite concentrations were found apart from a lower beta-hydroxybutyrate (beta-HB) level following the high CHO diet in EXP 2. Blood lactate was higher after exercise at 90% of VO2max following the high CHO diet. Blood lactate was higher, and beta-HB lower during exercise at 80% of VO2max following the high CHO diet. No differences were found in the other blood metabolites tested. The respiratory exchange ratio after 15 min of exercise at 80% of VO2max was higher on the high CHO diet. No differences in oxygen uptake, heart rate (EXP 2) or ratings of perceived exertion (both experiments) were found between conditions. These results indicate that moderate changes in diet composition during training do not affect the performance of high-intensity exercise in trained individuals when the total energy intake is moderately high.  相似文献

We compared the accuracy of an ingestible telemetry pill method of core temperature (Tc) measurement and an infrared tympanic membrane thermometer to values from a rectal thermistor during exercise-induced heat stress. Ten well-trained subjects completed four exercise trials consisting of 40 min constant-load exercise at 63% of maximum work rate followed by a 16.1 km time trial at 30 °C and 70% relative humidity. Temperature at rest was not different between the three methods of Tc measurement (Tre: 37.2±0.3 °C; Tp: 37.2±0.2 °C; Tty: 37.1±0.3 °C; P=0.40P=0.40). Temperature rose continuously during the exercise period (ΔTre: 2.2±0.5 °C; ΔTp: 2.2±0.5 °C; ΔTty: 1.9±0.5 ±°C and there were no differences between Tre and Tp measurements at any time throughout exercise (P=0.32P=0.32). While there were no differences between Tre and Tty after 10 min (P=0.11P=0.11) and 20 min (P=0.06P=0.06) of exercise, Tty was lower than Tre after 30 min of exercise (P<0.01P<0.01) and remained significantly lower throughout the remainder of the exercise period. These results demonstrate that the telemetry pill system provides a valid measurement of trunk temperature during rest and exercise-induced thermal strain. Tty was significantly lower than Tre when temperature exceeded 37.5 °C. However, whether these differences are due to selective brain cooling or imperfections in the tympanic membrane thermometer methodology remains to be determined.  相似文献

The relative importance of genetic and socio-cultural influences contributing to the success of east Africans in endurance athletics remains unknown in part because the pre-training phenotype of this population remains incompletely assessed. Here cardiopulmonary fitness, physical activity levels, distance travelled to school and daily energy expenditure in 15 habitually active male (13.9±1.6 years) and 15 habitually active female (13.9±1.2) adolescents from a rural Nandi primary school are assessed. Aerobic capacity () was evaluated during two maximal discontinuous incremental exercise tests; physical activity using accelerometry combined with a global positioning system; and energy expenditure using the doubly labelled water method. The of the male and female adolescents were 73.9±5.7 ml. kg−1. min−1 and 61.5±6.3 ml. kg−1. min−1, respectively. Total time spent in sedentary, light, moderate and vigorous physical activities per day was 406±63 min (50% of total monitored time), 244±56 min (30%), 75±18 min (9%) and 82±30 min (10%). Average total daily distance travelled to and from school was 7.5±3.0 km (0.8–13.4 km). Mean daily energy expenditure, activity-induced energy expenditure and physical activity level was 12.2±3.4 MJ. day−1, 5.4±3.0 MJ. day−1 and 2.2±0.6. 70.6% of the variation in was explained by sex (partial R2 = 54.7%) and body mass index (partial R2 = 15.9%). Energy expenditure and physical activity variables did not predict variation in once sex had been accounted for. The highly active and energy-demanding lifestyle of rural Kenyan adolescents may account for their exceptional aerobic fitness and collectively prime them for later training and athletic success.  相似文献
