Abstract: A sensitive radioisotopic assay has been used to examine the kinetic properties and regulation of biosynthesis of glutamine synthetase in C-6 glioma cultures. The Km values for glutamate, MgATP, and ammonium ion were 5mM, 14 mM, and 0.042 mM, respectively, when measured at the pH optimum of 7.2. There was an absolute requirement for a divalent metal ion, with 15 mM- Mg2+ being the preferred ion at pH 7.2. Activity was completely inhibited after 30 min with 8 mM-L-methionhe sulfoximine. The addition of 1 μM-cortisol to C-6 cultures caused a two to threefold increase in glutamine synthetase specific activity over a 96-h period, while dexamethasone at the same concentration elevated the level some 7-10-fold. This was specific for glucocorticoids, as other steroid hormones or catecholamines did not significantly affect glutamine synthetase specific activity. Cycloheximide (30 μM) or actinomycin D (0.01 μg/ml) blocked the hormone response. The continued presence of hormone was required in order to maintain an elevated enzyme level. The results suggest that glucocorticoids act to induce glutamine synthetase by stimulating new enzyme synthesis.  相似文献

Using an immunohistochemical technique and densitometric analysis in experiments on rats, we examined the expression of an early-response protein, c-Fos, in the medial parvicellular subnucleus of the hypothalamic paraventricular nucleus (mpcPVN). During 7 days, the animals were kept under conditions of (i) normal photoperiod (group LD), (ii) continuous illumination (group LL), and (iii) continuous darkness (light deprivation, group DD). It was found that both normal photoperiodicity and artificial modifications of the latter result in significant changes of c-Fos immunopositivity in the nuclei of mpcPVN neurons (cells that synthesize corticotropin-releasing factor). Changes in the indices of concentration and amount of c-Fos in mpcPVN neurons were, from some aspects, similar to those observed under analogous experimental conditions in neurons of a leading pacemaker of the circadian periodicity, the suprachiasmatic nuclei (SChNs) of the hypothalamus [10], but, at the same time, demonstrated noticeable specificity from other aspects. In all experimental groups, the concentration and amount of c-Fos in mpcPVN neurons were greater during daytime than at night. Dramatic increases in the above parameters observed at daytime in rats subjected to light deprivation were the most significant effect of modifications of the photoperiod. Both continuous illumination and light deprivation resulted in some increase of the number (density) of c-Fos-immunopositive units in the mpcPVN. The nuclei of neurons of this structure demonstrated the phenomenon found earlier in SChN neuron; geometrical dimensions of the cell nuclei of these structures depended noticeably on both illumination conditions and periodicity. Probable mechanisms of the dependence of c-Fos expression in the hypothalamic nuclei on the photoperiod and its modifications, interdependence of such changes observed in the circadian pacemaker (SChN) and an important component of the system controlling the stress reactions (mpcPVN), and the relation of the above phenomena to the level of melatonin produced by the epiphysis cerebri (main neuroendocrine intermediator organizing the above-mentioned periodicity) are discussed.  相似文献

The effects of a single-shot intraperitoneally administration of melatonin in a dose of 1 mg per kg body weight and epithalamin in a dose of 2.5 mg per kg body weight on the activities of Na+, K(+)-ATPase and 5'-nucleotidase were investigated in the forebrain of juvenile male white rats under the acute hypobaric hypoxia. The melatonin and epithalamin administration against the background of acute hypoxia prevented an acute hypoxia inducing decrease in the activity of Na+, K(+)-ATPase as well as increased in the activity of 5'-nucleotidase. Such effects of pineal hormones can promote antihypoxic protection of neurons.  相似文献

The neurofunctional system, which receives a photoperiod, is a photoperiodic brain system. As a part of chronoperiodic system of organism it is involved in perception and transfer of information about the main external Zeitgeber to the peripheral tissues. Such role of photoperiodic system allows it not only synchronize the chronorhythms of different somatic and visceral functions, but also realize the coordination and the modulation of adaptation mechanisms to the stressors influence. The present review is focused on the ways of conduction of photoperiodic information, role of suprachiasmatic nuclei of hypothalamus in endogenous oscillation of chronorhythms and pineal gland as a neuroendocrinal transducer; and also on characteristics of circadian and circannual parts of photoperiodic system. Special attention is given to vegetative part of photoperiodic system and melatonin--"the hormone of dark". It is supposed that adenosine, one of the humoral elements of photoperiodic system, is involved in transfer of duration of the light part of photoperiod. Due to presented data we have come to the conclusion that septohippocampal system is one of the center for saving photoperiodic information.  相似文献

A radioisotope assay for the measurement of glutamine synthetase activity has been developed in which tandemly arranged ion-exchange columns of Dowex 1-acetate and Amberlite CG-50 (H+) are used to separate the product, [14C]glutamine, from unreacted [U-14C]glutamate and other labeled compounds, particularly γ-aminobutyrate, that are formed by competing reactions. The technique is sensitive, reproducible, and suitable for multiple determinations. The assay has been used successfully to measure glutamine synthetase activity in neural and nonneural tissues which contain appreciable amounts of glutamate decarboxylase activity.  相似文献

Theobroma cacao L. plants over-expressing a cacao class I chitinase gene (TcChi1) under the control of a modified CaMV-35S promoter were obtained by Agrobacterium-mediated transformation of somatic embryo cotyledons. Southern blot analysis confirmed insertion of the transgene in eight independent lines. High levels of TcChi1 transgene expression in the transgenic lines were confirmed by northern blot analysis. Chitinase activity levels were measured using an in vitro fluorometric assay. The transgene was expressed at varying levels in the different transgenic lines with up to a sixfold increase of endochitinase activity compared to non-transgenic and transgenic control plants. The in vivo antifungal activity of the transgene against the foliar pathogen Colletotrichum gloeosporioides was evaluated using a cacao leaf disk bioassay. The assay demonstrated that the TcChi1 transgenic cacao leaves significantly inhibited the growth of the fungus and the development of leaf necrosis compared to controls when leaves were wound inoculated with 5,000 spores. These results demonstrate for the first time the utility of the cacao transformation system as a tool for gene functional analysis and the potential utility of the cacao chitinase gene for increasing fungal pathogen resistance in cacao.  相似文献

We studied the effect of injections of melatonin and modifications of the duration of illumination on the activity of 5-nucleotidase, an enzyme providing synthesis of adenosine, in the forebrain of juvenile male albino rats. The measurements were performed under conditions of acute hypobaric hypoxia. We found that, under conditions of natural illumination, neither isolated injections of melatonin nor acute hypoxia noticeably changed the activity of 5-nucleotidase. At the same time, acute hypoxia combined with melatonin injections increased the activity of this enzyme. A similar noticeable rise in the activity of 5-nucleotidase was observed after melatonin injections in normoxic animals kept in constant darkness, and in rats subjected to hypoxia without the above injections but under conditions of constant illumination. These data allow us to suppose that melatonin (whose level in the extracellular medium is a factor providing synchronization of endogenous temporal rhythms) stimulates 5-nucleotidase-mediated production of adenosine in brain neurons. Acute hypoxia promotes such an effect of melatonin.  相似文献

The influence of modifications of normal photoperiodicity on the state of c-fos (gene of immediate functional response) in neurons of the suprachiasmatic nuclei (SChNs) of the rat hypothalamus was examined; samples were taken during the subjective day and night. In animals kept under normal conditions of alternation of light and darkness, expression of the product of this gene and marker of its activation (c-Fos protein) demonstrated a rather clear circadian pattern, with a greater level of the immunoreactivity of this protein at the day period. Constant illumination for 7 days disturbed the rhythm of the c-fos activity and smoothed circadian variations of the level of immunoreactivity of c-Fos. Under conditions of light deprivation of a similar duration, we observed a significant (more than twofold) increase in the indices of concentration of c-Fos and its content in the nuclei of SChN neurons, as well of the total content of this protein in SChN slices, at the day phase. Possible mechanisms of the influence of modifications of the photoperiod on the state of the c-fos gene in SChN cells, in particular the relation of the respective changes to variations of the level of melatonin, are discussed. It is emphasized that natural photoperiodicity and its experimental modifications result in noticeable shifts of the geometrical dimensions of the nuclei of SChN neurons. The effect of acute hypoxic hypobaric hypoxia on the content of reduced glutathione and the activity of glutathione peroxidase, glutathione reductase, glucose-6-phosphate dehydrogenase and glutathione S-transferase, as well as 5'-nucleotidase in homogenates of juvenile male rats under conditions of varying photoperiodic duration: natural conditions of illumination, continuous illumination and continuous darkness were studied. Photoperiodic changes were revealed in the glutathione system of the control animals: the activity of glutathione peroxidase, glutathione reductase and glucose-6-phosphate dehydrogenase reduces under constant light, while the activity of glutathione peroxidase and glutathione S-transferase increases under conditions of constant darkness. The greatest inhibitory effect on the state of the glutathione system is brought about by constant light in case of acute hypoxia: the content of reduced glutathione decreases along with a sharp drop of the activity of glutathione S-transferase and glucose-6-phosphate dehydrogenase, observed against the background of decreased glutathione reductase activity. Permanent dark conditions eliminate partially or completely the negative effect of acute hypoxia on the glutathione system of the brain. The obtained results are indicator of a possibility of protecting role of melatonin in case of acute hypoxia.  相似文献

Somatic embryogenesis is an in vitro clonal propagation method with potential to contribute to the improvement of cacao varieties. Before using this technology for commercial production, it is essential that somatic embryogenesis-derived plants be tested in field conditions. Therefore, we established a field test at Union Vale Estate, Saint Lucia. Thirty- to 50-yr-old trees were selected for clonal propagation as potentially high yielding based on local farmers observations. Clonal plants were propagated in vitro from immature flowers by embryogenesis and micropropagation. Multiple plants from nine genotypes were acclimated to greenhouse conditions then returned to Saint Lucia and planted in a field. Orthotropic rooted cuttings and locally propagated open pollinated seedlings were also planted for a total of 214 trees. Growth data were collected every 4–6 mo. including: stem diameter, stem height, length of the longest jorquette branch, number of jorquette branches, and dates of first flowering and fruiting. At 4.5 yr after planting in the field there were no major differences in all growth parameters among the propagation methods evaluated with exception of the orthotropic rooted cuttings. Trees grown from seeds were slightly taller then trees propagated by the other methods. Trees propagated as orthotropic rooted cuttings exhibited smaller average stem diameters, shorter stem heights to the jorquette, and shorter jorquette branches. We concluded that somatic embryo-derived plants demonstrated normal phenotypes in field conditions and have growth parameters similar to plants propagated by traditional methods.  相似文献
