The effects of ethylene pretreatments (500 nl/l for 1 h) and treatments (100 nl/1 to 1000 nl/l for 6 h) on elongation and gravireaction of apical maize root segments were tested in light and in the dark. When growing roots are placed in a horizontal position gravity induces a positive curvature. It is classically considered to be the consequence of a faster elongation rate by the upper side compared to the lower side. A critical examination indicates that the gravireaction is caused by differential cell extension depending on several processes. Some of the endogenous regulators which may control the growth and gravitropism of elongating roots are briefly presented. The growth inhibitors produced or released from the root cap move preferentially in a basipetal direction and accumulate in the lower side of the elongation zone of horizontally maintained roots. The identity of these compounds is far from clear, but one of these inhibitors could be abscisic acid (ABA). However, indol-3y1 acetic acid (IAA) is also important for root growth and gravitropism. ABA may interact with IAA. Two other aspects of root cell extension have also to be carefully considered. An elongation gradient measured from the tip to the base of the root was found to be important for the growth of both vertical and horizontal gravireactive roots. It was changed significantly during the gravipresentation and can be considered as the origin of the differential elongation. Sephadex beads have been used as both growth markers and as monitors of surface pH changes when they contain some pH indicator. This technique has shown that the distribution of cell extension along the main root axis is related to a pH gradient, the proton efflux being larger for faster growing parts of roots. A lateral movement of calcium is obtained when Ca2+ is applied across the tips of horizontally placed roots with a preferential transport towards the lower side. Endogenous calcium, which may accumulate inside the endoplasmic reticulum of some cap cells, may also act in the gravireception. These observations and several others strongly suggest that calcium may play an essential role in controlling root growth and several steps of the root gravireaction. 相似文献

The effects of a synthetic growth promoter, 4-ethoxy-l-( p -tolyl)-S-triazine-2,6 (1H, 3H)-dione [TA], on growth and gravireaction of Zea mays L. (cv. LG 11) roots were investigated. In horizontal, intact roots, pretreatment with TA at 4 × 10−4 M inhibited the gravireaction. If the pretreated roots were rinsed with a buffer solution before incubation, the TA effect was reduced, indicating that a continuous presence of TA was necessary for its maximal activity. On the other hand, the TA pretreatment (1×10−5 , 1×10−4 and 4 × 10−4 M ) promoted the elongation of these roots. The TA effect was stronger for illuminated roots than for those kept in darkness. TA also decreased the lateral curvature of half-decapitated roots maintained vertically in light. This indicates that the action of TA could be associated with some growth inhibiting substances produced or released in cap cells. 相似文献

DNA-oriented samples of various origins were studied under different conditions of humidiity and sodium chloride content by means of infrared spectroscopy. (1) Oriented DNA (M. Lysodeikticus, E. coli, calf thymus and salmon sperm) films at 3–4% sodium chloride yield polarized spectra which show drastic changes at relative humidities (r.h.) between 94% and 0% indicative of conformational changes: B form → a form → disordered form The measurements of the infrared dichroism at frequencies of about 1230 cm?1 and at about 1090 cm?1 allow one to determine the orientation of the phosphate group, whereas the measurements at 1710 cm?1 characterize the base orientation. At humidities higher than 90% r.h. (B form) the bisector of OPO forms an angle of 70° relative to the helix axis, whereas at lower humidities, between 75% and 50% r.h. (A form) a rotation to about 45° is observed. Simultaneously, the 0—0 line of phosphate group changes its orientation from 55° to 65° to the helix when B → A transition takes place. The results are in general agreement with that of X-ray diffraction and allow one to determine the orientation of the phosphate group with greater precision. (2) The B–A conformational change is not observed for satellite DNA, isolated from Cancer pagurus, of which the guanine + cytosine content is below 5%. As a function of decreasing humidities, one observes the transition: B form → disordered form A diagram of conformational changes of DNA's as a function of base composition and of r.h., suggests that B–A transition will occur for DNA of relatively higher G + C content, whereas for high (A + T) content, base sequence may be of importance. Effect of zeatin on the growth and indolyl-3-acetic acid and abscisic acid levels in maize roots 总被引:1,自引:0,他引:1
Elongation, indolyl-3-acetic acid (IAA) and abscisic acid (ABA) levels, – gas chromatography-mass spectrometry quantification –, in the elongating zone were analysed for maize ( Zea mays L., Cv. LG11) roots immersed in buffer solution with or without zeatin (Z). The effect of Z depends on the initial extension rate of roots. The slower growing roots are more strongly inhibited by Z (10−7 −10−5 M ) and they show a greater increase in IAA and ABA content. When compared to the rapidly growing roots, the larger reactivity of the 'slow'ones cannot be attributed to a higher Z uptake as shown when using [14 C]-Z. It is suggested that Z could regulate root elongation by acting on the IAA and/or ABA level. The comparative action of these two hormones is discussed. 相似文献

Synthesis of indole-3-acetic acid (IAA), using stable-isotope incorporation, was investigated in Zea mays L. Incorporation of 2H from 2H2O into IAA molecules was shown to occur in intact plantlets and excised primary roots cultured in vitro. For several species of lepidoptera, most of the approximately 350-bp
mitochondrial control-region sequences were determined. Six of these
species are in one genus, Jalmenus; are closely related; and are believed
to have undergone recent rapid speciation. Recent speciation was supported
by the observation of low interspecific sequence divergence. Thus, no
useful phylogeny could be constructed for the genus. Despite a surprising
conservation of control-region length, there was little conservation of
primary sequences either among the three lepidopteran genera or between
lepidoptera and Drosophila. Analysis of secondary structure indicated only
one possible feature in common--inferred stem loops with higher-than-random
folding energies-- although the positions of the structures in different
species were unrelated to regions of primary sequence similarity. We
suggest that the conserved, short length of control regions is related to
the observed lack of heteroplasmy in lepidopteran mitochondrial genomes. In
addition, determination of flanking sequences for one Jalmenus species
indicated (i) only weak support for the available model of insect 12S rRNA
structure and (ii) that tRNA translocation is a frequent event in the
evolution of insect mitochondrial genomes.

The cell-wall polysaccharides from different parts of maize roots have been analysed. The arabinose, galactose and mannose contents are influenced by cell differentiation, whereas xylose, rhamnose and uronic-acid contents are not. In cap cells, the pectin content is low but rhamnose and fucose are present in larger quantities. The cell-wall polysaccharides from cells of the elongation zone and their respective regenerating protoplasts were also analysed. The walls of the protoplasts contained higher xylose and mannose levels and a much lower level of cellulose than the cells from which they were derived. 相似文献

Protoplasts were isolated from cortical cells of the elongating zone of maize (Zea mays L. cv. LG 11) roots and submitted to microelectrophoresis. Significant and transient differences in zeta potential between protoplasts from upper and lower root sides were compared with the gravireaction and the differential elongation of these roots. The maximum difference in the zeta potential was obtained between protoplasts from the upper and lower cortical cells after 90 min, exactly the time of gravipresentation for which the maximum rate of gravireaction was observed. In addition, this almost corresponded to the time for which the difference between the elongation rates of upper and lower sides of the extending zone began to increase. Consequently, the changes in the charges of the plasmalemma of the cortical cells from the growing part of roots could be more or less directly related to the root graviresponse. 相似文献
