The purpose of these studies was to determine whether treatment of newborn rats with exogenous FSH or hCG would alter the concentration or size of testicular macrophages. Animals were injected once daily with various doses of FSH, hCG, or vehicle for 8-10 days beginning the day following birth. After immunohistochemical labeling of the macrophages with a monoclonal antibody specific for rat macrophages, the concentration and size of macrophages were determined by use of a point-counting method. Body weight, testis weight, and serum levels of testosterone and FSH were also measured. It was found that hCG significantly increased the concentration of macrophages within the interstitium but did not affect the size of the cells. Both testicular weight and serum testosterone concentrations increased after hCG treatment. Although FSH increased the weight of the testis, neither the size nor concentration of macrophages was altered. These results raise the possibility that the number of macrophages within the interstitial compartment of the normally maturing rat testis is under the control of LH.  相似文献

Though predation, productivity (nutrient richness), spatial heterogeneity, and disturbance regimes are known to influence species diversity, interactions between these factors remain largely unknown. Predation has been shown to interact with productivity and with spatial heterogeneity, but few experimental studies have focused on how predation and disturbance interact to influence prey diversity. We used theory and experiments to investigate how these factors influence diversification of Pseudomonas fluorescens by manipulating both predation (presence or absence of Bdellovibrio bacteriovorus) and disturbance (frequency and intensity of disturbance). Our results show that in a homogeneous environment, predation is essential to promote prey species diversity. However, in most but not all treatments, elevated diversity was transitory, implying that the effect of predation on diversity was strongly influenced by disturbance. Both our experimental and theoretical results suggest that disturbance interacts with predation by modifying the interplay of resource and apparent competition among prey.  相似文献

In male rats, methionine-enkephalin immunoreactivity (enkephalin-ir) has been observed in the dorsal lateral nucleus (DLN), a longitudinal pool of motoneurons in the lumbar spinal cord. Within the DLN a mediodorsal crescent of intense enkephalin-ir staining surrounds the motoneurons innervating the ischiocavernosus muscle of the penis, which suggests a function of the enkephalinergic afferents in male copulatory activities. The present study attempted to determine the roles of gender and adult exposure to androgen in shaping the striking subnuclear distribution of enkephalin-ir. Transverse sections through L5-6 were obtained from mature male and female rats that were gonadally intact, gonadectomized, or gonadectomized and treated with testosterone, as well as from male rats genetically deficient in androgen receptors (Tfm). The sections were incubated with primary antiserum raised against methionine enkephalin and bound antibodies were visualized using the avidin-biotin-peroxidase technique. A microphotometer was used to compare the staining density in laminae I-II of the dorsal horn, ventral grey matter, and the DLN. In all groups the DLN stained more darkly than the ventral grey, demonstrating the presence of enkephalin-ir in the DLN regardless of gender or exposure to androgen. However, the mediodorsal crescent of dense staining in the DLN was obvious only in gonadally intact males, while the entire DLN stained darkly in both sexes of gonadectomized rats treated with androgen. Therefore, the preferential distribution of enkephalin-ir in the mediodorsal crescent of the DLN is sexually dimorphic though the overall content of enkephalin-ir within the DLN responds to androgen.  相似文献

Middle-aged female rats cease to display estrous cycles and exhibit a state of persistent estrus (PE). Under PE and chronic anovulatory conditions, there is a lack of spontaneous luteinizing hormone (LH) surges, but ovulations often occur after the females are caged with males. This study examined the effects of caging and mating with male rats on LH release in PE females, and assessed their reproductive capacity. Young cyclic rats received intra-atrial cannulae, and subsequently were sampled every 90 min during 1400-2130 h on proestrus for plasma LH measurement. PE females were similarly cannulated and sampled. Two days later, these PE rats received an s.c. injection of estradiol benzoate (EB) and were sampled on the following day. While young females exhibited the proestrous LH surge, PE rats maintained low plasma LH levels persistently and were unable to increase LH release after EB administration. On the other hand, when cannulated PE females were caged with fertile males, 92% displayed lordotic responses, and 75% of those sexually receptive PE females exhibited LH surges followed by ovulation. The initiations of the lordotic response and the LH surge both were more rapid in PE females caged with males beginning at 1500 h than at 1400 h. In contrast, when individual PE rats were placed in clean boxes without males, only one of 13 females showed an increase in LH release followed by ovulation. In contrast to other male mice, many hybrid B6D2F1 males continue to mate after being castrated in adulthood. This persistent sexual behavior in the castrated hybrids presumably is based upon heterozygosity that might occur within individual loci or between different loci. Those possibilities were discriminated by studying BXD recombinant inbred mice. In BXD strains, individual loci are homozygous, but heterozygosity has arisen between separate loci. Males in two of six BXD strains persisted in copulating for 5 mo after being castrated. Thus, it is concluded that retention of masculine sexual behavior in castrated mice is a function of heterozygosity between loci.  相似文献

It is commonly believed that the scent-marking activity of female mammals is elevated when they are sexually receptive, yet urine-marking by female mice does not vary in relation to their estrous cycles. We examined the marking behavior of female mice during their reproductive cycle with attention to their postpartum period of sexual receptivity. In the first experiment, females were allowed to mark prior to mating, during gestation, on the evening of parturition, during lactation, and after weaning their pups. They were exposed to a male or female mouse or nothing at each testing period in a counter-balanced fashion. Overall, the subjects marked most frequently in the presence of a male stimulus; their marking rates did not differ when they were exposed to another female or to nothing. The females' marking remained relatively steady through gestation, declined dramatically at parturition, then gradually returned to the levels observed prior to mating. In the second experiment, multiparous females were allowed to mark while they were simultaneously lactating and pregnant. Compared to unmated females, the subjects' marking frequency was low at parturition, declined further to its lowest level about 1 week later, then regained the preceding low level prior to weaning the initial litter and delivering the next. Together these results demonstrate that urine-marking by female mice is diminished just after parturition. Thus, in contrast to some other species, the marking behavior of female mice apparently is not enhanced around the time they are sexually receptive. Because female mice generally mark most frequently when males are nearby, we postulate that a major function of this behavior is to evoke endocrine and behavioral responses in proximate males rather than to attract distant males.  相似文献

Schizonts of all rodent Plasmodium studied (Plasmodium yoelii, P. chabaudi, P. vinckei) show a characteristic morphology when they are completely mature: rounded or slightly elongate merozoites, completely detached from the pigment mass. At this stage, they are localized principally in the spleen and the lungs but, in impression smears of these organs they show two different aspects. In the spleen, schizonts are either inside the host erythrocyte or extraglobular but still close to a pigment mass; free merozoites are rare. In the lungs, on the contrary, merozoites are often free and dispersed; electron microscopy showed them to lie against the endothelium. Work by physiologists has shown the blood circulation in the alveoli to be much slowed down. Free merozoites, lined against the endothelium of relatively rigid capillaries, are in the best possible conditions to make contact with the intact red blood cells. Lungs appear to be the privileged site for the invasion of erythrocytes by the merozoites.  相似文献

Experiments performed during the last few years, lead us to hypothesise the existence of latent asexual forms of murine Plasmodium. In the present report we examined the organs of infected animals and describe novel structures, which we call merophores, containing merozoites which have resisted lysis seen with other asexual stage parasites. We propose that these merozoites represent a latent form of the parasite. Merophores were also found in the lymphatic circulation, and were demonstrated by subinoculation to have retained their viability. Depending on the parasite species two types of merophores were observed. For P. yoelii nigeriensis merophore sacks, with the latent merozoites found inside vesicles, were usually observed. Merophore leucocytes, where latent merozoites dispersed in the cytoplasm of macrophages or neutrophils, were solely seen with P. vinckei petteri. Both structures were seen in P. chabaudi chabaudi infections. Merophores were found in lymph nodes of rodents after the asexual parasitaemia had apparently subsided. They were formed soon after schizogony, principally in the spleen, either by pitting or by macrophage phagocytosis. Merophore numbers appeared to be proportional to the number of maturing schizonts. We propose that merophore formation and their circulation in the lymphatics play an important role in the pattern of recrudescences and chronicity of rodent malaria infections. It is further suggested that the lymphatic network, a privileged pathway for many parasites, might play a similar role in human malaria infections.  相似文献

Successful adaptation relies on the ability to learn the consequence of our actions in different environments. However, understanding the neural bases of this ability still represents one of the great challenges of system neuroscience. In fact, the neuronal plasticity changes occurring during learning cannot be fully controlled experimentally and their evolution is hidden. Our approach is to provide hypotheses about the structure and dynamics of the hidden plasticity changes using behavioral learning theory. In fact, behavioral models of animal learning provide testable predictions about the hidden learning representations by formalizing their relation with the observables of the experiment (stimuli, actions and outcomes). Thus, we can understand whether and how the predicted learning processes are represented at the neural level by estimating their evolution and correlating them with neural data. Here, we present a bayesian model approach to estimate the evolution of the internal learning representations from the observations of the experiment (state estimation), and to identify the set of models' parameters (parameter estimation) and the class of behavioral model (model selection) that are most likely to have generated a given sequence of actions and outcomes. More precisely, we use Sequential Monte Carlo methods for state estimation and the maximum likelihood principle (MLP) for model selection and parameter estimation. We show that the method recovers simulated trajectories of learning sessions on a single-trial basis and provides predictions about the activity of different categories of neurons that should participate in the learning process. By correlating the estimated evolutions of the learning variables, we will be able to test the validity of different models of instrumental learning and possibly identify the neural bases of learning.  相似文献

This experiment was designed to determine whether the reflexive LH pulses induced in male mice by exposure to a female are dependent upon the presence of the testes. B6D2F1 males were studied because, unlike most males, these hybrids remain sexually active indefinitely after castration, demonstrating that they still recognize females and are sexually arousable. We reasoned that if LH reflexes occur independently of the testes, then exposing castrated B6D2F1 males to a female should elicit a pulse. We observed female-induced pulses in 67% of intact, naive males and 80% of intact, sexually experienced males, but they were absent in castrated, sexually experienced males. Spontaneous pulses were too frequent in castrated, naive males to distinguish potential LH reflexes. Thus, the induction of LH reflexes depends upon the presence of the testes in these hybrid males. Secondarily we found more frequent spontaneous pulses in sexually naive than in experienced mice regardless of their gonadal status. These unexpected results imply that long-term mating activity slows the pace of spontaneous LH pulses in these hybrids. Considered together, the differential effects of the testes and sexual experience on both types of LH pulses suggest that two distinct pulse-generating mechanisms exist in male mice.  相似文献
