The diversity of class II major histocompatibility complex (MHC) loci was investigated in the brushtail possum, an important marsupial pest species in New Zealand. Immunocontraception, a form of fertility control that generates an autoimmune response, is being developed as a population control method for the possum. Because the immune response is partly under genetic control, an understanding of immunogenetics in possum will be crucial to the development of immunocontraceptive vaccines. MHC molecules are critical in the vertebrate immune response. Class II MHC molecules bind and present exogenously derived peptides to T lymphocytes and may be important in the presentation of immunocontraceptives. We used polymerase chain reaction primers designed to amplify the peptide binding region of possum class II MHC genes to isolate sequences from 49 animals. We have previously described 19 novel alleles from the DAB locus in the possum (Holland et al., Immunogenetics 60:449–460, 2008). Here, we report on another 11 novel alleles isolated from possum DAB, making this the most diverse marsupial locus described so far. This high level of diversity indicates that DAB is an important MHC locus in the possum and will need to be taken into account in the design of immunocontraceptive vaccines.  相似文献

Sea surface temperature (SST) time-series from the southwest Atlantic and the El Ni?o 4 region in the western Pacific were compared to an index of annual calving success of the southern right whale (Eubalaena australis) breeding in Argentina. There was a strong relationship between right whale calving output and SST anomalies at South Georgia in the autumn of the previous year and also with mean El Ni?o 4 SST anomalies delayed by 6 years. These results extend similar observations from other krill predators and show clear linkages between global climate signals and the biological processes affecting whale population dynamics.  相似文献

Diagnostic catheters based on fibre Bragg gratings (FBG's) are proving to be highly effective for measurement of the muscular activity associated with motility in the human gut. While the primary muscular contractions that generate peristalsis are circumferential in nature, it has long been known that there is also a component of longitudinal contractility present, acting in harmony with the circumferential component to improve the overall efficiency of material movement. We report the detection of longitudinal motion in mammalian intestine using an FBG technique that should be viable for similar detection in humans. The longitudinal sensors have been combined with our previously reported FBG pressure sensing elements to form a composite catheter that allows the relative phase between the two components to be detected. Trypanosoma cruzi, the causative agent of human Chagas disease, is endemic to the southern region of the United States where it routinely infects many host species. The indoor/outdoor housing configuration used in many non-human primate research and breeding facilities in the southern of the USA provides the opportunity for infection by T. cruzi and thus provides source material for in-depth investigation of host and parasite dynamics in a natural host species under highly controlled and restricted conditions. For cynomolgus macaques housed at such a facility, we used a combination of serial blood quantitative PCR (qPCR) and hemoculture to confirm infection in >92% of seropositive animals, although each method alone failed to detect infection in >20% of cases. Parasite isolates obtained from 43 of the 64 seropositive macaques were of 2 broad genetic types (discrete typing units, (DTU’s) I and IV); both within and between these DTU groupings, isolates displayed a wide variation in growth characteristics and virulence, elicited host immune responses, and susceptibility to drug treatment in a mouse model. Likewise, the macaques displayed a diversity in T cell and antibody response profiles that rarely correlated with parasite DTU type, minimum length of infection, or age of the primate. This study reveals the complexity of infection dynamics, parasite phenotypes, and immune response patterns that can occur in a primate group, despite being housed in a uniform environment at a single location, and the limited time period over which the T. cruzi infections were established.  相似文献

Little is known about bee communities and pollination services conservation strategies in sub-Sahara Africa. A study was conducted at 26 different sites with varying local landscape characteristics in farmlands of central Uganda in 2006. Bees were sampled using coloured pantraps, handnet and line transect counts. Overall 80,883 bee individuals from 6 families and 652 species were encountered. The bee fauna was characterized by a lower diversity of Melittidae and Andrenidae and a high diversity of Apidae, Megachilidae and Halictidae. Megachile and Lasioglossum were the two most species-rich genera. The most abundant species was Apis mellifera adansonii Linnaeus (23 % of total individuals) followed by Hypotrigona gribodoi Magretti (19 %), Meliponula ferruginea Lepeletier (13 %), Lasioglossum ugandicum Cockerell (7 %), Apis mellifera scutellata Latreille (6 %), Allodapula acutigera Cockerell (6 %), Ceratina rufigastra Cockerell (5 %), Ceratina tanganyicensis Strand (5 %), Braunsapis angolensis Cockerell (5 %), Megachile rufipes Fabricius (5 %), Meliponula bocandei Spinola (5 %) and Seladonia jucundus Smith (5 %). The mean number of species recorded per study site per day ranged between 14 and 49, whereas the abundance ranged between 188 and 1,859 individuals. Study sites in areas with intense land-use had species-poor bee communities compared to sites with medium to low land-use intensities. Study sites with riparian forest fragments and wetlands, or with forest fallows in their vicinity had significantly (P < 0.05) higher species richness and diversity than sites dominated by small-scale monoculture/polyculture fields or sites dominated by either simple or complex traditional agroforestry systems. An ordination analysis also revealed that bee communities were significantly (P < 0.01) influenced by the presence of semi-natural habitats (woodlands, fallows) and forest fragments in the surrounding of fields. Thus, natural and semi-natural habitats are of great value for afrotropical farmland bee communities. There is a need to put in place strategies and policies for semi-natural and forest fragments preservation for spatio-temporal stability of pollination services in rural landscapes. Farmers are recommended to increase on-farm trees cover to safeguard and enhance pollination function and services in fields. Mimicking natural vegetation through promoting establishment of forest plantations and village community forestry in rural landscapes is also critical for conserving pollination services.  相似文献

A gene involved in N-acyl homoserine lactone (N-AHSL) degradation was identified by screening a genomic library of Rhodococcus erythropolis strain W2. This gene, named qsdA (for quorum-sensing signal degradation), encodes an N-AHSL lactonase unrelated to the two previously characterized N-AHSL-degrading enzymes, i.e., the lactonase AiiA and the amidohydrolase AiiD. QsdA is related to phosphotriesterases and constitutes the reference of a novel class of N-AHSL degradation enzymes. It confers the ability to inactivate N-AHSLs with an acyl chain ranging from C(6) to C(14), with or without substitution at carbon 3. Screening of a collection of 15 Rhodococcus strains and strains closely related to this genus clearly highlighted the relationship between the ability to degrade N-AHSLs and the presence of the qsdA gene in Rhodococcus. Bacteria harboring the qsdA gene interfere very efficiently with quorum-sensing-regulated functions, demonstrating that qsdA is a valuable tool for developing quorum-quenching procedures.  相似文献

The impuritiy profiles of acetonitrile solutions of the four standard O-cyanoethyl-N,N-diisopropyl-phosphoramidites of 5'-O-dimethoxytrityl (DMT) protected deoxyribonucleosides (dG(ib), dA(bz), dC(bz), T) were analyzed by HPLC-MS. The solution stability of the phosphoramidites decreases in the order T, dC>dA>dG. After five weeks storage under inert gas atmosphere the amidite purity was reduced by 2% (T, dC), 6% (dA), and 39% (dG), respectively. The main degradation pathways involve hydrolysis, elimination of acrylonitrile and autocatalytic acrylonitrile-induced formation of cyanoethyl phosphonoamidates. Consequently, the rate of degradation is reduced by reducing the water concentration in solution with molecular sieves and by lowering the amidite concentration. Acid-catalyzed hydrolysis could also be reduced by addition of small amounts of base.  相似文献

Many species carry out their most interesting activities wherethey cannot readily be observed or monitored. Marine mammalsare extreme among this group, accomplishing their most astoundingactivities both distant from land and deep in the sea. Collection,storage and transmission of data about these activities areconstrained by the energy requirements and size of the recordingloggers and transmitters. The more bits of information collected,stored and transmitted, the more battery is required and thelarger the tag must be. We therefore need to be selective aboutthe information we collect, while maintaining detail and fidelity.To accomplish this in the study of marine mammals, we have designed"intelligent" data logger/transmitters that provide context-drivendata compression, data relay, and automated data base storage.We later combine these data with remotely sensed environmentalinformation and other oceanographic data sets to recreate theenvironmental context for the animal's activity, and we displaythe combined data using computer animation techniques. In thisway, the system can provide near real time "observation" ofanimal behavior and physiology from the remotest parts of theglobe.  相似文献

1. AMPA receptor potentiators (ARPs) exhibit antidepressant-like activity in preclinical tests (for example, the forced swim test) that are highly predictive of efficacy in humans. Unlike most currently used antidepressants, ARPs do not elevate extracellular levels of biogenic amines (e.g., 5HT, NE) in prefrontal cortex at doses that are active in the forced swim test.2. The present series of experiments examined the effects of combining the ARP, LY 392098, with biogenic amine-based antidepressants in the forced swim test. Male, NIH Swiss mice were placed in a cylinder of water and observed for attempted escape behaviors and immobility.3. LY 392098 dose-dependently decreased immobility as did a range of classical antidepressants. At doses of LY 392098 below those that decreased immobility, this compound significantly increased the potency with which fluoxetine and citalopram (SSRI antidepressants), imipramine (tricyclic antidepressant), duoxetine (norepinephrine/serotonin uptake blocker), nisoxetine (norepinephrine uptake inhibitor), and rolipram (PDE4 inhibitor) decreased immobility in the forced swim test with potency shifts upward of 5-fold (fluoxetine, imipramine, and rolipram). Likewise, ineffective doses of the traditional antidepressants potentiated the effects LY 392098 with shifts in the dose-effect functions that were 10-fold or more for citalopram, fluoxetine, imipramine, and duloxetine.4. Combined with other evidence for a role of AMPA receptors in the efficacy of antidepressants, the current data suggest that the addition of an ARP may augment the activity and perhaps the onset of the therapeutic effects of biogenic amine and second messenger-based antidepressants.  相似文献
