The effects of 2-methoxyestradiol (2ME), a naturally occurring mammalian metabolite of 17 -estradiol, on adipocyte growth has been investigated in mouse brown adipose tissue precursor cells developed in primary culture. 2ME inhibits brown adipocyte proliferation in a dose-response manner (IC50 = 1.7 × 10-6 M for DNA synthesis), with much higher potency than its hormone precursor 17-estradiol, and cells acquire the typical differentiated morphology - more round with a higher content of triglycerides. 2ME causes similar effects in the immortal brown adipocyte tumor-derived hibernoma cell line HIB 1B and the immortal 3T3-F442A white adipocyte line. These findings suggest a possible role for 2ME in adipocyte proliferation, and probably in the differentiation process, entering the cells in the adipogenic program. 相似文献

Purpose
Porcelain stoneware tile (PST) is currently the ceramic tile of greatest commercial and innovation interest. An environmental life cycle assessment of different varieties of PST was undertaken to enable hotspots to be identified, strategies to be defined, differences between PST varieties to be evaluated and guidance for PST manufacturers to be provided in choosing the Environmental Product Declaration (EPD) programme that best suited their needs according to grouping criteria.Methods
Analysis of previous information allowed three main parameters (thickness, glaze content and mechanical treatment) to be identified in order to encompass all PST variations. Fifteen varieties of PST were thus studied. The coverage of 1 m2 of household floor surface with the different PST varieties for 50 years was defined as functional unit. The study sets out environmental data whose traceability was verified by independent third parties for obtaining 14 EPDs of PST under Spanish EPD programmes.Results and discussion
The study presents PST inventory analysis and environmental impact over the entire life cycle of the studied PST varieties. The natural gas consumed in the manufacturing stage accounted for more than 70% abiotic depletion–fossil fuels and global warming; electricity consumption accounted for more than 60% ozone layer depletion, while the electricity generated by the cogeneration systems avoided significant environmental impacts in the Spanish power grid mix. The variations in PST thickness, amount of glaze and mechanical treatments were evaluated. The PST variety with the lowest environmental impact was the one with the lowest thickness, was unglazed and had no mechanical treatments. Similarly, the PST variety with the highest environmental impact was the one with the greatest thickness, was glazed and had been mechanically treated.Conclusions
The PST life cycle stage with the highest environmental impact was the manufacturing stage. The main hotspots found were production and consumption of energy and raw materials extraction. Variation in thickness was a key factor that proportionally influenced almost all studied impact categories; the quantity of glaze strongly modified abiotic depletion–elements and eutrophication, while the mechanical treatments contributed mainly to ozone depletion. The study of all PST varieties led to the important conclusion, against the current trend, that differences among them were found to be so significant that declaring a number of PSTs within the same EPD is not directly possible, and it needs preliminary verification to ensure compliance with the product category rule.3.

The endogenous cardiac activity rhythm of the Norway lobster Nephrops norvegicus was studied under constant conditions of darkness by means of a computer-aided monitoring system (CAPMON). Time series recordings of the heart rate (beats min?1) were obtained from 47 adult males freshly collected from the continental slope (400–430?m) in the western Mediterranean. Periodogram analysis revealed the occurrence of circadian periodicity (of around 24?h) in most cases. A large percentage of animals showed significant ultradian periods (of around 12 and 18?h). The analysis of the circadian time series revealed the occurrence of peaks of heart rate activity during the expected night phase of the cycle. These results are discussed in relation to the emergence and locomotor activity rhythms of the species. 相似文献

TNF has been implicated in the pathogenesis of type 1 diabetes. When administered early in life, TNF accelerates and increases diabetes in NOD mice. However, when administered late, TNF decreases diabetes incidence and delays onset. TNFR1-deficient NOD mice were fully protected from diabetes and only showed mild peri-insulitis. To further dissect how TNFR1 deficiency affects type 1 diabetes, these mice were crossed to β cell-specific, highly diabetogenic TCR transgenic I-A(g7)-restricted NOD4.1 mice and Kd-restricted NOD8.3 mice. TNFR1-deficient NOD4.1 and NOD8.3 mice were protected from diabetes and had significantly less insulitis compared with wild type NOD4.1 and NOD8.3 controls. Diabetic NOD4.1 mice rejected TNFR1-deficient islet grafts as efficiently as control islets, confirming that TNFR1 signaling is not directly required for β cell destruction. Flow cytometric analysis showed a significant increase in the number of CD4(+)CD25(+)Foxp3(+) T regulatory cells in TNFR1-deficient mice. TNFR1-deficient T regulatory cells were functionally better at suppressing effector cells than were wild type T regulatory cells both in vitro and in vivo. This study suggests that blocking TNF signaling may be beneficial in increasing the function of T regulatory cells and suppression of type 1 diabetes. 相似文献

One of the applications of genomics is to identify genetic markers linked to loci responsible for variation in phenotypic traits, which could be used in breeding programs to select individuals with favorable alleles, particularly at the seedling stage. With this aim, in the framework of the European project FruitBreedomics, we selected five main peach fruit characters and a resistance trait, controlled by major genes with Mendelian inheritance: fruit flesh color Y, fruit skin pubescence G, fruit shape S, sub-acid fruit D, stone adhesion-flesh texture F-M, and resistance to green peach aphid Rm2. They were all previously mapped in Prunus. We then selected three F1 and three F2 progenies segregating for these characters and developed genetic maps of the linkage groups including the major genes, using the single nucleotide polymorphism (SNP) genome-wide scans obtained with the International Peach SNP Consortium (IPSC) 9K SNP array v1. We identified SNPs co-segregating with the characters in all cases. Their positions were in agreement with the known positions of the major genes. The number of SNPs linked to each of these, as well as the size of the physical regions encompassing them, varied depending on the maps. As a result, the number of useful SNPs for marker-assisted selection varied accordingly. As a whole, this study establishes a sound basis for further development of MAS on these characters. Summary The PRms protein is a pathogenesis-related (PR)-like protein whose mRNA accumulates during germination of maize seeds. Expression of the PRms gene is induced after infection of maize seeds with the fungus Fusarium moniliforme. To further our investigations on the expression of the PRms gene we examined the accumulation of PRms mRNA in different tissues of maize seedlings infected with E. moniliforme and studied the effect of fungal elicitors, the mycotoxin moniliformin, the hormone gibberellic acid, and specific chemical agents. Our results indicate that fungal infection, and treatment either with fungal elicitors or with moniliformin, a mycotoxin produced by F. monilforme, increase the steady-state level of PRms mRNA. PRms mRNA accumulation is also stimulated by the application of the hormone gibberellic acid or by treatment with silver nitrate, whereas acetylsalicylic acid has no effect. In situ RNA hybridization in isolated germinating embryo sections demonstrates that the PRms gene is expressed in the scutellum, particularly in a group of inner cells, and in the epithelium lying at the interface of the scutellum and the endosperm. The pattern of expression of the PRms gene closely resembles that found for hydrolytic enzymes, being confined to the scutellum and the aleurone layer of the germinating maize seed. Our results suggest that the PRms protein has a function during the normal process of seed germination that has become adapted to serve among the defence mechanisms induced in response to pathogens during maize seed germination. 相似文献

The transposon Mutator was first identified in maize, and is one of the most active mobile elements in plants. The Arabidopsis thaliana genome contains at least 200 Mutator-like elements (MULEs), which contain the Mutator-like transposase gene, and often additional genes. We have detected a novel type of MULEs in melon (CUMULE), which, besides
the transposase, contains two ubiquitin-like specific protease-like sequences (ULP1). This element is not present in the observed
location in some melon cultivars. Multiple copies of this element exist in the Cucumis melo genome, and it has been detected in other Cucurbitaceae species. Analysis of the A. thaliana genome revealed more than 90 CUMULE-like elements, containing one or two Ulp1-like sequences, although no evidence of mobility exists for these elements. We detected various putative transposable elements
containing ULP1-like sequences in rice. The discovery of these MULEs in melon and Arabidopsis, and the existence of similar
elements in rice and maize, suggest that a proteolytic function may be important for this subset of the MULE transposable
elements.

Retrospective studies have shown the occurrence of episodes of deep or superficial fungal infections in 58 to 81% of HIV/AIDS patients as a result of impairment of cell immunity. We describe a case of disseminated cutaneous dermatophytoses caused by Trichophyton mentagrophytes and Microsporum canis in a patients with AIDS. Diagnostic and therapeutic problems in relation to this unsual presentation are emphasized as well as the importance of an early mycologic diagnosis to prescribe antifungal therapy. 相似文献

The isolation and characterization of cDNA and homologous genomic clones encoding the lignin O-methyltransferase (OMT) from maize is reported. The cDNA clone has been isolated by differential screening of maize root cDNA library. Southern analysis indicates that a single gene codes for this protein. The genomic sequence contains a single 916 bp intron. The deduced protein sequence from DNA shares significant homology with the recently reported lignin-bispecific caffeic acid/5-hydroxyferulic OMTs from alfalfa and aspen. It also shares homology with OMTs from bovine pineal glands and a purple non-sulfur photosynthetic bacterium. The mRNA of this gene is present at different levels in distinct organs of the plant with the highest accumulation detected in the elongation zone of roots. Bacterial extracts from clones containing the maize OMT cDNA show an activity in methylation of caffeic acid to ferulic acid comparable to that existing in the plant extracts. These results indicate that the described gene encodes the caffeic acid 3-O-methyltransferase (COMT) involved in the lignin biosynthesis of maize. 相似文献
