Abstract The composition of the polyol pools in Aspergillus nidulans mycelium during active growth on sucrose depends strongly on pH. At pH 2.5, only mannitol is present. A comparison between nitrate- and ammonium-grown cultures shows stimulation of the arabitol content with nitrate a former nitrogen source. When starved mycelium is incubated either with natural-abundance or 13C-enriched glucose, label appears rapidly in mannitol and arabitol, regardless of the nitrogen source or the pH used.  相似文献

Rat hepatocytes were incubated in the presence of a high concentrationof the hepatopathogenic agent D-galactosamine (GalN), and theeffect on the cellular concentrations of pyrimidine nucleotidesand nucleotide sugars was determined. The UTP pool became depleted.The pools of UMP and CMP in RNA decreased to 72%, indicativefor an inhibition of RNA synthesis. UDP-HexNAc (where HexNAcis GlcNAc + GalNAc) and UDP-HexN (where HexN is GlcN + GalN)levels increased, and those of UDP-hexose and UDP-GlcA (whereGlcA is glucuronic acid) decreased. The cellular concentrationof CTP did not change, whereas that of CMP-NeuAc (where NeuAcis N-acetylneuraminic add) showed a 2-fold increase. Labellingwith [14C]orotic acid and [3H]cytidine showed that the metabolicflow via the de novo pathway was not changed. The depletionof the so-called overflow pool of UTP [Pels Rijcken et al, Biochem.J., 293, 207–213, 1993] caused a release of the feedbackinhibition by UTP and thus an increased flow through the salvagepathway. Finally, it appeared that GalN, when added to hepatocytes,gives rise to a pool of UDP-GlcNAc (where GlcNAc is N-acetylglueosamine)that is separate from the pool of UDP-GlcNAc that is derivedfrom GlcN. The multidrug resistance gene MDR1 (ABCB1) codes for a P-glycoprotein that acts as an ATP-dependent transporter and is involved in removing drugs, xenobiotics, and peptides from the cell. MDR1 is expressed in the brain, kidneys, liver, and gastrointestinal tract. The P-glycoprotein is thought to play a role in individual resistance to xenobiotics and infections. Several polymorphisms of MDR1 are associated with the level of its expression and resistance to various neurodegenerative and gastrointestinal diseases. The allele and haplotype frequencies, genetic differentiation, and linkage disequilibrium for five MDR1 single nucleotide polymorphisms (3435C/T, 2677G/T/A, 1236C/T, +139C/T, and ?1G/A) were studied in the Russian, Tuvinian, and northern and southern Kyrgyz populations. Significant genetic differences were observed between Russians and northern Kyrgyz and between Tuvinians and northern Kyrgyz. The linkage disequilibrium pattern was characterized by high population specificity.  相似文献

If predators lack information on the prey's position, prey have more chance to escape predation and will therefore reach higher population densities. One of the many possible cues that predators may use to find their prey are herbivore-induced plant volatiles. Although their effects on the behaviour of foraging predators have been well studied, little is known about how these prey-related odours affect predator–prey dynamics on a plant. We hypothesise that herbivore-induced plant volatiles provide the major cue eliciting predator arrestment on prey-infested leaves and that the response to these volatiles ultimately leads to lower prey densities. To test this hypothesis experimentally, we created two types of odour-saturated environments: one with herbivore-induced plant volatiles (treatment), and one with green-leaf volatiles (control). An odour-free environment could not be tested because herbivores require plants for population growth. We measured the rate at which predatory mites (Phytoseiulus persimilis) immigrate, emigrate and exploit a single leaf infested by two-spotted spider mites (Tetranychus urticae). The experiments did not show a significant difference between treatment and control. At best, there was a somewhat higher rate of predator (and possibly also prey) emigration in the treatment. The lack of a pronounced difference between treatment and control indicates that at the spatial scale of the experiments random searching for prey was as effective as directional searching. Alternatively, predators were arrested in the prey patch by responding not merely to herbivore-induced plant volatiles, but also to other prey-related cues, such as web and faeces. Based on our current experience we advocate to increase the spatial scale of the experiment (>1m2) and we provide other suggestions for improving the set-up.  相似文献

Background

Clinical practice guidelines (CPGs) are developed to assist health care providers in decision-making. We systematically reviewed existing CPGs on the HDPs (hypertensive disorders of pregnancy) to inform clinical practice.

Methodology & Principal Findings

MEDLINE, EMBASE, Cochrane Central Register of Controlled Trials, Cochrane Methodology Register, Health Technology Assessments, and Database of Abstracts of Reviews of Effects (Ovid interface), Grey Matters, Google Scholar, and personal records were searched for CPGs on the HDPs (Jan/03 to Nov/13) in English, French, Dutch, or German. Of 13 CPGs identified, three were multinational and three developed for community/midwifery use. Length varied from 3–1188 pages and three guidelines did not formulate recommendations. Eight different grading systems were identified for assessing evidence quality and recommendation strength. No guideline scored ≧80% on every domain of the AGREE II, a tool for assessing guideline methodological quality; two CPGs did so for 5/6 domains. Consistency was seen for (i) definitions of hypertension, proteinuria, chronic and gestational hypertension; (ii) pre-eclampsia prevention for women at increased risk: calcium when intake is low and low-dose aspirin, but not vitamins C and E or diuretics; (iii) antihypertensive treatment of severe hypertension; (iv) MgSO4 for eclampsia and severe pre-eclampsia; (v) antenatal corticosteroids at <34 wks when delivery is probable within 7 days; (vi) delivery for women with severe pre-eclampsia pre-viability or pre-eclampsia at term; and (vii) active management of the third stage of labour with oxytocin. Notable inconsistencies were in: (i) definitions of pre-eclampsia and severe pre-eclampsia; (ii) target BP for non-severe hypertension; (iii) timing of delivery for women with pre-eclampsia and severe pre-eclampsia; (iv) MgSO4 for non-severe pre-eclampsia, and (v) postpartum maternal monitoring.

Conclusions

Existing international HDP CPGs have areas of consistency with which clinicians and researchers can work to develop auditable standards, and areas of inconsistency that should be addressed by future research.  相似文献

The limited availability of fossil fuel sources, worldwide rising energy demands and anticipated climate changes attributed to an increase of greenhouse gasses are important driving forces for finding alternative energy sources. Chemically modified biomaterial surfaces (titanium and glass) covered with polyelectrolyte self-assembled films formed by the alternating adsorption of cationic poly-L-lysine (PLL) and anionic poly-L-glutamic acid (PGA) were structurally characterized by atomic force microscopy. Complementary information concerning the thickness and layer-by-layer growth of the films was provided by optical waveguide light-mode spectroscopy. The frequently used ex situ and the rarely used in situ build-up methods were compared. Important aspects of the industrial applicability of these films, their stability in time, and possible differences in their morphology were investigated. The films revealed a granular pattern, with grain diameters of 270 +/- 87 nm for glass (up to 8 bilayers) and 303 +/- 89 nm for titanium (up to 10 bilayers), independently of the build-up procedure. Both surfaces displayed a rehydration capability, the titanium surface exhibiting a better stability in time. The high roughness values observed at acidic or basic pH are related to the degree of ionization of PGA and PLL.  相似文献

Fluorides may affect the oxide layer on titanium surface. Caries preventive mouthwashes or gels contain fluorides and are applied at low pH. The aim of the present work was to study whether various concentrations of fluoride at acidic pH cause changes in the surface structure on the polished region of Ti implants, and alter the adherence and colonization of bacteria. Commercially pure Ti grade 4 discs with a polished surface were treated with a mouthwash containing 0.025% fluoride, a gel containing 1.25% fluoride or a 1% aqueous solution of NaF (pH 4.5). The change of surface roughness of the samples and the colonization of Porphyromonas gingivalis strains were studied by scanning electron microscopy after 5 days of anaerobic incubation. The quantity of the bacterial protein was determined by protein assay analysis. Agents with high fluoride concentration at acidic pH increased the roughness of the Ti surface. A slight increase in the amount of bacteria was found on the surfaces treated with 1% NaF and gel in comparison with the control surface. This study suggested that a high fluoride concentration at acidic pH may hinder the development of a healthy transgingival epithelial junction on Ti implants, due to bacterial colonization.  相似文献

In well-mixed populations of predators and prey, natural selection favors predators with high rates of prey consumption and population growth. When spatial structure prevents the populations from being well mixed, such predators may have a selective disadvantage because they do not make full use of the prey's growth capacity and hence produce fewer propagules. The best strategy then depends on the degree to which predators can monopolize the exploitation of local prey populations, which in turn depends on the spatial structure, the number of migrants, and, in particular, the stochastic nature of the colonization process. To analyze the evolutionary dynamics of predators in a spatially structured predator-prey system, we performed simulations with a metapopulation model that has explicit local dynamics of nonpersistent populations, keeps track of the number of emigrants entering the migration pool, assumes individuals within local populations as well as within the migration pool to be well mixed, and takes stochastic colonization into account. We investigated which of the predator's exploitation strategies are evolutionarily stable and whether these strategies minimize the overall density of prey, as is the case in Lotka-Volterra-type models of competitive exclusion. This was analyzed by pairwise invasibility plots based on short-term simulations and tested by long-term simulation experiments of competition between resident and mutant predator-types that differed in one of the following parameters: the prey-to-predator conversion efficiency, the per capita prey consumption rate, or the per capita emigration rate from local populations. In addition, we asked which of these three strategies are most likely to evolve. Our simulations showed that under selection for conversion efficiency the predator-prey system always goes globally extinct yet persists under selection for consumption or emigration rates and that the evolutionarily stable (ES) exploitation strategies do not maximize local population growth rates. The most successful exploitation strategy minimizes the overall density of prey but does not make it settle exactly at the minimum. The system did not settle at the point where the mean time to co-invasion (i.e., immigration of a second predator in a local prey population) equals the mean local interaction time (an idea borne out from studies on host exploitation strategies in host-pathogen systems) but rather where the mean time to co-invasion was larger. The ES exploitation strategies represent more prudent strategies than the ones that minimize prey density. Finally, we show that-compared to consumption-emigration is a more likely target for selection to achieve prudent exploitation and that prudent exploitation strategies can evolve only provided the prey-to-predator conversion efficiency is subject to constraints.  相似文献
