The formation of dynamic membrane microdomains is an important phenomenon in many signal transduction and membrane trafficking events. It is driven by intrinsic properties of membrane lipids and integral as well as membrane-associated proteins. Here we analyzed the ability of one peripherally associated membrane protein, annexin A2 (AnxA2), to induce the formation of phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2)-rich domains in giant unilamellar vesicles (GUVs) of complex lipid composition. AnxA2 is a cytosolic protein that can bind PI(4,5)P2 and other acidic phospholipids in a Ca2+-dependent manner and that has been implicated in cellular membrane dynamics in endocytosis and exocytosis. We show that AnxA2 binding to GUVs induces lipid phase separation and the recruitment of PI(4,5)P2, cholesterol and glycosphingolipids into larger clusters. This property is observed for the full-length monomeric protein, a mutant derivative comprising the C-terminal protein core domain and for AnxA2 residing in a heterotetrameric complex with its intracellular binding partner S100A10. All AnxA2 derivatives inducing PI(4,5)P2 clustering are also capable of forming interconnections between PI(4,5)P2-rich microdomains of adjacent GUVs. Furthermore, they can induce membrane indentations rich in PI(4,5)P2 and inward budding of these membrane domains into the lumen of GUVs. This inward vesiculation is specific for AnxA2 and not shared with other PI(4,5)P2-binding proteins such as the pleckstrin homology (PH) domain of phospholipase Cδ1. Together our results indicate that annexins such as AnxA2 can efficiently induce membrane deformations after lipid segregation, a mechanism possibly underlying annexin functions in membrane trafficking.  相似文献

Biological membranes are organized into dynamic microdomains that serve as sites for signal transduction and membrane trafficking. The formation and expansion of these microdomains are driven by intrinsic properties of membrane lipids and integral as well as membrane-associated proteins. Annexin A2 (AnxA2) is a peripherally associated membrane protein that can support microdomain formation in a Ca2+-dependent manner and has been implicated in membrane transport processes. Here, we performed a quantitative analysis of the binding of AnxA2 to solid supported membranes containing the annexin binding lipids phosphatidylinositol-4,5-bisphosphate and phosphatidylserine in different compositions. We show that the binding is of high specificity and affinity with dissociation constants ranging between 22.1 and 32.2 nM. We also analyzed binding parameters of a heterotetrameric complex of AnxA2 with its S100A10 protein ligand and show that this complex has a higher affinity for the same membranes with Kd values of 12 to 16.4 nM. Interestingly, binding of the monomeric AnxA2 and the AnxA2-S100A10 complex are characterized by positive cooperativity. This cooperative binding is mediated by the conserved C-terminal annexin core domain of the protein and requires the presence of cholesterol. Together our results reveal for the first time, to our knowledge, that AnxA2 and its derivatives bind cooperatively to membranes containing cholesterol, phosphatidylserine, and/or phosphatidylinositol-4,5-bisphosphate, thus providing a mechanistic model for the lipid clustering activity of AnxA2.  相似文献

Hemp fibers are very interesting natural material for textile and technical applications now. Applying hemp fibers to the apparel sector requires improved quality fibers. In this paper, hemp fibers were modified with sodium hydroxide solutions (5% and 18% w/v), at room and boiling temperature, for different periods of time, and both under tension and slack, in order to partially extract noncellulosic substances, and separate the fiber bundles. The quality of hemp fibers was characterised by determining their chemical composition, fineness, mechanical and sorption properties. The modified hemp fibers were finer, with lower content of lignin, increased flexibility, and in some cases tensile properties were improved. An original method for evaluation of tensile properties of hemp fibers was developed.  相似文献

Sorption properties of TEMPO-oxidized natural and man-made cellulose fibers   总被引:1,自引:0,他引:1  
Cotton and lyocell fibers were oxidized with sodium hypochlorite and catalytic amount of sodium bromide and 2,2,6,6-tetramethylpiperidine-1-oxy radical (TEMPO), under various conditions. Water-insoluble fractions, collected after TEMPO-mediated oxidation, were analyzed and characterized in terms of weight loss, aldehyde and carboxyl contents, and sorption properties. Aldehyde and carboxyl groups were introduced into the oxidized cotton up to 0.321 and 0.795 mmol/g, and into the oxidized lyocell up to 0.634 and 0.7 mmol/g, respectively, where weight loss was generally lower than 12% for cotton and 27% for lyocell. Oxidized cotton and lyocell were shown to exhibit 1.55 and 2.28 times higher moisture sorption than the original fibers, respectively, and water retention values up to about 85% for cotton and 335% for lyocell, while iodine sorption values of oxidized fibers were lower up to 35% for cotton and up to 18% for lyocell than the original fibers.  相似文献

Genotypic and Phenotypic Diversity of Cherry Species Collected in Serbia   总被引:1,自引:0,他引:1  
Genetic diversity of cherry species collected in Serbia has been investigated using 26 simple sequence repeat (SSR) markers developed in Prunus. This material consisted of 77 cherry accessions corresponding to the five following species, Prunus cerasus, Prunus avium, Prunus fruticosa, Prunus mahaleb, and Prunus serrulata. A total of 98 alleles were detected, with an average of 3.7 putative alleles per primer combination. Sixteen unique, species-specific, alleles were detected with nine primer pairs in four species, P. avium, P. cerasus, P. mahaleb, and P. serrulata. The highest number of unique alleles, 8, was observed in P. mahaleb and no species-specific alleles were detected in P. fruticosa. SSR markers generated unique fingerprints for all cherry accessions. Cluster analysis classified accessions into four groups according to their taxonomy, where P. avium and P. cerasus were grouped together, supporting P. avium as one of the progenitors of sour cherry. The highest genetic variability and potential value in rootstock breeding was observed in P. mahaleb and P. serrulata material. Principal component (PC) analysis explained more than 50 % of the total observed phenotypic variability using the first two components. The most important characteristics of PC1 were leaf length and width, fruit taste, color of leaf nectaries, fruit weight, leaf blade margin incisions, petiole length, size of vegetative buds, and length of internode. The most important characteristics of PC2 were shape of leaf blade at base, fruit skin color, and leaf blade length and tip angle. The investigated germplasm proved to be sufficiently genetically diverse for use in breeding programs and development of new cherry cultivars and rootstocks.  相似文献

The domestication of the Eurasian grape (Vitis vinifera ssp. sativa) from its wild ancestor (Vitis vinifera ssp. sylvestris) has long been claimed to have occurred in Transcaucasia where its greatest genetic diversity is found and where very early archaeological evidence, including grape pips and artefacts of a 'wine culture', have been excavated. Whether from Transcaucasia or the nearby Taurus or Zagros Mountains, it is hypothesized that this wine culture spread southwards and eventually westwards around the Mediterranean basin, together with the transplantation of cultivated grape cuttings. However, the existence of morphological differentiation between cultivars from eastern and western ends of the modern distribution of the Eurasian grape suggests the existence of different genetic contribution from local sylvestris populations or multilocal selection and domestication of sylvestris genotypes. To tackle this issue, we analysed chlorotype variation and distribution in 1201 samples of sylvestris and sativa genotypes from the whole area of the species' distribution and studied their genetic relationships. The results suggest the existence of at least two important origins for the cultivated germplasm, one in the Near East and another in the western Mediterranean region, the latter of which gave rise to many of the current Western European cultivars. Indeed, over 70% of the Iberian Peninsula cultivars display chlorotypes that are only compatible with their having derived from western sylvestris populations.  相似文献

Soybean cultivation holds great potential for a sustainable agriculture in Europe, but adaptation remains a central issue. In this large mega‐environment (MEV) study, 75 European cultivars from five early maturity groups (MGs 000–II) were evaluated for maturity‐related traits at 22 locations in 10 countries across Europe. Clustering of the locations based on phenotypic similarity revealed six MEVs in latitudinal direction and suggested several more. Analysis of maturity identified several groups of cultivars with phenotypic similarity that are optimally adapted to the different growing regions in Europe. We identified several haplotypes for the allelic variants at the E1, E2, E3 and E4 genes, with each E haplotype comprising cultivars from different MGs. Cultivars with the same E haplotype can exhibit different flowering and maturity characteristics, suggesting that the genetic control of these traits is more complex and that adaptation involves additional genetic pathways, for example temperature requirement. Taken together, our study allowed the first unified assessment of soybean‐growing regions in Europe and illustrates the strong effect of photoperiod on soybean adaptation and MEV classification, as well as the effects of the E maturity loci for soybean adaptation in Europe.  相似文献

This study investigated the individual influences of hemicelluloses and lignin removal on the water uptake behavior of hemp fibers. Hemp fibers with different content of either hemicelluloses or lignin were obtained by chemical treatment with 17.5% sodium hydroxide or 0.7% sodium chlorite. Various tests (capillary rise method, moisture sorption, water retention power) were applied to evaluate the change in water uptake of modified hemp fibers. The obtained results show that when the content of either hemicelluloses or lignin is reduced progressively by chemical treatment, the capillary properties of hemp fibers are improved, i.e. capillary rise height of modified fibers is increased up to 2.7 times in relation to unmodified fibers. Furthermore, hemicelluloses removal increases the moisture sorption and decreases the water retention values of hemp fibers, while lignin removal decreases the moisture sorption and increases the water retention ability of hemp fibers.  相似文献

 DNA-based fingerprinting technologies have proven useful in genetic similarity studies. RFLP is still most commonly used in the estimation of genetic diversity in plant species, but the recently developed PCR-based marker techniques, RAPDs, SSRs and AFLPs, are playing an increasingly important role in these investigations. Using a set of 33 maize inbred lines we report on a comparison of techniques to evaluate their informativeness and applicability for the study of genetic diversity. The four assays differed in the amount of polymorphism detected. The information content, measured by the expected heterozygosity and the average number of alleles, was higher for SSRs, while the lowest level of polymorphism was obtained with AFLPs. However, AFLPs were the most efficient marker system because of their capacity to reveal several bands in a single amplification. In fact, the assay efficiency index was more than ten-fold higher for AFLPs compared to the other methods. Except for RAPDs, the genetic similarity trees were highly correlated. SSR and AFLP technologies can replace RFLP marker in genetic similarity studies because of their comparable accuracy in genotyping inbred lines selected by pedigree. Bootstrap analysis revealed that, in the set of lines analysed, the number of markers used was sufficient for a reliable estimation of genetic similarity and for a meaningful comparison of marker technologies. Symptoms of bitter rot were observed on apple and pear fruits in the field and in storage in Croatia between 2009 and 2011. Fifteen Colletotrichum isolates from apple and two from pear were collected and identified by sequencing of the ITS1 and ITS2 regions of the ribosomal DNA. Phylogenetic analysis revealed that ten isolates from apple and two isolates from pear could be identified as Colletotrichum fioriniae, five isolates from apple were clustered in Colletotrichum clavatum, while one isolate was in the Colletotrichum acutatum A7 group. All isolates caused typical bitter rot symptoms when inoculated on apple and pear fruits.  相似文献
