This research determined the relationships between blood lead level and zinc protoporphyrin (ZnPROTO), protoporphyrin IX (PROTO), and free erythrocyte protoporphyrin (FEP) levels in a group of 18 ring teal ducks. Blood samples were drawn from two groups of ring teal ducks as part of the routine health maintenance program of the New York Zoological Park. One group of six teals had been exposed to what is believed to be lead-contaminated dust, while the second group of twelve teals was unexposed. Blood samples were analyzed for lead by flameless atomic absorption and for protoporphyrins by fluorescence. Blood lead level and log blood lead level had positive correlations with each of the protoporphyrins: the logarithmic correlations were better than the nonlogarithmic correlations, and PROTO correlated better than ZnPROTO. With one exception, PROTO levels were higher than ZnPROTO levels. The results suggest that PROTO, FEP, or ZnPROTO could serve as a biological indicator of lead poisoning in ring teal ducks.  相似文献

Purpose

Life cycle assessment (LCA) is a data-intensive methodology; therefore, experts usually focus collection efforts on a few activities, while generic data on remaining activities are taken from databases. Even though increased availability of databases has facilitated LCA takeoff, assuring data quality is fundamental to ensure meaningful results and reliable interpretation.

Methods

Ecoinvent has become a global reference for inventory data. Its current version released three impact partition modeling options—the recycled content, “allocation at the point of substitution” (APOS), and consequential models—whose adequate choice is crucial for yielding meaningful assessments. Tutorials and manuals describe the distribution algorithm that backs each system model, to ground decision-making regarding the best fit to a study’s goals. We performed a systematic literature review to investigate—within the papers published on the International Journal of LCA (IJLCA)—how transparently authors addressed the system model choices.

Results and discussion

About 70% of LCA practitioners continued to use earlier versions of ecoinvent after version 3 was launched in 2013. The number of papers using versions 3.x only showed an increased growth trend 2 years later. Eighty-three papers actually adopted the newest version of the database. From those, only 29 papers clearly mentioned the adopted system model. Our SLR also suggests a trend regarding authorship profile of LCA-related studies: the number of studies conducted by practitioners aware of the intricacies of sound modeling of background and foreground data might have been surpassed by those conducted by non-LCA specialists who use LCA as a supporting tool for investigations in applied fields, and merely scratch the surface.

Conclusions

Our results point to a need for a caveat: ecoinvent users must take time to understand the general concept behind each system model and practice one of the most important actions when performing an LCA—state methodological choices clearly.

The familial Alzheimer's disease gene products, presenilin-1 and presenilin-2, have been reported to be functionally involved in amyloid precursor protein processing, notch receptor signaling, and programmed cell death or apoptosis. However, the molecular mechanisms by which presenilins regulate these processes remain unknown. With regard to the latter, we describe a molecular link between presenilins and the apoptotic pathway. Bcl-X(L), an anti-apoptotic member of the Bcl-2 family was shown to interact with the carboxyl-terminal fragments of PS1 and PS2 by the yeast two-hybrid system. In vivo interaction analysis revealed that both PS2 and its naturally occurring carboxyl-terminal products, PS2short and PS2Ccas, associated with Bcl-X(L), whereas the caspase-3-generated amino-terminal PS2Ncas fragment did not. This interaction was corroborated by demonstrating that Bcl-X(L) and PS2 partially co-localized to sites of the vesicular transport system. Functional analysis revealed that presenilins can influence mitochondrial-dependent apoptotic activities, such as cytochrome c release and Bax-mediated apoptosis. Together, these data support a possible role of the Alzheimer's presenilins in modulating the anti-apoptotic effects of Bcl-X(L).  相似文献

Alzheimer's disease (AD) is a progressive neurodegenerative disorder. Although the pathogenesis of AD is unknown, it is widely accepted that AD is caused by extracellular accumulation of a neurotoxic peptide, known as Abeta. Mutations in the beta-amyloid precursor protein (APP), from which Abeta arises by proteolysis, are associated with some forms of familial AD (FAD) and result in increased Abeta production. Two other FAD genes, presenilin-1 and -2, have also been shown to regulate Abeta production; however, studies examining the biological role of these FAD genes suggest an alternative theory for the pathogenesis of AD. In fact, all three genes have been shown to regulate programmed cell death, hinting at the possibility that dysregulation of apoptosis plays a primary role in causing neuronal loss in AD. In an attempt to reconcile these two hypotheses, we investigated APP processing during apoptosis and found that APP is processed by the cell death proteases caspase-6 and -8. APP is cleaved by caspases in the intracellular portion of the protein, in a site distinct from those processed by secretases. Moreover, it represents a general effect of apoptosis, because it occurs during cell death induced by several stimuli both in T cells and in neuronal cells.  相似文献

A previously described derivatization method using trichlorophenylhydrazine was developed for the estimation of malonaldehyde measured by gas-chromatography (GC) with electron-capture detection. The precision and reliability of the procedure are improved here by the use of methylmalonaldehyde as internal standard and by the introduction of a diverter valve at the end of the capillary column to protect the electron-capture detector, respectively.The method was applied to determine malonaldehyde content in bovine plasma samples.  相似文献

Regulated intramembrane proteolysis (RIP) is an emerging paradigm in signal transduction. RIP is mediated by intramembrane-cleaving proteases (I-CliPs), which liberate biologically active nuclear or secreted domains from their membrane-tethered precursor proteins. The yeast Pcp1p/Rbd1p protein is a Rhomboid-like I-CliP that regulates mitochondrial membrane remodeling and fusion through cleavage of Mgm1p, a regulator of these essential activities. Although this ancient function is conserved in PARL (Presenilins-associated Rhomboid-like protein), the mammalian ortholog of Pcp1p/Rbd1p, the two proteins show a strong divergence at their N termini. However, the N terminus of PARL is significantly conserved among vertebrates, particularly among mammals, suggesting that this domain evolved a distinct but still unknown function. Here, we show that the cytosolic N-terminal domain of PARL is cleaved at positions 52-53 (alpha-site) and 77-78 (beta-site). Whereas alpha-cleavage is constitutive and removes the mitochondrial targeting sequence, beta-cleavage appears to be developmentally controlled and dependent on PARL I-CliP activity supplied in trans. The beta-cleavage of PARL liberates Pbeta, a nuclear targeted peptide whose sequence is conserved only in mammals. Thus, in addition to its evolutionarily conserved function in regulating mitochondrial dynamics, PARL might mediate a mammalian-specific, developmentally regulated mitochondria-to-nuclei signaling through regulated proteolysis of its N terminus and release of the Pbeta peptide.  相似文献

Purpose

Sustainability assessments of buildings using the life cycle approach have become more and more common. This includes the assessment of the environmental performance of buildings. However, the influence of the construction products used for the fabric, the finishing, and the technical building equipment of buildings has hardly been described in literature. For this reason, we evaluated the influence of the technical building equipment and its impact on the environment for different residential buildings.

Materials and methods

Five residential buildings were evaluated by applying the methodology of life cycle assessment (LCA) (ISO14040) expressed using quantitative assessment categories according to prEN15978.

Results and discussion

Results show that the optimization of energy performance has already reached a high level in Austria, so that the overall potential for possible improvements is quite low. Especially in low-energy and passive?Chouse-standard residential buildings, the limits for energy optimization in the use phase have mostly been achieved. In contrast to this, the integrated LCA (iLCA) findings attribute a high optimization potential to the construction products used for the technical building equipment as well as to the building fabric and finishing. Additionally, the passive house shows the lowest contribution of the technical building equipment on the overall LCA results.

Conclusions

The iLCA findings suggest that it is recommended to include the technical building equipment for future assessments of the environmental performance of buildings. It is also suggested to use a broad number of environmental indicators for building LCA.  相似文献
