Interaction of Proteus mirabilis with the intestine epithelium in white mongrel mice has been studied by means of bacteriological, toxicological and electron microscopic methods. Introduction of the agar culture of bacteria to the intestine lumen has permitted finding its general toxic action on enterocytes and other cell elements of epithelium which was accompanied by a potentiation of the striated margin membrane degradation but caused no destruction of cells. A damaging factor of protei was connected with the thermostable large-molecular fraction of cultural fluid of bacteria and its action was traced at the first hours of the bacterial contact with the mucous surface of thin intestine. Preliminary weakening of protective barriers of the intestine mucosa permitted protei to penetrate deep into the tissue through the intercellular slots. Colonization of the intestine lumen wall zone by the proteus is a necessary condition of the above-mentioned changes.  相似文献

Using the model of experimental colibacillosis in mice morphological, immunomorphological and morphometrical studies of duodenum 1 to 24 hours after inoculation were performed. Typical dynamics of cellular composition in the intestinal wall, accompanied with increased lymphoplasmacellular infiltration, and dystrophic changes in cells of submucosal and intermuscular neuroplexuses were found. The data obtained testify, that the infectious process at the acute state of experimental colibacillosis has an immune character (immediate hypersensitivity of local type), characterised by increasing in immunoglobulin-containing cells and including in this process of mast and endocrine cells.  相似文献

We studied genetic diversity in 54 populations of nine sexual and apomictic species of the genus Chondrilla (C. acantholepis, C. ambigua, C. brevirostris, C. canescens, C. graminea, C. juncea, C. laticoronata, C. latifolia and C. pauciflora) in SE European Russia and neighboring territories of NW Kazakhstan. We analysed the trnT–trnF region of plastid DNA and the internal transcribed spacer of ribosomal DNA (ITS1–5.8S–ITS2) using statistical parsimony, maximum likelihood and neighbor net methods. Two major evolutionary lineages, roughly corresponding to the two subgenera traditionally recognized in the region, were revealed. Within the first evolutionary lineage (subgenus Brachyrhynchus), the sexual diploid C. ambigua and its putatively hybrid apomictic derivatives C. brevirostris, C. laticoronata and C. pauciflora could be recognized. Their identity was also confirmed by analyses of ISSR markers. The second evolutionary lineage (subgenus Chondrilla) comprises C. juncea, C. acantholepis, C. canescens, C. graminea and C. latifolia in European Russia, but analyses of morphological variability and the genealogy of plastid and nuclear markers favor their treatment as the single facultatively apomictic species C. juncea. The results demonstrate that an apomictic mode of reproduction does not necessarily result in the formation of genetically separated microspecies.  相似文献

Energy status of the novel alkalitolerant Yarrowia lipolytica yeast strain grown at alkaline conditions (pH 9.7) was examined. Cells grown under such severe conditions were found to preserve high respiratory activity. The oxidative phosphorylation system dominated in the energy budget of the cell. A procedure was specially design to isolate tightly coupled mitochondria from yeast cells grown at alkaline conditions. The isolated mitochondrial preparations met known criteria of physiological intactness, as inferred from their ability to maintain distinctive state 4–3 respiration transition upon addition of ADP, high respiratory rates, good respiratory control values, and ADP/O ratios close to the theoretically expected maxima for the substrates used.  相似文献

Biochemistry of vitamins is one of the leading trends in the fundamental researches of A. V. Palladin Institute of Biochemistry from the moment of its foundation in 1925. The Laboratory of Vitamins Biochemistry was organised in 1994, it was reorganized into the Department of Vitamins Biochemistry in 1966, and later it was renamed as the Department of Coenzymes Biochemistry. Now the investigations at the Coenzymes Biochemistry Department headed (from 1986) by G. V. Donchenko, Corr.-Member of the National Academy of Sciences of Ukraine, are directed to estimation of vitamins A, E, B1 and PP action molecular mechanisms. Investigation of specific protein-acceptors of vitamins and their biologically active derivatives is a contemporary and effective methodological approach to the estimation of some molecular mechanisms of vitamins action on cellular metabolism. Considering the challenging theoretical and practical aspects of the further fundamental investigation development in the molecular vitaminology the following items are currently being worked in the Department last time: 1. Study of some molecular mechanisms of thiamine and vitamin PP neurotropic action. These investigations are oriented to clearing some new aspects of noncoenzymic mechanism of its influence on the nervous cell functioning both in the norm and at some nervous diseases. 2. Study of some molecular mechanisms of regulation by means of fat-soluble vitamins A, E and their specific proteins-acceptors of DNA, RNA and protein biosynthesis in the nuclei and mitochondria of actively proliferous cells. These investigations are aimed to the estimation of molecular mechanisms of fat-soluble vitamins participation in the regulation of DNA-dependent synthesis of RNA, RNA-polymerase activity, mechanism of their anticancerogenous effect, vitamin E participation in the realisation of nuclear genetic information. 3. Study of intracellular protein-receptors, which take part in realisation of vitamins and their biologically active derivatives functions in the human and animals' organism. The investigations, directed to study of a role of retinol-binding proteins in exchange of the vitamin A and in biosynthesis of DNA, RNA and proteins, the role of tocopherol-binding proteins in realisation of biological action of vitamin E in cells and thiamine-binding proteins in realisation of neurotropic action of vitamin B1 are actively developed. 4. Investigation of mechanisms of antioxidizing and antiradical biological action of vitamin D3, ecdisterone and related biologically active compounds. Basing on the fundamental researches some vitamins preparations have been created, such as "Carotin-M", "Cardiovit", "Evit-1", "Soevit", "Metovit", "Caratel'ka" and others. The results of fundamental investigation of noncoenzymic thiamine function led us to elaboration of a new hypothesis about molecular mechanism of vitamin B1 neurotropic action. According to the hypothesis the thiamine high neuroactivity is a result of existence in the nervous ending a specific mobile thiamine pool and connection thiamine metabolism with nervous cell membrane potential and acetylcholine metabolism.  相似文献

In the researches carried out on rats with models of chronic alcoholism and alcohol abstinence the most vulnerable to chronic action of alcohol biochemical parameters are revealed: a level of reduced glutathione (it was estimated by the content of free SH-groups in tissues), the content of thiamine diphosphate and thiaminekinase activity in a brain, the content of folic acid in the blood, the content of ubiquinone in the cardiac muscle, RNA/DNA relation in the liver. The data obtained demonstrate first of all the negative influence of alcohol on metabolism of sulfur-containing substances and processes of transmethylation. The results of our investigation have also shown that the part of metabolic changes caused by long-term usage of alcohol, can be caused by direct influence of ethanol or its metabolites on those or other enzymatic proteins or receptors, and their functions can quickly be normalized after the abolition of alcohol (NAD+ contents, alpha-ketoglutarate dehydrogenase activity and some others). More stable changes in other parts of metabolism, that were specified earlier, may be also a result of long-term alcohol consumption.  相似文献
