A crude endo-xylanase produced by Aspergillus niger BCC14405 was investigated for its potential in pre-bleaching of chemical pulp from eucalyptus. The optimal fermentation conditions on the basis of optimization using response surface methodology included cultivation in a complex medium comprising wheat bran, rice bran, and soybean meal supplemented with yeast extract, glucose, peptone, and lactose with a starting pH of 6.0 for 7 d. This resulted in production of 89.5 IU/mL of xylanase with minor cellulase activity. Proteomic analysis using LC/MS/MS revealed that the crude enzyme was a composite of hemicellulolytic enzymes, including endo-β-1,4-xylanase and other hemicellulolytic enzymes attacking arabinoxylan and mannan. Pretreatment of the pulp at a xylanase dosage of 10 IU/g increased the brightness ceiling after the C-Eop-H bleaching step up to 3.0% using a chlorine charge with a C-factor of 0.16-0.20. Xylanase treatment also led to reduction in chlorine charge of at least 20%, with an acceptable brightness level. The enzyme pretreatment resulted in a slight increase in pulp viscosity, suggesting an increase in relative cellulose content. The crude enzyme was potent in the enzyme-aided bleaching of chemical pulp in an environmentally friendly pulping process.  相似文献

Enzymatic modification of pulp is receiving increasing interest for energy reduction at the refining step of the paper-making process. In this study, the production of a multi-fiber modifying enzyme from Mamillisphaeria sp. BCC8893 was optimized in submerged fermentation using a response-surface methodology. Maximal production was obtained in a complex medium comprising wheat bran, soybean, and rice bran supplemented with yeast extract at pH 6.0 and a harvest time of 7 d, resulting in 9.2 IU/mL of carboxymethyl cellulase (CMCase), 14.9 IU/mL of filter paper activity (FPase), and 242.7 IU/mL of xylanase. Treatment of old corrugated container pulp at 0.2-0.3 IU of CMCase/g of pulp led to reductions in refining energy of 8.5-14.8%. The major physical properties were retained, including tensile and compression strength. Proteomic analysis showed that the enzyme was a complex composite of endo-glucanases, cellobiohydrolases, beta-1,4-xylanases, and beta-glucanases belonging to various glycosyl hydrolase families, suggestive of cooperative enzyme action in fiber modification, providing the basis for refining efficiency.  相似文献

Papaya is an economically important plant in Thailand for domestic consumption and export. However, papaya is extremely susceptible to disease caused by the papaya ring spot virus. Although transgenic papaya has been developed, commercial cultivation of transgenic plants in Thailand is still illegal. One concern is cross-pollination to conventional varieties. In this study, windborne-pollen dispersion of papaya (Carica papaya L.) was investigated using geographic information systems (GIS) and remotely sensed data. Pollen traps were placed around a papaya plot in eight geographic directions, with radiuses varying from 5 to 900 m from the plot. Pollen counts were made for 12 different dates, and data were input into a GIS database. The distribution of pollen and its relation to land use were analyzed using land use data obtained from Quickbird imagery acquired during 2007. Comparative analyses of pollen dispersal, wind direction, and speed were made using data collected from a micro-climatic station set up at a papaya plot. The furthest distance from the plot that pollen was found was at 0.9 km, a distance at which only 1 pollen grain was found. The number of pollen grains carried by wind decreased as distance increased. The direction of dispersal was not in accordance with wind direction data. Most pollen grains were found in agricultural areas and bare land. The total number of pollen grains found in exposed areas was considerably higher than the total found in areas sheltered by dense tree lines.  相似文献

Autoantibodies against interferon-gamma (IFN-γ) can cause immunodeficiency and are associated with various opportunistic infections. In the present study, we investigated other cellular immune parameters for a better understanding of the immunodeficiency condition in the patients. The numbers of WBC, monocytes and NK cells were increased in patients with anti-IFN-γ autoantibodies (AAbs). Upon TCR activation, T cell proliferation and IL-2 receptor of the patients remained intact. Nonetheless, the Th1 cytokine (IFN-γ and TNF-α) production was up-regulated. The production of Th2 (IL-4) and Th17 (IL-17) cytokines was unchanged. We suggest that, in addition to the presence of anti-IFN-γ autoantibodies, alterations in the cellular immune functions may also contribute to this immunodeficiency.  相似文献

The genetic variation of seven genes in the carotenoid biosynthetic pathway was investigated in thirteen papaya cultivars. DNA analysis revealed a total of 46 polymorphisms; 42 SNPs (29 positions in introns and 13 positions in exons) and four insertions/deletions. Six SNPs were transversions resulting in amino acid changes. Most variations were found in cpLCYE (21), followed by cpVDE (10). In cpLCYB and cpNCED3, only one polymorphism was found in each gene. Expression studies indicated the differential expression of caroteogenic genes in red, orange and yellow-fleshed papaya during ripening. At ripe stage, the expression level of cpZEP, cpLYCE, cpVDE, cpLCYB and cpNCED3 was higher in red-fleshed than in orange- and yellow-fleshed papaya. The expression level of cpBCH in ripe red-fleshed papaya was significantly lower than that of both orange and yellow-fleshed papaya. Yellow-fleshed papaya cv. Krung Yellow had highest level of beta-carotene at 1428 ± 32 µg/g FW. Lycopene content of red-fleshed papaya cv. Mexico was 1830 ± 208 µg/g FW was highest among the three cultivars. This study revealed DNA polymorphisms in cartenogenic genes among papaya cultivars and their differential expressions in red, orange and yellow-fleshed papaya correlated with carotenoid content and flesh colour.  相似文献

Along with the rapid advances of the nextgen sequencing technologies, more and more species are added to the list of organisms whose whole genomes are sequenced. However, the assembled draft genome of many organisms consists of numerous small contigs, due to the short length of the reads generated by nextgen sequencing platforms. In order to improve the assembly and bring the genome contigs together, more genome resources are needed. In this study, we developed a strategy to generate a valuable genome resource, physical map contig-specific sequences, which are randomly distributed genome sequences in each physical contig. Two-dimensional tagging method was used to create specific tags for 1,824 physical contigs, in which the cost was dramatically reduced. A total of 94,111,841 100-bp reads and 315,277 assembled contigs are identified containing physical map contig-specific tags. The physical map contig-specific sequences along with the currently available BAC end sequences were then used to anchor the catfish draft genome contigs. A total of 156,457 genome contigs (~79% of whole genome sequencing assembly) were anchored and grouped into 1,824 pools, in which 16,680 unique genes were annotated. The physical map contig-specific sequences are valuable resources to link physical map, genetic linkage map and draft whole genome sequences, consequently have the capability to improve the whole genome sequences assembly and scaffolding, and improve the genome-wide comparative analysis as well. The strategy developed in this study could also be adopted in other species whose whole genome assembly is still facing a challenge.  相似文献
