A novel moderately thermophilic bacterium, strain STGHT, was isolated from Severo-Stavropolskoye underground gas storage (Russia). Cells of strain STGHT were spore-forming motile straight rods 0.3 μm in diameter and 2.0–4.0 μm in length having a Gram-positive cell wall structure. The temperature range for growth was 36–65 °C, with an optimum at 50–52 °C. The pH range for growth was 5.5–8.0, with an optimum at pH 7.0–7.5. Growth of strain STGHT was observed at NaCl concentrations ranging from 0 to 4.0 % (w/v) with an optimum at 1.0 % (w/v). Strain STGHT grew anaerobically by reduction of nitrate, thiosulfate, S0 and AQDS using a number of complex proteinaceous compounds, organic acids and carbohydrates as electron donors. Nitrate was reduced to nitrite; thiosulfate and sulfur were reduced to sulfide. It also was able to ferment pyruvate, glucose, fructose, and maltose. The strain STGHT did not grow under aerobic conditions during incubation with atmospheric concentration of oxygen but was able to microaerobic growth (up to 10 % of oxygen in gas phase). The G+C content of DNA of strain STGHT was 34.8 mol%. 16S rRNA gene sequence analysis revealed that the isolated organism belongs to the class Bacilli. We propose to assign strain STGHT to a new species of a novel genus Tepidibacillus fermentans gen. nov., sp.nov. The type strain is STGHT (=DSM 23802T, =VKM B-2671T).  相似文献

The fatty acid composition (FAC) of 43 strains of purple nonsulfur bacteria belonging to six genera--Rubrivivax, Rhodopseudomonas, Rhodoplanes, Blastochloris, Rhodobium, and Rhodomicrobium--was studied by capillary gas chromatography. The cultures were grown on standard medium under standard conditions. Automatic identification of the fatty acid methyl esters and statistical processing of the results were performed by the computerized Microbial Identification System (M.I.S). Significant differences between the FACs of different genera, species, and, sometimes, strains were revealed. 16S rRNA genes of some of the new isolates, primarily those having a specific FAC, were sequenced. The taxonomic status of a number of the strains in question was determined using the FAC characteristics as one of the criteria. It was shown that the FAC characteristics may be used both for affiliating the isolates to known species and for revealing new taxa.  相似文献

A critical step of neuronal terminal differentiation is the permanent withdrawal from the cell cycle that requires the silencing of genes that drive mitosis. Here, we describe that the alpha isoform of the heterochromatin protein 1 (HP1) protein family exerts such silencing on several E2F-targeted genes. Among the different isoforms, HP1alpha levels progressively increase throughout differentiation and take over HP1gamma binding on E2F sites in mature neurons. When overexpressed, only HP1alpha is able to ensure a timed repression of E2F genes. Specific inhibition of HP1alpha expression drives neuronal progenitors either towards death or cell cycle progression, yet preventing the expression of the neuronal marker microtubule-associated protein 2. Furthermore, we provide evidence that this mechanism occurs in cerebellar granule neurons in vivo, during the postnatal development of the cerebellum. Finally, our results suggest that E2F-targeted genes are packaged into higher-order chromatin structures in mature neurons relative to neuroblasts, likely reflecting a transition from a 'repressed' versus 'silenced' status of these genes. Together, these data present new epigenetic regulations orchestrated by HP1 isoforms, critical for permanent cell cycle exit during neuronal differentiation.  相似文献

Anaerobic enrichment with pectin at pH 10 and moderate salinity inoculated with sediments from soda lakes of the Kulunda Steppe (Altai, Russia) resulted in the isolation of a novel member of the Bacteroidetes, strain AP1T. The cells are long, flexible, Gram-negative rods forming pink carotenoids. The isolate is an obligate anaerobe, fermenting various carbohydrates to acetate and succinate. It can hydrolyze and utilize pectin, xylan, starch, laminarin and pullulan as growth substrates. Growth is possible in a pH range from 8 to 10.5, with an optimum at pH 9.5, and at a salinity range from 0.1 to 2 M Na+. Phylogenetic analysis based on 16S rRNA sequences placed the isolate into the phylum Bacteroidetes as a separate lineage within the family Marinilabilaceae. On the basis of distinct phenotype and phylogeny, the soda lake isolate AP1T is proposed to be assigned in a new genus and species Natronoflexus pectinivorans (=DSM24179T = UNIQEM U807T).  相似文献

Decreased levels of early and advanced glycation products were found in exercise-training diabetic rats as compared with sedentary diabetic rats. The components of the nitrogenase complex, MoFe-protein and FeMo-cofactor, possessing no ATPase or nitrogen-fixing activity, maintain the 18O-exchange at the level of 1 atom of 18O per molecule of Pi, which is inhibited by ATP. The Fe-protein complex does not catalyze the 18O-exchange. The nitrogenase components do not hydrolyze the substrates for phosphatase (p-nitrophenylphosphate, beta-glycerophosphate, glucose 1-phosphate and ribose 5-phosphate). The artificial albumin-containing MoFe- and Fe-proteins and the carboxyl group-containing proteins (albumin, hemoglobin, lysozyme) as well as sodium molibdate do not catalyze the 18O-exchange. It is assumed that the site of the ATPase center which is subjected to phosphorylation, is located on the MoFe-protein.  相似文献

We compare predatory behaviour towards a mobile insect in three species of small mammals: the granivorous striped field mouse, the insectivorous common shrew and the Norway rat (a generalist). The striped field mouse displays a surprisingly efficient hunting stereotype. We apply the data compression method (Ryabko et al. Theory Comput Syst 52:133–147, 2013) to compare the complexity of hunting behavioural patterns and to evaluate the flexibility of stereotypes and their succinctness. Norway rats demonstrated the highest level of complexity of hunting behaviour, with the highest proportion of ‘auxiliary’ and ‘noise’ elements and relatively low proportion of ‘key’ elements in their behaviours. The predominance of ‘key’ elements resulted in similarly low levels of complexity of hunting stereotypes in striped field mice and shrews. The similarity between hunting stereotypes of the insectivorous shrew and the granivorous striped field mouse enables us to argue about evolutionary roots of hunting behaviour in small mammals. We show that this method is a useful tool for comparing ethograms as ‘biological texts’.  相似文献

Oscillochloris trichoides is a mesophilic, filamentous, photoautotrophic, nonsulfur, diazotrophic bacterium which is capable of carbon dioxide fixation via the reductive pentose phosphate cycle and possesses no assimilative sulfate reduction. Here, we present the draft genome sequence of Oscillochloris trichoides subsp. DG-6, the type strain of the species, which has permitted the prediction of genes for carbon and nitrogen metabolism and for the light-harvesting apparatus.  相似文献

Species composition of anoxygenic phototrophic bacteria in microbial mats of the Goryachinsk thermal spring was investigated along the temperature gradient. The spring belonging to nitrogenous alkaline hydrotherms is located at the shore of Lake Baikal 188 km north-east from Ulan-Ude. The water is of the sulfate-sodium type, contains trace amounts of sulfide, and salinity does not exceed 0.64 g/L, pH 9.5. The temperature at the outlet of the spring may reach 54°C. The cultures of filamentous anoxygenic phototrophic bacteria, nonsulfur and sulfur purple bacteria, and aerobic anoxygenic phototrophic bacteria were identified using the pufLM molecular marker. The fmoA marker was used for identification of green sulfur bacteria. Filamentous cyanobacteria predominated in the mats, with anoxygenic phototrophs comprising a minor component of the phototrophic communities. Thermophilic bacteria Chloroflexus aurantiacus were detected in the samples from both the thermophilic and mesophilic mats. Cultures of nonsulfur purple bacteria similar to Blastochloris sulfoviridis and Rhodomicrobium vannielii were isolated from the mats developed at high (50.6–49.4°C) and low temperatures (45–20°C). Purple sulfur bacteria Allochromatium sp. and Thiocapsa sp., as well as green sulfur bacteria Chlorobium sp., were revealed in low-temperature mats. Truly thermophilic purple and green sulfur bacteria were not found in the spring. Anoxygenic phototrophic bacteria found in the spring were typical of the sulfur communities, for which the sulfur cycle is mandatory. The presence of aerobic bacteriochlorophyll a-containing bacteria identified as Agrobacterium (Rhizobium) tumifaciens in the mesophilic (20°C) mat is of interest.  相似文献
