Comparisons of nematode communities among ecosystems have indicated that, unlike many organisms, nematode communities have less diversity in the tropics than in temperate ecosystems. There are, however, few studies of tropical nematode diversity on which to base conclusions of global patterns of diversity. This study reports an attempt to estimate nematode diversity in the lowland tropical rainforest of La Selva Biological Research Station in Costa Rica. We suggest one reason that previous estimates of tropical nematode diversity were low is because habitats above the mineral soil are seldom sampled. As much as 62% of the overall genetic diversity, measured by an 18S ribosomal barcode, existed in litter and understorey habitats and not in soil. A maximum-likelihood tree of barcodes from 360 individual nematodes indicated most major terrestrial nematode lineages were represented in the samples. Estimated 'species' richness ranged from 464 to 502 within the four 40 × 40 m plots. Directed sampling of insects and their associated nematodes produced a second set of barcodes that were not recovered by habitat sampling, yet may constitute a major class of tropical nematode diversity. While the generation of novel nematode barcodes proved relatively easy, their identity remains obscure due to deficiencies in existing taxonomic databases. Specimens of Criconematina, a monophyletic group of soil-dwelling plant-parasitic nematodes were examined in detail to assess the steps necessary for associating barcodes with nominal species. Our results highlight the difficulties associated with studying poorly understood organisms in an understudied ecosystem using a destructive (i.e. barcode) sampling method. 相似文献

AFTER electrophoresis of human serum in starch gel1, there are at least four bands of Cholinesterase activity. Most of the enzyme is concentrated in the band with least mobility (C4) while three faster moving bands (C1, C2, C3) are of lower molecular weight as shown by gel filtration2. Although electrophoresis in Polyacrylamide reveals additional bands3, all seem to be a single genetic entity as shown by experiments on persons homozygous for the silent gene4,5. The C4 enzyme has a molecular weight of at least 250,000 (ref. 6) and can be assumed to be composed of more than one polypeptide chain. By preparing an enzyme of altered mobility and forming hybrids of it with the usual enzyme, we have obtained results that suggest that Cholinesterase is a tetramer. 相似文献

The British Standards Institution and the International Standards Organization specify that when mixtures of tea and milk are to be evaluated sensorially, the tea should be added to the milk. Four cups of tea prepared by pouring the tea into the cup first (TIF) and four cups prepared by placing the milk in the cup first (MIF) were presented to 155 panelists in the dark. They were instructed to select by taste the four cups containing the MIF tea. The frequency of correct selection was well above the chance level, indicating that the panelists had detected a difference in flavor. The same task was performed by 131 panelists except they evaluated the tea in light. The panelists were not highly trained tea tasters; in fact, most were not regular drinkers of hot tea. Notwithstanding that, their discriminating power was sufficient to resolve the subtle differences which did exist. 相似文献

Global temperatures are rising, and are expected to produce a poleward shift in the distribution of many organisms. We quantified changes in fish assemblages within seagrass meadows of the northern Gulf of Mexico (GOM) between the 1970s and 2006–2007, and observed changes consistent with this forecast. During 2006–2007 we sampled seagrass meadows using the same gears and methods previously employed by R. J. Livingston in coastal waters of northwest Florida throughout the 1970s. Comparisons between datasets revealed numerous additions to the fish fauna during 2006–2007 that were completely absent in the 1970s, including: Lutjanus synagris (lane snapper), Epinephelus morio (red grouper), Chaetodon ocellatus (spotfin butterflyfish), Mycteroperca sp (grouper, non gag), Centropristis philadelphica (rock sea bass), Fistularia tabacaria (bluespotted cornetfish), Ocyurus chrysurus (yellowtail snapper), Thalassoma bifasciatum (bluehead wrasse), Abudefduf saxatilis (sergeant major), Acanthuridae spp. (surgeonfishes) and Sparisoma viride (stoplight parrotfish). Several other species showed large increases in abundance during the interval between 1979 and 2006, including Mycteroperca microlepis (gag grouper, up ∼200 ×), Lutjanus griseus (gray snapper, up ∼105 ×), and Nicholsina usta (emerald parrotfish, up ∼22 ×). All of these are tropical or subtropical species that now make up a greater percentage of seagrass-associated fish assemblages in the northern GOM than in the past. Additionally, we observed regional increases in air and sea surface temperatures (> 3 °C) during the ∼30 years that separate Livingston's samples and ours that correlate with northern shifts in the distribution of warm-water fishes. Documenting these range shifts is a critical first step in investigating the consequences of global warming for endemic marine communities and fishery production in the northern GOM. 相似文献
