Although microsatellite markers have become exceedingly popular in molecular studies of wild organisms, their development in some taxonomic groups is challenging. This is partly because of repetitive flanking sequences, which lead to the simultaneous amplification of alleles from multiple loci. Until now, these microsatellite DNA families have been considered unsuitable for population genetics studies, but here we describe our development of these repetitive flanking sequences (ReFS) as novel molecular markers. We illustrate the utility of these markers by using them to address an outstanding taxonomic question in the moth genus Schrankia.  相似文献

A chamber for the simulation of radiation freezing of plants   总被引:1,自引:0,他引:1  
Frost injury to plants can occur following episodic radiation frosts. In the UK this is particularly important to spring sown crops such as potatoes. Most laboratory based frost studies simulate freezing using either conductive or convective freezing chambers. Such frost tests do not simulate overnight freezing events adequately. A freezing chamber based on radiative cooling is described which mimics overnight radiative freezing. The chamber is rectangular in design (1 m × lm × 2 m high) with a radiative cooling plate at the top of the chamber cooled to -40°C to -45°C using HFC coolants, which acts as a cold black body. The sides of the chamber are also cooled to variable temperatures down to -5°C in order to prevent the chamber walls radiating to the plant material during testing. Using thermocouples to measure air temperature and plant temperature the chamber has been characterised to simulate the radiative cooling conditions found in the UK during autumn and spring. Exotherm detection upon plant freezing is simplified by virtue of the reduction in temperature fluctuation normally experienced at the plant surface during natural freezing. Radiation frosts and subsequent frost damage to potatoes have been recorded in the temperature range -4°C to –5°C. The equipment is recommended for studies of frost damage to plants normally caused by episodic radiation frost events.  相似文献

Abstract:  Ornitholestes hermanni is a Late Jurassic theropod dinosaur from North America. This kinematic study of Ornitholestes uses manual manipulations of forelimb casts to determine range of motion. The manual phalanges of the O. hermanni holotype, previously unidentified, are here identified as phalanges I-1, I-2 (ungual), II-2 and II-3 (ungual). At all represented manual joints, hyperextensibility is small or absent, whereas flexion is strong, as in most other theropods. The elbow can be strongly flexed beyond a right angle. When data on range of forelimb motion in Ornitholestes are added to such data from other theropods, high elbow flexion is present in maniraptoriform coelurosaurs but not in basal theropods. Forelimb functions requiring strong elbow flexion (such as holding objects to the chest, or tucking the forearms in for their protection or to reduce wind resistance or heat loss) were therefore available to maniraptoriform coelurosaurs but not to basal theropods.  相似文献

Papua New Guinea (PNG) is an extensively forested country. Recent research suggests that despite commencing a trajectory of deforestation and degradation later than many counties in the Asia–Pacific region, PNG is now undergoing comparable rates of forest change. Here we explore the bioregional distribution of changes in the forest estate over the period 1972–2002 and examine their implications for forest protection. This is undertaken through the development of a novel bioregional classification of the country based on biogeographic regions and climatic zones, and its application to existing forest cover and forest‐cover change data. We found that degradation and deforestation varied considerably across the 11 defined biogeographic regions. We report that the majority of deforestation and degradation has occurred within all the lowland forests, and that it is these forests that have the greatest potential for further losses in the near term. The largest percentage of total change occurred in the east of PNG, in the islands and lowlands of the Bismarck, D'Entrecasteaux, East Papuan Islands and in the South‐East Papua–Oro region. The only region with a significant highlands component to undergo deforestation at a comparable magnitude to the islands and lowland regions was the Huon Peninsula and Adelbert region. Significant changes have also occurred at higher elevations, especially at the interface of subalpine grasslands and upper montane forests. Lower montane forests have experienced proportionally less change, yet it is these forests that constitute the majority of forests enclosed within the protected area system. We find that protected areas are not convincingly protecting either representative areas of PNG's ecosystems, nor the forests within their borders. We conclude by suggesting a more expansive and integrated approach to managing the national forest estate.  相似文献

Xing, L, Bell, P.R., Currie, P.J., Shibata, M., Tseng, K. & Dong, Z. 2012: A sauropod rib with an embedded theropod tooth: direct evidence for feeding behaviour in the Jehol group, China. Lethaia, Vol. 45, pp. 500–506. A fragmentary rib from the Lower Cretaceous (Barremian) Yixian Formation in northeastern China preserves rare, direct evidence of feeding behaviour by an unidentified theropod. The rib, which comes from the holotype of Dongbetitan, preserves an embedded, broken theropod tooth. Comparison of the tooth with all known theropods from the Yixian Formation suggests that it belongs to a new taxon of medium‐sized theropod. Given the large size difference between the sauropod and the theropod and the absence of reactive bone growth around the tooth, the bite likely occurred post‐mortem during scavenging. Recognition of a new, medium‐sized theropod increases the known diversity of taxa from the Yixian Formation and helps fill a gap in the theropod palaeoecology of that formation, which previously consisted of only small (<2 m) forms. □China, Cretaceous, feeding behaviour, theropod, titanosauriformes, sauropod.  相似文献

The genus Prosopis contains 44 species of trees and shrubs, the majority of which originate in the Americas. Most species are reported to be diploid, with a somatic chromosome number of 2 n  = 28. There are rare reports of polyploidy, although it is thought that these may represent polysomaty in root tissues. However, flow cytometry has recently indicated that P. juliflora is entirely tetraploid with a somatic chromosome number of 2 n  = 56. In order to clarify the situation, an extensive review of ploidy in Prosopis was undertaken, the first of its kind. The ploidy levels of 124 samples of Prosopis from 21 countries, including both the natural and introduced ranges, were analysed using flow cytometry. In addition, a comprehensive literature review was carried out, examining 305 published ploidy values and covering 32 of the 44 species of Prosopis . Flow cytometry analysis suggests that P. juliflora is the only tetraploid species, with a somatic chromosome number of 2 n  = 4 x  = 56, whilst the remainder of the species analysed are diploid with 2 n  = 2 x  = 28, including the first report for P. articulata (2 n  = 28). A critical review of published ploidy values shows that all species of Prosopis are reported to be entirely diploid, except P. glandulosa , P. juliflora , and P. koelziana , for which both diploid and tetraploid values have been recorded. © 2008 The Linnean Society of London, Botanical Journal of the Linnean Society , 2008, 156 , 425–438.  相似文献

Abstract Monitoring the biodiversity of Australian rangelands has been identified as a means of informing policy and supporting funding decisions in relation to the conservation of biodiversity. Australian rangelands are subject to invasion by alien plants that have the potential to have major impacts on ecosystem function and biodiversity, although there has been little quantitative documentation of these effects. Research is needed to improve our understanding of how and to what extent alien plants affect biodiversity in Australian rangelands so that this relationship can be considered when developing and implementing programmes to monitor biodiversity. It is also important to consolidate existing efforts to quantify the extent of alien plant invasions and monitor their progress, thus documenting a process that threatens biodiversity. Information on the presence and abundance of alien plant species should be considered for inclusion as a component of biodiversity monitoring programmes that are undertaken. Monitoring components of biodiversity can itself provide a basis for evaluating weed management strategies.  相似文献

Abstract To prioritize weed management activities it is necessary to predict the extent of future infestations of individual weed species. Management effort should focus on populations which are likely to spread and produce new satellite populations. Deciding which aspects of an invading population to manage, new emerging distant satellites or the existing core from which most seed originates has been the subject of much debate. We use extensive survey data to define the core and satellite populations and dispersal function of Parkinsonia aculeata in the semi‐arid rangelands of the Northern Territory of Australia. We described populations in terms of three size classes, classified on the basis of plant height (small (<0.4 m), medium (>0.4 and <2.5 m) and large (>2.5 m)). Data were recorded as a spatial point pattern and a dispersal function was derived from the distribution of the distance of each medium plant to the nearest P. aculeata neighbour that was greater than 2.5 m. All large and medium P. aculeata were classified as members of core or satellite populations. From these classifications, we tested critical assumptions of Moody and Mack's model, namely (i) the satellites form multiple disjunct foci that do not overlap; (ii) the system is homogenous and there are no abiotic or biotic restrictions to the weeds expansion enabling it to expand radially in any direction; (iii) the densities of all foci are uniform; (iv) there is no extinction through stochastic processes; and (v) there are no threshold effects. When the medium size class was considered, the population density and area occupied by the weed increased. Two satellite populations were comprised solely of medium plants, suggesting the number of satellites increased marginally from eight to 10 when data from both size classes were considered. Most individuals were part of the core population. The core population increased in density and we argue this core population is most likely to give rise to new satellites and should be managed in preference to emerging satellite populations. We suggest the invasion is behaving like a source‐sink system, where the core population is the source of most new medium plants.  相似文献

How soil carbon balance will be affected by plant–mycorrhizal interactions under future climate scenarios remains a significant unknown in our ability to forecast ecosystem carbon storage and fluxes. We examined the effects of soil temperature (14, 20, 26 °C) on the structure and extent of a multispecies community of arbuscular mycorrhizal (AM) fungi associated with Plantago lanceolata. To isolate fungi from roots, we used a mesh‐divided pot system with separate hyphal compartments near and away from the plant. A 13C pulse label was then used to trace the flow of recently fixed photosynthate from plants into belowground pools and respiration. Temperature significantly altered the structure and allocation of the AM hyphal network, with a switch from more vesicles (storage) in cooled soils to more extensive extraradical hyphal networks (growth) in warmed soils. As soil temperature increased, we also observed an increase in the speed at which plant photosynthate was transferred to and respired by roots and AM fungi coupled with an increase in the amount of carbon respired per unit hyphal length. These differences were largely independent of plant size and rates of photosynthesis. In a warmer world, we would therefore expect more carbon losses to the atmosphere from AM fungal respiration, which are unlikely to be balanced by increased growth of AM fungal hyphae.  相似文献
