We characterized nine microsatellite loci and identified an additional 60 genomic regions containing microsatellites in the red hind grouper, Epinephelus guttatus. The nine loci were highly polymorphic, and primers designed from red hind genomic DNA produced a strong amplification product in a test panel of 16 other groupers in the genera Epinephelus and Mycteroperca collected from across the world. Most of the amplified regions were homologous to the red hind locus and a well‐defined microsatellite repeat was generally evident. The nine loci, together with the 60 uncharacterized microsatellite‐containing regions, provide a powerful tool for ecological and evolutionary studies in groupers.  相似文献

The nymphalid butterfly tribe Preponini includes some of the Neotropical region's most spectacular and familiar butterflies, but the taxonomy of the group nevertheless remains unstable. Several recent studies of Nymphalidae phylogeny have suggested that both the tribe itself and several genera might not be monophyletic, but to date taxon sampling has not been sufficiently comprehensive to allow informed revision of the group's systematics. We therefore conducted the first complete species‐level phylogenetic study of the tribe to establish a firm higher classification. We used DNA sequence data from three genes, the two mitochondrial genes cytochrome oxidase subunits I and II (COI and COII), and the nuclear gene elongation factor‐1α (EF‐1α), to reconstruct the phylogeny of the tribe using maximum likelihood (ML), maximum parsimony (MP) and Bayesian inference (BI). We included 48 individuals representing the 22 recognised Preponini species, and an additional 25 out‐group taxa to explore taxonomic limits at different levels. Firstly, we found that Anaeomorpha splendida Rothschild never grouped with remaining Preponini, so that maintaining monophyly of the tribe requires the taxon to be excluded, and we thus reinstate the tribe Anaeomorphini stat.rev. Secondly, we investigated generic limits, in particular the relationship of Noreppa Rydon to Archaeoprepona Fruhstorfer, and that of Agrias Doubleday to Prepona Boisduval. The molecular results coupled with previous morphological studies suggest that Noreppa syn.n should be synonymised with Archaeoprepona, and that Agrias syn.n should be synonymised with Prepona. We found Prepona pheridamas (Cramer) to be sister to all other Prepona, and markedly divergent from them in both morphology and DNA sequences, suggesting the possibility that it should be placed in a separate genus. We also found a number of cases of significant DNA sequence divergence and paraphyly or polyphyly within putative species that require further taxonomic attention, including Prepona claudina (Godart) stat.n. and Prepona narcissus (Staudinger) stat.n., Prepona pylene Hewitson and Prepona deiphile (Godart). Future research should focus on a broader population sampling of widespread, polymorphic Preponini species to thoroughly revise the current species‐level taxonomy, thus creating a solid foundation for studies in ecology and conservation.  相似文献

  • 1 Tuberculosis (TB) is a chronic disease caused by Mycobacterium bovis and related members of the Mycobacterium tuberculosis complex. Infection affects not only cattle but also other livestock species, companion animals and wild mammals. Humans are also susceptible; hence, zoonotic infection is a driver for disease control in animal hosts. As bovine TB prevalence has been reduced in livestock, the relative epidemiological and socio‐economic importance of wildlife reservoirs has increased, and there is a need for disease management strategies.
  • 2 We review the current status of TB in European wild mammals, identifying epidemiological trends and areas for future research and management. TB has a complex epidemiology, which may involve multiple hosts, and is influenced by climate and habitat.
  • 3 Consequently, the role of wild and domestic hosts in the epidemiology of TB varies among regions. In Europe, there are three regional examples of M. bovis maintenance hosts: the Eurasian badger Meles meles in Great Britain and Ireland, the Eurasian wild boar Sus scrofa in the Iberian Peninsula and deer belonging to the subfamily Cervinae in several European regions. In other parts of Europe, these species are currently regarded as spillover hosts, although in time their status may change depending on local or regional risk factors. Nevertheless, in most situations, the relative contribution of wild mammals to M. bovis infection in cattle is still a matter of debate. Also, the outcome of management interventions to control disease in wildlife populations may be complex and counter‐intuitive.
  • 4 As our knowledge of disease dynamics in wild mammals improves, options for disease control in wildlife reservoirs, such as vaccination, improved biosecurity and population management, are likely to broaden. In order to evaluate our existing control options, we must monitor the effects of interventions on TB occurrence in the affected regions of Europe and share our collective experiences.

  • 1 In studies of birds and their pathogens, spleen size has frequently been used to make inferences about immune system strength. However, the use of spleen size in mammals is more complicated because, in addition to having an immune function, the mammalian spleen is also a reservoir for red blood cells.
  • 2 To assess the reliability of mammalian spleen mass as an indicator of immune activity, we quantified the white and red pulp mass by histology of spleens from shot red deer Cervus elaphus. We then analysed the relationships among spleen mass, the amounts of white and red pulp, and the deer's body condition relative to faecal counts of the nematode parasite Elaphostrongylus cervi.
  • 3 White and red pulp mass were positively correlated so that an increase in spleen mass was a positive function of both components of the spleen. In male deer, which had significantly lower body condition and higher parasite loads than females, parasite counts were negatively correlated with spleen mass, white pulp mass, and red pulp mass.
  • 4 Our findings suggest that (i) spleen mass in shot red deer is a reliable measure of white and red pulp content; and (ii) when looking at the red deer life history, which is greatly influenced by sex of the deer, splenic mass and white pulp mass could be used as reflections of immune system strength.
  • 5 Future studies of mammalian spleens can contribute to the understanding of evolved strategies of immune response investment in mammals. However, determination of the white and red pulp spleen components using various sampling methods must be made prior to their application.

Leaf water (Ψ) and solute (ψ) potential were measured in field sorghum and maize under well irrigated (I) and dryland (NI) conditions throughout a season. Despite decreases in ψ due to slow soil water depletion and to apparent increases in liquid phase plant resistance, midday leaf turgor (ψp) in the NI sorghum was maintained at similar levels as in the I treatment throughout the season due to concomitant decreases in ψs. Osmotic adjustment was also observed in maize, although ψp was significantly lower in the NI treatment as compared to I during the final stages of grain filling. A seasonal shift in the ψ vs. relative water content relation of NI sorghum leaves was observed, more water being retained by the older leaf at any particular ψ. The major factor for turgor maintenance was a net increase in solutes per unit of tissue. The role played by increases in the proportion of tissue volume occupied by cell wall was also evaluated. No stomatal closure due to water stress was found in NI sorghum even though leaf ψ reached —20 bars late in the season. Under similar conditions, stomata closed at —14 to —16 bars in younger plants where water stress was made to develop much faster.  相似文献

There is limited information on the impacts of present-day solar ultraviolet-B radiation (UV-B) on biomass and grain yield of field crops and on the mechanisms that confer tolerance to UV-B radiation under field conditions. We investigated the effects of solar UV-B on aspects of the biochemistry, growth and yield of barley crops using replicated field plots and two barley strains, a catalase (CAT)-deficient mutant (RPr 79/4) and its wild-type mother line (Maris Mink). Solar UV-B reduced biomass accumulation and grain yield in both strains. The effects on crop biomass accumulation tended to be more severe in RPr 79/4 (≈ 32% reduction) than in the mother line (≈ 20% reduction). Solar UV-B caused measurable DNA damage in leaf tissue, in spite of inducing a significant increase in UV-absorbing sunscreens in the two lines. Maris Mink responded to solar UV-B with increased CAT and ascorbate peroxidase (APx) activity. No effects of UV-B on total superoxide dismutase (SOD) activity were detected. Compared with the wild type, RPr 79/4 had lower CAT activity, as expected, but higher APx activity. Neither of these activities increased in response to UV-B in RPr 79/4. These results suggest that growth inhibition by solar UV-B involves DNA damage and oxidative stress, and that constitutive and UV-B-induced antioxidant capacity may play an important role in UV-B tolerance.  相似文献
