This study was designed to assess the changes in fiber-type distribution of the extensor digitorum longus (EDL) muscle of the mouse during the first 21 days of age following neonatal sciatic neurectomy. Denervated and normal muscles were compared at 7, 14, and 21 days of age and the normal EDL was also studied at 1 day of age. Frozen sections of the EDL were treated histochemically to detect NADH-tetrazolium reductase and myosin ATPase reactions. Quantitative assessment included measurements of cross-sectional areas and fiber counting. Denervation resulted in muscle atrophy which was due primarily to a decrease in individual fiber area as opposed to fiber loss. Histochemical maturation of the EDL was severely affected by neonatal denervation during the first three postnatal weeks. By 21 days, two extrafusal fiber types which were both oxidative could be distinguished. One type was highly atrophied and resembled an immature fiber exhibiting myosin ATPase staining at both acid and alkaline preincubation conditions, whereas another type was less atrophied and showed myosin ATPase staining resembling fast-twitch (type IIA) fibers. These findings emphasize the importance of an intact nerve supply in determining the phenotypic expression of skeletal muscle, and point to the early postnatal period as a critical stage in fiber type differentiation. 相似文献

The in vitro deproteinized vastus lateralis muscle buffer capacity, carnosine, and histidine levels were examined in 20 men from 4 distinct populations (5 sprinters, 800-m runners; 5 rowers; 5 marathoners; 5 untrained). Needle biopsies were obtained at rest from the vastus lateralis muscle. The buffer capacity was determined in deproteinized homogenates by repeatedly titrating supernatant extracts over the pH range of 7.0-6.0 with 0.01 N HCl. Carnosine and histidine levels were determined on an amino acid AutoAnalyzer. Fast-twitch fiber percentage was determined by staining intensity of myosin adenosinetriphosphatase. High-intensity running performance was assessed on an inclined treadmill run to fatigue (20% incline; 3.5 m X s-1). Significantly (P less than 0.01) elevated buffer capacities, carnosine levels, and high-intensity running performances were demonstrated by the sprinters and rowers, but no significant differences existed between these variables for the marathoners vs. untrained subjects. Low but significant (P less than 0.05) interrelationships were demonstrated between buffer capacity, carnosine levels, and fast-twitch fiber composition. These findings indicate that the sprinters and rowers possess elevated buffering capabilities and carnosine levels compared with marathon runners and untrained subjects. 相似文献

An ultrastructural comparison of the two types of intrafusal muscle fibers in muscle spindles of the rat was undertaken. Discrete myofibrils with abundant interfibrillar sarcoplasm and organelles characterize the nuclear chain muscle fiber, while a continuous myofibril-like bundle with sparse interfibrillar sarcoplasm distinguishes the nuclear bag muscle fiber. Nuclear chain fibers possess well-defined and typical M bands in the center of each sarcomere, while nuclear bag fibers contain ill-defined M bands composed of two parallel thin densities in the center of the pseudo-H zone of each sarcomere. Mitochondria of nuclear chain fibers are larger and more numerous than they are in nuclear bag fibers. Mitochondria of chain fibers, in addition, often contain conspicuous dense granules, and they are frequently intimately related to elements of the sarcoplasmic reticulum (SR). Striking differences are noted in the organization and degree of development of the sarcotubular system. Nuclear bag fibers contain a poorly developed SR and T system with only occasional junctional couplings (dyads and triads). Nuclear chain fibers, in contrast, possess an unusually well-developed SR and T system and a variety of multiple junctional couplings (dyads, triads, quatrads, pentads, septads). Greatly dilated SR cisternae are common features of nuclear chain fibers, often forming intimate associations with T tubules, mitochondria, and the sarcolemma. Such dilatations of the SR were not encountered in nuclear bag fibers. The functional significance of these structural findings is discussed. 相似文献

Reconstructions of the human-African great ape phylogeny by using
mitochondrial DNA (mtDNA) have been subject to considerable debate. One
confounding factor may be the lack of data on intraspecific variation. To
test this hypothesis, we examined the effect of intraspecific mtDNA
diversity on the phylogenetic reconstruction of another Plio- Pleistocene
radiation of higher primates, the fascicularis group of macaque (Macaca)
monkey species. Fifteen endonucleases were used to identify 10 haplotypes
of 40-47 restriction sites in M. mulatta, which were compared with similar
data for the other members of this species group. Interpopulational,
intraspecific mtDNA diversity was large (0.5%- 4.5%), and estimates of
divergence time and branching order incorporating this variation were
substantially different from those based on single representatives of each
species. We conclude that intraspecific mtDNA diversity is substantial in
at least some primate species. Consequently, without prior information on
the extent of genetic diversity within a particular species, intraspecific
variation must be assessed and accounted for when reconstructing primate
phylogenies. Further, we question the reliability of hominoid mtDNA
phylogenies, based as they are on one or a few representatives of each
species, in an already depauperate superfamily of primates.

Toxoplasmosis is a parasitic zoonosis. The importance of the cat as related to this parasitosis lies in the fact that it is not only the definite host of the parasite, but responsible for its dissemination through the release of oocysts, which subsequently infect both humans and other animals. The objective of this study was to determine the serological prevalence of Toxoplasma gondii in the feline population of the city of Valdivia (Chile) and to establish the possible epidemiological implications of this prevalence. With these goals in mind the technique of indirect immunofluorescence was implemented to detect anti- T. gondii species--specific IgG. Blood samples from 97 cats (selected using a directed sampling process) from different sectors of Valdivia were collected. The sample included 46 males and 51 females of different ages, Positive and negative control sera obtained from de United States were used to verify the results, observed by UV microscopy. The anti-feline IgG antibody was used as directed by the manufacturer (Sigma). Of the 97 selected cats, 32 were found to be positive with a titre higher than 1:4, a prevalence of 33.0%. The number of infected males as compared to the number of infected females was found to be statistically insignificant, using the chi-square analysis with p less than 0.1. In contrast, a definite correlation between age and seropositivity was found; infection levels were higher in older animals. These results are consistent with those obtained in different studies on this topic that have been performed in different parts of the world. However, they would seem to be in conflict with other, similar studies that have been done recently in Chile; this may be due to the fact that the cats selected for this study were exposed to different climatic conditions than those examined in previous works. In conclusion, it has been determined that in Valdivia exist cats infected by T. gondii, which indicate the presence of the necessary epidemiological conditions for the persistence of this parasitic cycle and the source of infection for humans and other animals. 相似文献

This study describes the spatial and temporal dynamics of several physical, chemical and biological variables in the Grussai
lagoon, and their relationship to ephemeral sand bar openings and to a constant in natura waste water input. The spatial variation
in pH, dissolved oxygen, electrical conductivity, total alkalinity and nutrients (e.g. soluble reactive silicate, soluble
reactive phosphate and ammonium) was associated to the anoxic and nutrient rich groundwater discharge, the development of
aquatic macrophytes, the biological activities of phytoplanktonic community and the marine influence. During the period when
the sand bar was closed (isolated), the lagoon water was supersaturated with dissolved oxygen and exhibited high values of
pH (8–10), total alkalinity (3.000–5.000 μeq l-1), and chlorophyll a contents (60-300 μg l-1), and had low values of dissolved nutrients (nearly undetectable). These suggest a biological processes dominance. When the
sand bar was opened, there was an enrichment with dissolved inorganic nutrients (e.g. ammonium and soluble reactive phosphorus
up to 120 and 5 μM, respectively) and a decrease in pH (below 8), total alkalinity (below 3.000 μeq l-1) and dissolved oxygen during the initial second to eight days. Subsequently there was a period when the physical and chemical
characteristics of seawater prevailed. The lagoon returned chemical to the pre-opening water conditions in a few days (∼ 10–20).
This quick return implies highly efficient biological mechanisms. The high levels of chlorophyll a, total nitrogen and phosphorus
in the water column indicate a high eutrophication stage in the Grussai lagoon during the sand bar closed periods.

Burr Medic (Medicago polymorpha) is awidespread, polymorphic annual legume of Mediterranean origin, and as aself-reseeding, highly effective nitrogen fixer it has important value invarious dryland farming systems. In central Chile, it is naturalized along a1000 km long environmental gradient, from an arid zone(29°–31° S) to a perhumid one(37°–38°24' S). Comparative and experimental studiesof its ecotypic differentiation patterns along this gradient should be usefulinelucidating the evolutionary forces at work along environmental gradients, andalso for efforts to select annual legumes for pastures and fallow fieldimprovement in mediterranean climate areas. In this first paper of atwo-part series, we present results of two common-environmentexperiments conducted at a subhumid site in central Chile to compare vegetativeand reproductive traits in 69 populations of Burr Medic collected in 1988 and1994. Multivariate analyses based on 11 phenological and agronomic characters,including phenology and biomass accumulation, winter vigour and degree ofhard-seededness, showed that three principal components (PCs)explained nearly 70% of total variability, in both collections. For thelarger collection (1988), the diagram of dispersion for the first twoPCs – phenology, winter vigour and above-ground biomass –provided good discrimination between accessions from arid-semiarid andhumid-perhumid mediterranean zones. Onset of flowering was positivelycorrelated with both latitude and longitude, as well as mean annualprecipitation/potential evapotranspiration (PP/ETP) ofcollecting site; days from first flower to pod ripening was negativelycorrelated with PP/ETP. In contrast to reports from Syria and Sardinia, noclear trends in Burr Medic seed or pod-related traits was found along theChilean gradient, apart from the concentration of spiny pod accessions in thesouthern, more mesic end of the gradient. Winter vigour (according to asemi-quantitative analysis of growth) was greater in accessions fromthe warmer, arid and semiarid zones than in those from cooler, subhumid tohumidones. Both winter vigour and harvest index were negatively correlated with daysto first flower. Applied and evolutionary aspects of the results are brieflydiscussed. 相似文献

The ultrastructure of inner capsule cells of the vertebrate muscle spindle was studied by transmission electron microscopy and compared with that of homologous cells in the tendon organ. Aside from variations in their complexity and pattern of organization, cells of the inner capsule in these two sensory receptors exhibited marked similarities in fine structure. The virtual absence of basal lamina in the region of the nucleated soma as well as on the branching cytoplasmic extensions of these cells was noted. In the inner capsule of both end organs, three kinds of intercellular specialization were encountered. Cell processes were typically linked together at multiple sites by intermediate junctions. In addition, focal points of membrane fusion between two or more cellular profiles were identified as tight junctions. In more extensive regions of plasma membrane overlap, gap junctions were also discerned. It seems probable that these sites along the inner capsule represent areas of mechanical and electrical linkage, enabling contiguous cells to function as a synchronous unit. Tight junctions may also provide the inner capsular sheath with specific permeability-barrier characteristics. Elements of the Golgi complex and associated presecretory vesicles and cytoplasmic granules were prominent. Their presence implicates these cells in the elaboration of the paracellular connective-tissue matrix occupying the intracapsular spaces of both receptors. The close resemblance of these cells to endoneurial fibroblasts of peripheral nerve and to hyalocytes of the vitreous body is emphasized. It is likely that, regardless of species examined, cells of the inner capsule in both receptors play an overall protective role in the formation, maintenance and regulation of their luminal paracellular contents. 相似文献

The phenological development of nine Chilean accessions of Medicagopolymorpha, collected along a northsouth aridity gradient,and of two commercial cultivars of the same species, were comparedin 12 sequential outdoor sowings at Cauquenes (35°58'S,72°17'W, elev. 177 m), in the sub-humid Mediterranean climatezone of Chile. A glasshouse experiment was also conducted toevaluate the effect of photoperiod on phenophase timing. Therewas a clear gradient in precocity among the Chilean accessionsin both experiments: accessions MPO-9-88 and MPO-7-88, fromthe arid zone, were the earliest-flowering accessions, whereasMPO-36-88 from the humid Mediterranean zone was the latest.Both experiments revealed significant variation among the Chileanaccessions in the response of flowering time to variation inphotoperiod regime. Differences in days to flowering betweenthe least- (8 h) and the most- (16 h) inductive photoperiodswere lower in precocious accessions from arid and semi-aridzones, than in late-flowering accessions from more humid zones.Rate of progress to flowering, defined as the inverse of timefrom emergence to first flower appearance (1/ f), was relatedto mean diurnal temperature, or to both mean diurnal temperatureand mean photoperiod. In two early-flowering accessions fromthe arid zone, and in the Australian cultivar CircleValley, 1/ f was affected significantly (P < 0.05)by both temperature and photoperiod.
