Proliferative response of splenic lymphocytes from adult (2 months old) male mice to human luteotrophic hormone (hLH) was studied in vitro. hLH induced lymphoblastic transformation at a concentration of 0.1 micrograms/ml or higher. This hormone potentiated the mitogen activities of concanavalin A (ConA) and phytohemagglutinin (PHA) for T cells and of pokweed mitogen for B and T cells. The data suggest that peripherally circulating LH may regulate a certain function of lymphoid cells.  相似文献

Metastasis mechanisms depend on cell metabolism changes, migration and adhesion to different tissues. To understand their choice of interaction site, the tumoral cell adhesion to model surfaces was studied. The response of Caco-2 tumoral cells cultured on polyelectrolyte film-functionalized surfaces with or without adhesion proteins (fibronectin or collagen IV) was analyzed. Using the layer-by- layer method, multilayer films were prepared with cationic poly(allylamine hydrochloride) and anionic poly(sodium 4-styrenesulfonate) polyelectrolytes. Film surface wettability was evaluated. The electrochemical impedance spectroscopy analyses were carried out to control the elaborated surfaces on which Caco-2 tumoral cells were cultured. The cell velocity was studied by video-microscopy and a cell colorimetric assay (WST-1) was used to quantify cell viability. The film surface parameters as well as the protein nature and localization in the film were found to modulate cell response. Results demonstrated that the cancer cell motility and proliferation were higher when cultured onto pure collagen located above the polyelectrolyte film and that the reverse surprisingly was observed when proteins were inserted into the polyelectrolyte film. Data also showed that cell motility was correlated with a high charge transfer resistance (Rct) and a low surface free energy (SFE) polar component (electron donor character). This relationship was valid only for pure external proteins. Thus, fibronectin exhibited a low Rct and a high SFE polar component, which decreased cell motility and proliferation.  相似文献

Fanconi anemia (FA) is a recessive chromosomal instability syndrome that is clinically characterized by multiple symptoms. Chromosome breakage hypersensitivity to alkylating agents is the gold standard test for FA diagnosis. In this study, we provide a detailed laboratory protocol for accurate assessment of FA diagnosis based on mitomycin C (MMC) test. Induced chromosomal breakage study was successful in 171 out of 205 aplastic anemia (AA) patients. According to the sensitivity of MMC at 50 ng/ml, 38 patients (22.22%) were diagnosed as affected and 132 patients (77.17%) as unaffected. Somatic mosaicism was suspected in an 11-year-old patient with a FA phenotype. Twenty-six siblings of FA patients were also evaluated and five of them (19.23%) were diagnosed as FA. From this study, a standard protocol for diagnosis of FA was developed. It is routinely used as a diagnostic test of FA in Tunisia.  相似文献

In the present study the atomic force microscope (AFM) was used to image the surface morphology of red blood cells (RBC) for the first time. The AFM yielded very reproducible images without appreciable modifications of the sample surfaces. In addition to this topographical imaging, we have developed an experimental approach to measure the binding strength between antibody (anti-A), and the RBC antigen A, when reversible bonds between specific molecules such as antigen and antibody mediate the adhesion. The experimental results suggest that the procedure established here may be used for specific antibody detection. This study has also enhanced our understanding under physiological conditions of molecular interaction in particular antigen-antibody.  相似文献

This study aimed to evaluate the in vitro antifungal activity (AA) of the essential oil (EO) of lemon (Citrus limon L.) against three pathogenic fungi attacking grapevine wood. The composition of the EO was also studied. Ten volatile components were identified by gas chromatography–mass spectrometry. The results showed that the EO consists of volatile components where monoterpene hydrocarbons are the most abundant ones. Four major components were identified, which represent 99.9% of the total EO (limonene, neral, ß-pinene, and ?-terpinene). The AA of the EO was evaluated against three pathogenic fungi attacking grapevine wood (Eutypa sp., Botryosphaeria dothidea, and Fomitiporia mediterranea). The results showed that the EO exerts AA against all tested fungi and significantly inhibits their growth. Eutypa sp. is the most sensitive fungus. These results show, for the first time, a new use for the EO of lemon (C. limon L.) to control fungal diseases of grapevine wood.  相似文献

The electrochemical impedance spectroscopy (EIS) technique has been used as a sensitive method to explore the effect of antibacterial molecules on immobilized bacteria and biofilm formation. In this work, we describe the electrochemical spectroscopy as a powerful method to monitor the effect of Chlorhexidine Digluconate (CHX-Dg) on polyelectrolyte immobilized Escherichia coli K12 MG1655 and the kinetics of cell adhesion on gold electrodes. The experimental impedance data were modelised with a Zview program to find the best equivalent electrical circuit and analyse its parameter's properties. Polyelectrolyte multilayer formation on the electrode surface and bacteria immobilization greatly increased the electron-transfer resistance (Ret) and reduced the constant phase element (CPEdl). The effect of CHX-Dg was studied in a 0.5 × 10−4 mmol l−1 to 0.5 mmol l−1 range. The relation between the evolution of Ret and CHX-Dg concentration was found to be negatively correlated. When CHX-Dg was added, the electrochemical monitoring of the bacterial kinetic adhesion showed that the electrode's capacity (CP) variation remained stable, demonstrating that the addition of CHX-Dg in the broth inhibited bacterial adhesion.  相似文献

In order to study seminal roots morphology in barley grown under different water treatments, experiments were carried out under glasshouse-controlled conditions. Eight genotypes were cultivated under four water treatments (100, 75, 50 and 25% of field capacity). Seminal root length and root-to-shoot dry matters' ratio were measured. Root volume was assessed at three soil depths. Results showed broad genotypic differences for all traits. The effect of low and moderate water deficit was slight. In contrast, the impact of severe water treatment was strongly marked on all traits. The impact of water deficit intensity on root traits at different soil depths is discussed.  相似文献
