A study was performed to investigate the effect of weaning at 4 weeks of age on the activity of digestive enzymes in the stomach and pancreatic tissue and in digesta from 3 days prior to weaning to 9 days postweaning in 64 piglets. In stomach tissue the activity of pepsin and gastric lipase was determined. Pepsin activity declined abruptly after weaning but 5 days postweaning the weaning level was regained and in the gastric contents no change in pepsin activity was observed. Weaning did not influence the activity of gastric lipase. The activity of eight enzymes and a cofactor was measured in pancreatic tissue. The effect of weaning on the enzyme activity was highly significant for all enzymes except elastase. The activity of all enzymes remained at the weaning level during day 1–2 postweaning followed by a reduction of the activity. The activity of trypsin, carboxypeptidase A, amylase and lipase exhibited minimum activity 5 days postweaning. Trypsin activity increased to the preweaning level on day 7–9 whereas the activity of the others increased but did not reach the preweaning level. The activity of chymotrypsin, carboxypeptidase B and carboxyl ester hydrolase decreased during the entire experimental period. In digesta no effect of weaning was observed on the activity of amylase and trypsin. The activity of chymotrypsin was reduced after weaning in the proximal third of the small intestine and lipase and carboxyl ester hydrolase activity was reduced in the middle and distal parts of the small intestine after weaning. The present study shows that the activities of the digestive enzymes in the pancreatic tissue are affected by weaning. Even though the pancreatic secretion cannot be judged from these results they show that the enzymes respond differently to weaning. In general the activity of the digestive enzymes in pancreatic tissue is low on day 5 postweaning which in interaction with other factors may increase the risk of developing postweaning diarrhoea.  相似文献

We present the results of an individual-based simulation model, showing that increasing the mortality of non-breeding dispersers within settlement areas can lead to the extinction of species and (meta)populations in a subtle way. This is because the areas where dispersers settle are generally unknown or difficult to detect. Consequently, fewer efforts are devoted to the conservation of these sites than to the conservation of breeding territories. Additionally, high mortality rates affecting the floater sector of a population become evident in the breeding sector only after several of years, when it is too difficult or too late to halt the decline. As a result, because most conservation projects on endangered species and populations mainly focus on breeding areas, many current efforts may be wasted in locations other than those in which conservation would be really necessary and effective.  相似文献

Alzheimer disease (AD) is a neurodegenerative disorder that primarily causes β-amyloid accumulation in the brain, resulting in cognitive and behavioral deficits. AD patients, however, also suffer from severe circadian rhythm disruptions, and the underlying causes are still not fully known. Patients with AD show reduced systemic melatonin levels. This may contribute to their symptoms, since melatonin is an effective chronobiotic and antioxidant with neuroprotective properties. Here, the authors critically assessed the effects of long-term melatonin treatment on circadian system function, hippocampal oxidative stress, and spatial memory performance in the APPswe/PS1 double transgenic (Tg) mouse model of AD. To test if melatonin MT1/MT2 receptor activation, alone, was involved, the authors chronically treated some mice with the selective MT1/MT2 receptor agonist ramelteon. The results indicate that many of the circadian and behavioral parameters measured, including oxidative stress markers, were not significantly affected in these AD mice. During the day, though, Tg controls (Tg-CON) showed significantly higher mean activity and body temperature (BT) than wild-type (WT) mice. Overall, BT rhythm amplitude was significantly lower in Tg than in WT mice. Although melatonin treatment had no effect, ramelteon significantly reduced the amplitude of the BT rhythm in Tg mice. Towards the end of the experiment, Tg mice treated with ramelteon (Tg-RAM) showed significantly higher circadian rhythm fragmentation than Tg-CON and reduced circadian BT rhythm strength. The free-running period (τ) for the BT and locomotor activity (LA) rhythms of Tg-CON was <24 h. Whereas melatonin maintained τ at 24 h for BT and LA in both genotypes, ramelteon treatment had no effect. In the behavioral tests, the number of approaches and time spent exploring novel objects were significantly higher in Tg-CON than WT controls. Brain tissue analysis revealed significant reduction in hippocampal protein oxidation in Tg-MEL and Tg-RAM compared with Tg-CON animals. These results suggest that not all aspects of the circadian system are affected in the APPswe/PS1 mice. Therefore, care should be taken when extending the results obtained in Tg mice to develop new therapies in humans. This study also revealed the complexity in the therapeutic actions of melatonin and ramelteon in this mouse model of AD.  相似文献

We develop two individual-based models using a large and detailed data set (information gathered over more than a century) on a population of a longlived and territorial predator, the Spanish imperial eagle. We investigated the relationship between survival and predator pressure, prey behaviour and patch availability (i.e. settlement areas). Survival of dispersing individuals was highly dependent on the number of available settlement areas, mediated by prey availability. Changes in prey behaviour due to predation pressure (e.g. shifting from diurnal to nocturnal activity) can decrease their availability for predators even if the density significantly exceeds the predator needs. Environmental stochasticity had a strong influence on population viability when it occurred in a synchroneous way between breeding and settlement areas, and an increase in floater mortality negatively influenced stability and dynamics of the breeding segment of populations in reproductive areas. Our simulations demonstrated the link between the dynamics in settlement and breeding areas: factors affecting floater survival also influence whole population dynamics. Moreover, model outputs provided insights into the relationship between environmental stochasticity and population dynamics.  相似文献

Introduction

Angiogenesis is an important process in the development of destructive synovial pannus in rheumatoid arthritis (RA). The ITGAV +gene encodes a cell cycle-associated antigen, integrin ανβ 3, which plays a role in RA angiogenesis. Previously, two independent studies identified an association between the major allele of the ITGAV single-nucleotide polymorphism (SNP) rs3738919 and RA. We therefore tested this association in an independent study using New Zealand (NZ) and Oxford (UK) RA case control samples.

Methods

We compared genotype frequencies in 740 NZ Caucasian RA patients and 553 controls genotyped for rs3738919, using a polymerase chain reaction-restriction fragment length polymorphism assay. A TaqMan genotyping SNP assay was used to type 713 Caucasian RA patients and 515 control samples from Oxford for the rs3738919 variant. Association of rs3738919 with RA was tested in these two sample sets using the chi-square goodness-of-fit test. The Mantel-Haenszel test was used to perform a meta-analysis, combining the genetic results from four independent Caucasian case control cohorts, consisting of 3,527 cases and 4,126 controls. Haplotype analysis was also performed using SNPs rs3911238, rs10174098 and rs3738919 in the Wellcome Trust Case Control Consortium, NZ and Oxford case control samples.

Results

We found no evidence for association between ITGAV and RA in either the NZ or Oxford sample set (odds ratio [OR] = 0.88, Pallelic = 0.11 and OR = 1.18, Pallelic = 0.07, respectively). Inclusion of these data in a meta-analysis (random effects) of four independent cohorts (3,527 cases and 4,126 controls) weakens support for the hypothesis that rs3738919 plays a role in the development of RA (ORcombined = 0.92, 95% confidence interval 0.80 to 1.07; P = 0.29). No consistent haplotype associations were evident.

Conclusions

Association of ITGAV SNP rs7378919 with RA was not replicated in NZ or Oxford case control sample sets. Meta-analysis of these and previously published data lends limited support for a role for the ITGAV in RA in Caucasians of European ancestry.  相似文献

Background

The objective of this study was to evaluate angiogenesis according to CD34 antigen expression in estrogen receptor (ER)-positive and negative breast carcinomas.

Methods

This study comprised 64 cases of infiltrating ductal carcinoma in postmenopausal women divided into two groups: Group A: ER-positive, n = 35; and Group B: ER-negative, n = 29. The anti-CD34 monoclonal antibody was used as a marker for endothelial cells. Microvessel count was carried out in 10 fields per slide using a 40× objective lens (magnification 400×). Statistical analysis of the data was performed using Student's t-test (p < 0.05).

Results

The mean number of vessels stained with the anti-CD34 antibody in the estrogen receptor-positive and negative tumors was 23.51 ± 1.15 and 40.24 ± 0.42, respectively. The number of microvessels was significantly greater in the estrogen receptor-negative tumors (p < 0.001).

Conclusion

ER-negative tumors have significantly greater CD34 antigen expression compared to ER-positive tumors.

After some 70 years of debate on density-dependent regulation of animal populations, there is still poor understanding of where spatial and temporal density dependence occurs. Clearly defining the portion of the population that shapes density-dependent patterns may help to solve some of the ambiguities that encircle density dependence and its patterns. In fact, individuals of the same species and population can show different dynamics and behaviors depending on their locations (e.g., breeding vs. dispersal areas). Considering this form of intrapopulation heterogeneity may improve our understanding of density dependence and population dynamics in general. We present the results of individual-based simulations on a metapopulation of the Spanish imperial eagle Aquila adalberti. Our results suggest that high rates of floater mortality within settlement areas can determine a shift in the classical relationship (from negative to positive) between the fecundity (i.e., fledglings per pair) and density (i.e., number of pairs) of the breeding population. Finally, we proved that different initial conditions affecting the breeder portion of the population can lead to the same values of fecundity. Our results can represent a starting point for new and more complex approaches studying the regulation of animal populations, where the forgotten and invisible component--the floater--is taken into account.  相似文献

Objective

Patients undergoing immune modulatory therapies for the treatment of autoimmune diseases such as multiple sclerosis, and individuals with an impaired-immune system, most notably AIDS patients, are in the high risk group of developing progressive multifocal leukoencephalopathy (PML), an often lethal disease of the brain characterized by lytic infection of oligodendrocytes in the central nervous system (CNS) with JC virus (JCV). The immune system plays an important regulatory role in controlling JCV reactivation from latent sites by limiting viral gene expression and replication. However, little is known regarding the molecular mechanisms responsible for this regulation.

Methods and Results

Here, we investigated the impact of soluble immune mediators secreted by activated PBMCs on viral replication and gene expression by cell culture models and molecular virology techniques. Our data revealed that viral gene expression and viral replication were suppressed by soluble immune mediators. Further studies demonstrated that soluble immune mediators secreted by activated PBMCs inhibit viral replication induced by T-antigen, the major viral regulatory protein, by suppressing its expression in glial cells. This unexpected suppression of T-antigen was mainly associated with the suppression of translational initiation. Cytokine/chemokine array studies using conditioned media from activated PBMCs revealed several candidate cytokines with possible roles in this regulation. Among them, only IFN-γ showed a robust inhibition of T-antigen expression. While potential roles for IFN-β, and to a lesser extent IFN-α have been described for JCV, IFN-γ has not been previously implicated. Further analysis of IFN-γ signaling pathway revealed a novel role of Jak1 signaling in control of viral T-antigen expression. Furthermore, IFN-γ suppressed JCV replication and viral propagation in primary human fetal glial cells, and showed a strong anti-JCV activity.

Conclusions

Our results suggest a novel role for IFN-γ in the regulation of JCV gene expression via downregulation of the major viral regulatory protein, T-antigen, and provide a new avenue of research to understand molecular mechanisms for downregulation of viral reactivation that may lead to development of novel strategies for the treatment of PML.  相似文献

Spatio-temporal patterns of predator aggregations within their settlement areas (i.e. temporary settling zones used during dispersal or, more generally, foraging patches) were studied. By integrating the main behavioural rules of juveniles of Spanish imperial eagles Aquila adalberti during dispersal with the temporal availability of the eagles' main prey, we have developed several individual-based models under different simulation scenarios. The results suggest that the spatially synchrony of the aggregation patterns that we observed were derived from a combination of behavioural responses of the prey and the distances between available settlement areas. These results are discussed in the context of predator–prey relationships, optimal foraging and self-organizing processes.  相似文献

Furcraea foetida (Asparagaceae) is a native plant of Central America and northern South America but there is no information about its country of origin. The species was introduced into Brazil and is now considered invasive, particularly in coastal ecosystems. To date, nothing is known about the environmental factors that constrain its distribution and there is only inconclusive information about its location of origin. We used reciprocal distribution models (RDM) to assess invasion risk of F. foetida across Brazil and to identify source regions in its native range. We also tested the niche conservatism hypothesis using Principal Components Analyses and statistical tests of niche equivalency and similarity between its native and invaded ranges. For RDM analysis, we built two models using maximum entropy, one using records in the native range to predict the invaded distribution (forward‐Ecological Niche Model or forward‐ENM) and one using records in the invaded range to predict the native distribution (reverse‐ENM). Forward‐ENM indicated invasion risk in the Cerrado region and the innermost region of the Atlantic Forest, however, failed to predict the current occurrence in southern Brazil. Reverse‐ENM supported an existing hypothesis that F. foetida originated in the Orinoco river basin, Amazon basin and Caribbean islands. Prediction errors in the RDM and multivariate analysis indicated that the species expanded its realized niche in Brazil. The niche similarity test further suggested that the niche differences are because of differences in habitat availability between the two ranges, not because of evolutionary changes. We hypothesize that physiological pre‐adaptation (especially, the crassulacean acid metabolism), human‐driven propagule pressure and high competitive ability are the main factors determining the current spatial distribution of the species in Brazil. Our study highlights the need to include F. foetida in plant invasion monitoring programs, especially in priority conservation areas where the species has still not been introduced.  相似文献
