With the use of a patch-clamp technique in the whole-cell configuration, we studied the effects of pinacidil and its fluorine derivatives on A-type potassium current (I A) through the membrane of pyramidal neurons of the rat hippocampus. Hydrogen peroxide (10 mM) exerted no influence on the rate of inactivation ofI A; therefore, this current is probably mediated by Shal Kv4.2 potassium channels. Pinacidil demonstrated the properties of a weakI A blocker: in the 500 μM concentration it blocked about 45% of the current, while 50 μM of pinacidil fluorine derivatives were capable of blocking up to 30% ofI A. The effects of pinacidil and its derivatives showed no dependence on the stimulating potential. A similar pattern of the effects of pinacidil fluorine derivatives, which are an order of magnitude stronger than those of pinacidil itself, allows us to suppose that the imine nitrogen of the tested compounds is significantly more involved in the molecular interaction with the site of an A-type potassium channel than the pyridine nitrogen.  相似文献

The antitumor dose-dependent effect of binuclear dinitrosyl iron complexes with glutathione as NO donors on a solid tumor in the mouse, Lewis lung carcinoma, was detected. The complexes being injected at doses of 21, 42, 105 mg/kg daily for 10 days blocked completely the development of the tumor for the first week after tumor cell implantation into animals. After that, the part of tumor cells which remained in intact alive state began to grow at a rate equal to that for control animals. The effect was proposed to be caused via formation of an antinitrosative defense system in the cells as a response to NO attack on cells. It was also hypothesized that this system can be inactivated by higher doses of dinitrosyl iron complexes. Data were obtained which were in line with the hypothesis.  相似文献

The coenzymes Q and Vitamin K(3), as well as their synthetic analogues, have been found to inhibit free-radical processes of fragmentation of hydroxyl-containing organic compounds and oxidation of the latter by molecular oxygen. Effects of curcumin and related compounds on product formation in radiolysis of aerated and deaerated ethanol were studied. Ab initio calculations of enthalpy values relating to O-H bond dissociation and H-atom addition to > C = O bonds of the compounds under study have been performed. The obtained data allowed the conclusion that the presence of a 7-carbon chain containing conjugated > C = C < and > C = O bonds in the structures of curcumin and its analogues makes these compounds capable of inhibiting the reactions involving α-hydroxyl-containing carbon-centered radicals. This finding broadens the existing views concerning radical-regulating properties of curcuminoids, and it should be taken into account when practical use of these compounds is envisaged.  相似文献

Results are presented from experimental studies of the spatial electron density distribution in current sheets formed in three-dimensional magnetic configurations with X-lines. The electron density is measured by using two-exposure holographic interferometry. It is shown that plasma sheets can form in a magnetic configuration with an X-line in the presence of a sufficiently strong longitudinal magnetic-field component B when the electric current is excited along the X-line. As the longitudinal magnetic-field component increases, the electron density decreases and the plasma sheet thickness increases; i.e., the plasma is compressed into a sheet less efficiently.  相似文献

On modeled monolayer phospholipid (formed from azolectin) membranes, we studied the surface activity of optical isomers of a dipeptide, kyotorphin (Tyr-Arg), and a cardiotonic agent, suphan (N-succinyltryptophan potassium salt). It was found that the membranotropic activity of four studied isomers of kyotorphin is distributed in the order: LL>DL≈LD>DD, and two isomers (by tryptophan) of suphan as LL>DL. The data obtained suggest that the primary mechanism underlying binding of kyotorphin and suphan to the plasma membrane can be considered based on interaction of their molecules with the molecules of membrane phospholipids. Binding of molecules of kyotorphin and suphan by the lipid matrix to the plasma membrane and/or their incorporation into the matrix is a result of the above interaction.  相似文献

Results are presented from studies of the structure and dynamics of current sheets in three-dimensional magnetic configurations with an X line by means of holographic interferometry. It is found that the efficiency of plasma compression into the sheet is reduced as the longitudinal magnetic field B z , directed along the X line, increases. This effect is attributed to the enhancement of the longitudinal component of the magnetic field within the sheet and to the corresponding increase in the magnetic pressure. It is shown that the formation of a plasma sheet lags behind the beginning of the plasma current pulse, the delay time being close to the characteristic Alfvén time.  相似文献

Complete genome DNA sequence and analysis is presented for Wolbachia, the obligate alpha-proteobacterial endosymbiont required for fertility and survival of the human filarial parasitic nematode Brugia malayi. Although, quantitatively, the genome is even more degraded than those of closely related Rickettsia species, Wolbachia has retained more intact metabolic pathways. The ability to provide riboflavin, flavin adenine dinucleotide, heme, and nucleotides is likely to be Wolbachia's principal contribution to the mutualistic relationship, whereas the host nematode likely supplies amino acids required for Wolbachia growth. Genome comparison of the Wolbachia endosymbiont of B. malayi (wBm) with the Wolbachia endosymbiont of Drosophila melanogaster (wMel) shows that they share similar metabolic trends, although their genomes show a high degree of genome shuffling. In contrast to wMel, wBm contains no prophage and has a reduced level of repeated DNA. Both Wolbachia have lost a considerable number of membrane biogenesis genes that apparently make them unable to synthesize lipid A, the usual component of proteobacterial membranes. However, differences in their peptidoglycan structures may reflect the mutualistic lifestyle of wBm in contrast to the parasitic lifestyle of wMel. The smaller genome size of wBm, relative to wMel, may reflect the loss of genes required for infecting host cells and avoiding host defense systems. Analysis of this first sequenced endosymbiont genome from a filarial nematode provides insight into endosymbiont evolution and additionally provides new potential targets for elimination of cutaneous and lymphatic human filarial disease.  相似文献
