The distribution of ATP in different parts of the frog olfactory organ was investigated. It is demonstrated that the highest macroerg amount is characteristic of the receptor part of olfactory mucosa. The amount of ATP decreased after the stimulation of the olfactory mucosa by odorants. The role of ATP in olfactory receptor process is discussed.  相似文献

Macroelectrodes and electrophysiological setup were used in experiments with stems of 15-day-old pumpkin (Cucurbita pepo L.) seedlings for computer-assisted data recording. It is shown that local bioelectric responses induced by graded local chilling are similar to the receptor potentials of animals. These responses increase gradually with stimulus strength and trigger the action potential generation when a temperature threshold is attained. The excitation threshold of cells in seedling stems displays the phenomenon of accommodation. Parameters of local bioelectric responses induced by intermittent cooling can undergo changes similar to sensitization-habituation patterns. The results indicate that local electrical responses may be involved in early stages of cooling perception in cells of higher plants devoid of locomotive functions.  相似文献

The signaling role of action potential (AP) in higher plants is considered. The principles underlying realization of this role and the significance of AP-induced short-term effector response are discussed. Bioluminescent analysis has been made of the effect of oxygen supply on the content of ATP in the isolated olfactory epithelium of the frog. It was shown that storage of epithelium preparations in the air increases their ATP content. When preparations are kept in the atmosphere of an inert gas, ATP level in the epithelium rapidly decreases, being recovered after transition of preparations to the air medium. The data obtained indicate the existence of apical type of respiration in the olfactory epithelium of the frog.  相似文献

The nature of subthreshold changes in excitable plasma membranes has been investigated in stem parenchyma cells of Cucurbita pepo L. during action-potential generation induced by gradual cooling (from 23 to 10 ° C). The character of the subthreshold depolarization of excitable cells is shown to be mainly defined by a decrease in the activity of the plasma-membrane electrogenie pump (H+-ATPase). In its turn, the pump activity is controlled by thermal changes in the structure of the membrane lipid matrix. Generation of electric (delta psi) and chemical (delta pH) components of electrochemical proton gradient delta muH+, in plasma membrane vesicles of Heracleum sosnovskyi phloem cells was investigated. ATP-dependent generation of delta psi at pH 6.0 in the presence of Mg2+ and K+ was established with the help of fluorescent probes AU+ and ANS-. Protonophore CCCP and proton ATPase inhibitor DCCD suppressed generation, whereas oligomycin, the inhibitor of mitochondrial ATPases did not affect it. Measurings of delta psi value indicated its oscillations within the limits from 10 to 60 mV. ATP-dependent generation of delta pH was established by means of fluorescent probe 9-AA. The effect was eliminated by CCCP and stimulated by K+, that may testify to the transformation of a part of delta psi into delta pH at antiport H+/K+. Existence of H+-ATPase in the plasma membranes of higher plant cells insuring generation of delta muH+ is supposed.  相似文献

A mathematical model is built for the kinetics of delayed luminescence, considering the states of the donor and acceptor sides of photosystem II and the electrochemical gradient on the thylakoid membrane. It is used to assess the possible mechanisms of the experimentally observed biphasic changes in delayed luminescence caused by variation potentials (Sukhov et al., Biophysics 53 (3), 223 (2008)).  相似文献

The inside-out vesicles of plasma membranes were isolated from pumpkin stem cells, and the kinetics of sucrose efflux induced by the K+-diffusion potential (D) was studied by measuring light transmission. Two phases differing in their rates and duration were identified in D-dependent changes of light transmission. The increase in Delevated the rate and magnitude of the fast phase in light transmission changes but did not markedly affect the rate of the slow phase. These two phases also differed in their sensitivity to inhibitors and to changes in sucrose concentration in the external medium. Measurements of Dduring sucrose transport by means of the fluorescence probe dis-C3-(5) revealed differences in the magnitude of Dand its stability in vesicles loaded with sucrose and mannitol, as well as under the action of inhibitors. The two-phase dependence of sucrose efflux from vesicles on the applied diffusion potential is discussed in the context of modern concepts on the functioning of sucrose carriers in the membranes.  相似文献

A pH-sensitive electrode was applied to measure activity of H+ ions in the medium surrounding excitable cells of pumpkin (Cucurbita pepo L.) seedlings during cooling-induced generation of action potential (AP). Reversible alkalization shifts were found to occur synchronously with AP, which could be due to the influx of H+ ions from external medium into excitable cells. Ethacrynic acid (an anion channel blocker) reduced the AP amplitude but had no effect on the transient alkalization of the medium. An inhibitor of plasma membrane H+-ATPase, N,N’-dicyclohexylcarbodiimide suppressed both the AP amplitude and the extent of alkalization. In experiments with plasma membrane vesicles, the hydrolytic H+-ATPase activity was subjected to inhibition by Ca2+ concentrations in the range characteristic of cytosolic changes during AP generation. The addition of a calcium channel blocker verapamil and a chelating agent EGTA to inhibit Ca2+ influx from the medium eliminated the AP spike and diminished reversible alkalization of the external solution. An inhibitor of protein kinase, H-7 alleviated the inhibitory effect of Ca2+ on hydrolytic H+-ATPase activity in plasma membrane vesicles and suppressed the reversible alkalization of the medium during AP generation. The results provide evidence that the depolarization phase of AP is associated not only with activation of chloride channels and Cl? efflux but also with temporary suppression of plasma membrane H+-ATPase manifested as H+ influx. The Ca2+-induced inhibition of the plasma membrane H+-ATPase is supposedly mediated by protein kinases.  相似文献

It was shown that local damaging stimulation of geranium graft induced the generation and propagation of electrical signals (variation potential), which changed the parameters of delayed luminescence in the leaf blade. The responses of delayed luminescence included two stages, which probably occur by different mechanisms.  相似文献
