Hepatic sarcocystosis was diagnosed in a horse in association with refractory bacterial osteomyelitis and plasma cell tumor of the maxilla and hepatic salmonellosis. Gross lesions included pleural, pericardial, and peritoneal effusions, hepatomegaly, gastric ulceration, colonic edema, and proliferative tissues filling 2 maxillary dental alveoli. Histologically, liver was characterized by severe suppurative, necrotizing, periportal hepatitis, and severe periacinar necrosis. Hepatocytes frequently contained protozoal schizonts in various stages of development. In mature schizonts, merozoites were often arranged radially around a central residual body, consistent with asexual division by endopolygeny. Ultrastructural features of merozoites included an apical conoid and polar ring, anterior micronemes, central nuclei, and absence of rhoptries. These protozoa did not react to antisera raised against Neospora caninum, Sarcocystis neurona, Toxoplasma gondii, or Hammondia hammondi. The microscopic and ultrastructural characteristics and immunoreactivity of this organism are consistent with a Sarcocystis sp. other than S. neurona. This is the first report of Sarcocystis-associated hepatitis in a horse. The life cycle of this organism and source of infection are unknown.  相似文献

Primary amines covalently bonded to the surface of poly(dimethylsiloxane) were obtained by hydrosilylation grafting of aminopropyl vinyl ether to Si-H groups formed during argon plasma treatment. The amine groups were derivatized using pentafluorobenzaldehyde and characterized by X-ray photoelectron spectroscopy. The graft yield was about 3% grafted molecules within the depth of the analysis. The terminal aldehyde groups of diazotized heparin was also coupled to the primary amines. This led to a silicone elastomer with covalently bonded heparin which was expected to be hydrolytically stable. This method of bonding primary amines to the surface of silicone elastomers and the subsequent coupling of aldehyde-containing molecules is a promising way of obtaining novel biomaterials.  相似文献

A phenomenological mixture model is presented for interactions between biosynthesis of extracellular matrix (ECM) constituents and ECM linking in a scaffold seeded with chondrocytes. A system of three ordinary differential equations for average apparent densities of unlinked ECM, linked ECM and scaffold is developed along with associated initial conditions for scaffold material properties. Equations for unlinked ECM synthesis and ECM linking include an inhibitory mechanism where associated rates decrease as unlinked ECM concentration in the interstitial fluid increases. Linking rates are proposed to depend on average porosity in the evolving tissue construct. The resulting initial value problem contains nine independent parameters that account for scaffold biomaterial properties and interacting mechanisms in the engineered system. Effects of parameter variations on model variables are analyzed relative to a baseline case with emphasis on the evolution of solid phase apparent density, which is often correlated with the compressive elastic modulus of the tissue construct. The new model provides an additional quantitative framework for assessing and optimizing the design of engineered cell-scaffold systems and guiding strategies for articular cartilage tissue engineering.  相似文献

The majority of deforested land in the Amazon Basin has become cattle pasture, making forest‐to‐pasture conversion an important contributor to the carbon (C) and climate dynamics of the region. However, our understanding of biogeochemical dynamics in pasturelands remains poor, especially when attempting to scale up predictions of C cycle changes. A wide range of pasture ages, soil types, management strategies, and climates make remote sensing the only realistic means to regionalize our understanding of pasture biogeochemistry and C cycling over such an enormous geographic area. However, the use of remote sensing has been impeded by a lack of effective links between variables that can be observed from satellites (e.g. live and senescent biomass) and variables that cannot be observed, but which may drive key changes in C storage and trace gas fluxes (e.g. soil nutrient status). We studied patterns in canopy biophysical–biochemical properties and soil biogeochemical processes along pasture age gradients on two important soil types in the central Amazon. Our goals were to (1) improve our understanding of the plot‐scale biogeochemical dynamics of this land‐use change, (2) evaluate the effects of pasture development on two contrasting soil types (clayey Oxisols and sandy Entisols), and (3) attempt to use remotely sensed variables to scale up the site‐specific variability in biogeochemical conditions of pasturelands. The biogeochemical analyses showed that (1) aboveground and soil C stocks decreased with pasture age on both clayey and sandy soils, (2) declines in plant biomass were well correlated with declines in soil C and with available phosphorus (P) and calcium (Ca), and (3) despite low initial values for total and available soil P, ecosystem P stocks declined further with pasture age, as did a number of other nutrients. Spectral mixture analysis of Landsat imagery provided estimates of photosynthetic vegetation (PV) and non‐photosynthetic vegetation (NPV) that were highly correlated with field measurements of these variables and plant biomass. In turn, the remotely sensed sum PV+NPV was well correlated with the changes in soil organic carbon and nitrogen, and available P and Ca. These results suggest that remote sensing can be an excellent indicator of not only pasture area, but of pasture condition and C storage, thereby greatly improving regional estimates of the environmental consequences of such land‐use change.  相似文献

This study investigated beliefs about climate change among Swedish secondary school students at the end of their K-12 education. An embedded mixed method approach was used to analyse 51 secondary school students’ written responses to two questions: (1) What implies climate change? (2) What affects climate? A quantitative analysis of the responses revealed that ‘Earth’, ‘human’ and ‘greenhouse effect’ were frequent topics regarding the first question, and ‘pollution’, ‘atmosphere’ and ‘Earth’ were frequent regarding the second. A qualitative analysis, based on a ‘conceptual elements’ framework, focused on three elements within responses: atmosphere (causes and/or consequences), Earth (causes and consequences) and living beings (humans and/or animals and their impacts on climate change). It revealed a predominantly general or societal, rather than individual, perspective underlying students’ responses to the second question. The ability to connect general/societal issues with individual issues relating to climate change could prompt students to reflect on the contributions of individuals towards climate change mitigation, thereby constituting a basis for decision-making to promote a sustainable environment. Although the students did not discuss climate changes from an individual perspective, their statements revealed their understanding of climate change as a system comprising various components affecting the overall situation. They also revealed an understanding of the difference between weather and climate.  相似文献

Fibrin-enhanced endothelial cell organization   总被引:12,自引:0,他引:12  
We examined the synthesis of extracellular matrix macromolecules by human microvascular endothelial cells isolated from the dermis of neonatal (foreskin) and adult (abdominal) skin. Electron microscopy showed that both cell types produced an extracellular matrix that was strictly localized to the subendothelial space. The subendothelial matrices were initially deposited as a single discontinuous layer of filamentous, electron-dense material that progressively became multilayered. Biosynthetic studies indicated that 2-4% of the newly synthesized protein was deposited in the subendothelial matrices by both cell types. Approximately 15-20% of the radiolabeled protein was secreted into the culture medium, and the remainder was confined to the cellular compartment. Biochemical and immunochemical analyses demonstrated the extracellular secretion of type IV collagen, laminin, fibronectin, and thrombospondin by the newborn and adult cells. Whereas type IV collagen was the predominant constituent of the matrix, fibronectin was secreted into the medium, with only small amounts being deposited in the matrix. Thrombospondin was a major constituent of the matrix produced by the newborn foreskin cells but was virtually absent in the matrix elaborated by the adult cells. However, both cell types did release comparable amounts of thrombospondin into their medium. Immunoperoxidase staining for type IV collagen revealed a fibrillar network in the subendothelial matrices produced by both adult and neonatal cells. In contrast, thrombospondin, which was detected only in the matrix of newborn cells, exhibited a spotty and granular staining pattern. The results indicate that the extracellular matrices synthesized by cultured human microvascular endothelial cells isolated from anatomically distinct sites and different stages of development and age are similar in ultrastructure but differ in their macromolecular composition.  相似文献

A growth inhibition test method was developed using the macroalga Gracilaria tenuistipitata as the test organism. This alga was chosen because of its high laboratory growth rates, commonly 30–40% d–1, which are reached in salinities between 5 and 40, and its epiphyte resistance. The toxicity of a number of substances, including heavy metals, herbicides and complex wastewaters towards the alga was assayed. Anti-fouling paints were tested with a modification of the method. EC50 values for heavy metals varied between 0.05 and 17 mg l–1 and for herbicides between 0.002 and 0.02 mg l–l. The sensitivity to the toxicant was generally higher at low salinity. Omitting nitrogen and phosphorus additions to the test medium increased the sensitivity and a semi-static performance was possible with maintained or increased sensitivity. Preliminary tests done with a computerised photosynthesis inhibition method produced promising results.In conclusion, this is a simple, sensitive and reproducible test method for assessing the toxicity of substances, wastewaters and anti-fouling paints in brackish and marine environments.  相似文献

Human vascular permeability factor (hVPF) is a glycoprotein that promotes fluid and protein leakage from blood vessels. The function of hVPF is at present unknown, but the potent bioactivities of this protein suggest that it could act during inflammation, wound healing, and tumor angiogenesis. hVPF was purified from serum-free conditioned medium of the human histiocytic lymphoma cell line U937 as a disulfide-linked dimeric 40-kDa protein that promoted dermal blood vessel leakage in guinea pigs at a dose of 20 ng (3 x 10(-9) M) and promoted in vitro endothelial cell growth at concentrations as low as 50 PM. Multiple forms of hVPF with apparent pI values greater than 7.5 were resolved using pH gradient electrophoresis. Antibodies against guinea pig vascular permeability factor were found to cross-react with hVPF. The N-terminal amino acid sequence of hVPF was similar to, but not identical with, the N-terminal sequence of guinea pig vascular permeability factor.  相似文献
