The inside-out patch-clamp technique was applied to the plasmolyzedplasmalemma of inter-nodes of Chara corallina without enzymatictreatment. We found two different types of channel activitythat were CP-sensitive. Both types of channel were Ca2+-dependent.However, the one that exhibited greater dependence on Ca2+ ionswas the focus of our studies, and we named it the Ca2+-dependentCP-sensitive anion channel. When the concentration ofCa2+ ions on the cyto-plasmic side was 1.0 µM, the Ca2+-dependentCP-sensitive channel opened most frequently between approximately–80 and –100 mV. At 10 µM Ca2+, it openedless frequently, and at 0.1 µM Ca2+ it scarcely openedat all. These observations indicate that the anion channel ofinterest is voltage-dependent over a restricted range of concentrationsof Ca2+ ions. The dependence on Ca2+ and voltage of the channelcan explain the behavior of the excitable Ca2+-activated Clchannel in the Chara plasmalemma. A search was made for inhibitors of Pi uptake that act directlyon the Pi transporter in the plasma membranes of Catharanthusroseus cells to inhibit Pi uptake without inhibition of protonpumping. Using standard electrodes, we monitored changes inpH and in the concentration of K+ ions, as well as the rateof Pi uptake, when an inhibitor to be tested was applied tothe cells in unbuffered medium. A9C (28 µM), a blockerof anion channels, inhibited Pi uptake but it also inhibitedthe proton pump. However, a structurally similar inhibitor,furosemide, inhibited Pi uptake without inhibiting proton pumping. It is suggested that the carboxylic group of these inhibitorsinteracts with the Pi-binding site (probably an amino group)of the Pi transporter in the plasma membrane and that the hydrophobicstructure of these inhibitors facilitates their accumulationin the plasma membrane. 3Present address: Department of Biology, Hitotsubashi University,2-1 Naka, Kunitachi, Tokyo, 186 Japan  相似文献

In our previous paper (Kimura, Y., et al., Biosci. Biotechnol. Biochem., 67, 1852-1856, 2003), we found that a complex type N-glycans containing beta1-3 galactose residue occurs on royal jelly glycoproteins. During structural analysis of minor components of royal jelly N-glycans, we found complex type N-glycans bearing both galactose and N-acetylgalactosamine residues. Detailed structural analysis of pyridylaminated oligosaccharide revealed that the newly found N-glycan had a complex type structure harboring a tumor marker (T-antigen) unit: Galbeta1-3GalNAcbeta1-4GlcNAcbeta1-2Manalpha1-6 (Galbeta1-3GalNAcbeta1-4GlcNAcbeta1-2Manalpha1-3) Manbeta1-4GlcNAcbeta1-4GlcNAc. To our knowledge, this may be the first report of the presence of the T-antigen unit in the N-glycan moiety of eucaryotic glycoproteins.  相似文献

The 3- and 21-monosulfates of pentadeuterated 5β-tetrahydrocorticosteroides were synthesized, starting from cortisol and 11-deoxycotisol. The principal reactions used were (1) perdeuteration of the methylene groups adjacent to the 3-oxo group of 17,20:20,21-bismethylendioxy-5β-3-ketosteroids with NaOD in CH3OD followed by stereoselective reduction with NaBD4, (2) sulfation of hydroxy groups with sulfur trioxide–trimethylamine complex, and (3) removal of the 17,20:20,21-bismethylendioxy group with hydrogen fluoride. The labeled compounds can be used as internal standards in liquid chromatography/mass spectrometry assays for clinical and biochemical studies.  相似文献

The effects of calmodulin (from spinach) on the Ca2+-dependentCl--sensitive anion channel in the Chara plasmalemma (Okiharaet al. 1991) were studied by the inside-out patch-clamp technique.The current of Cl- ions, which flowed through the channel at1.0 µM Ca2+, tended to decrease irregularly with time.This tendency toward a decrease in the current was no longerapparent after application of calmodulin for some time. Theactivity of the channel was restored to a small extent or tendedto increase during the early stages of application of calmodulin.Such a transient action of calmodulin on the channel activitywas evident, at voltages more negative than –100 mV. The effects of lidocaine, tetracaine, procaine and bupivacaine (less than 1000 microM) on the Chara corallina internodal cell were studied. These local anesthetics depolarized the membrane at rest, while they affected the rising phase and the peak level of action potential not appreciably. Instead, they prolonged the time course of the falling phase of action potential as slowly as the repolarization was imperfect, even after enough lapse beyond the refractory period. Consequently, an action potential appeared to enhance the degree of depolarization at rest. Such a depolarization with stimulus/excitation was named use-dependent depolarization, while the depolarization without excitation, the resting one. The order of the potency of the use-dependent depolarization almost coincided with that of the nerve-blocking potency. During depolarization the change in membrane conductance was not simple. However, the conductance-voltage (Gm-Vm) relationship curve in the presence of local anesthetic suggested that depolarization was due to, not only the decrease in the electrogenic H(+)-pump, but also the increase in the diffusion conductance.  相似文献

On incubation with rat liver microsomes plus NADPH, Denmert (S-n-butyl S′-p-tert-butylbenzyl N-3-pyridyldithiocarbonimidate) underwent at least two different types of oxidation; hydroxylation and sulfoxidation. Hydroxylation of Denmert at the tert-butyl group was one of the major metabolic attacks in mammalian metabolism. Sulfoxidation of Denmert gave two isomers of Denmert sulfoxides which were intermediates in the metabolism and readily transformed into 2-(3′-pyridylimmo)-4-carboxylthiazolidine (HM) in the presence of l-cysteine without enzymatic mediation. This type of conjugation with cysteine appears to be a new class of metabolic reactions in mammals. Denmert S-oxide showed increased fungicidal activity when assayed in liquid cultures, but not on plant leaves.  相似文献

Elsewhere, we characterized the structure of twelve N-glycans purified from royal jelly glycoproteins (Kimura, Y. et al., Biosci. Biotechnol. Biochem., 64, 2109-2120 (2000)). Structural analysis showed that the typical high-mannose type structure (Man9-4GlcNAc2) accounts for about 72% of total N-glycans, a biantennary-type structure (GlcNAc2Man3GlcNAc2) about 8%, and a hybrid-type structure (GlcNAc1Man4GlcNAc2) about 3%. During structural analysis of minor N-glycans of royal jelly glycoproteins, we found that one had an N-acetyl-galactosaminyl residue at the non reducing end; most of such residues have been found in N-glycans of mammalian glycoproteins. By exoglycosidase digestion, methylation analysis, ion-spray (IS)-MS analysis, and 1H NMR spectroscopy, we identified the structure of the N-glycan containing GalNAc as; GlcNAc(beta)1-2Man(alpha)1-6(GalNAcbeta1 - 4GIcNAcbeta1 - 2Man(alpha)1 - 3)Manbeta1 - 4GlcNAc(beta)1-4GlcNAc. This result suggested that a beta1-4 GalNAc transferase is present in hypopharyngeal and mandibular glands of honeybees.  相似文献

Detection of peripheral nerve tissues during surgery is required to avoid neural disturbance following surgery as an aspect of realizing better functional outcome. We provide a proof-of-principle demonstration of a label-free detection technique of peripheral nerve tissues, including myelinated and unmyelinated nerves, against adjacent tissues that employ spontaneous Raman microspectroscopy. To investigate the Raman spectral features of peripheral nerves in detail, we used unfixed sectioned samples. Raman spectra of myelinated nerve, unmyelinated nerve, fibrous connective tissue, skeletal muscle, tunica media of blood vessel, and adipose tissue of Wistar rats were analyzed, and Raman images of the tissue distribution were constructed using the map of the ordinary least squares regression (OLSR) estimates. We found that nerve tissues exhibited a specific Raman spectrum arising from axon or myelin sheath, and that the nerve tissues can be selectively detected against the other tissues. Moreover, myelinated and unmyelinated nerves can be distinguished by the intensity differences of 2,855 cm?1, and 2,945 cm?1, which are mainly derived from lipid and protein contents of nerve fibers. We applied this method to unfixed section samples of human periprostatic tissues excised from prostatic cancer patients. Myelinated nerves, unmyelinated nerves, fibrous connective tissues, and adipose tissues of the periprostatic tissues were separately detected by OLSR analysis. These results suggest the potential of the Raman spectroscopic observation for noninvasive and label-free nerve detection, and we expect this method could be a key technique for nerve-sparing surgery.  相似文献

14C-Denmert(S-n-butyl S′-p-tert-butylbenzyl N-3-pyridyldithiocarbonimidate, S–1358) labeled at the methylene of the benzyl group was gradually decomposed to yield a number of products, when exposed to sunlight on thin-layer plates or in water solution, applied to plant foliage or nutrient solution, and added to soils under upland conditions. The identified products were almost common to plants, soils and light. The primary reactions occurred: (1), oxidation of the sulfur atoms; (2), cleavage of the dithiocarbonimidate linkage, and (3), oxidation of the methylene of the benzyl group. Also, hydroxylation at the tert-butyl group attached to the benzyl moiety slightly took place in soils. Although radioactivity was absorbed by the plant through leaves or roots, translocation into other parts of the plant occurred to a very small extent. Denmert and its degradation products were hardly leached through the acidic soils tested.  相似文献
