Non-chromotropic substances such as fibrin and gelatin and most tissue and cellular structures stain orthochromatically with internal dye concentrations of such metachromatic dyes as methylene blue and toluidine blue which, if in solution, would be metachromatic. Therefore, at ordinary levels of staining these substances depress the natural tendency of these dyes to change color. However, at elevated levels of dye-binding metachromasy eventually occurs. This phenomenon is explained on the basis of the distribution of dye-binding sites. In these substrates, by contrast with chromotropic substances, many binding sites are too far removed for dye interaction, consequently the interaction frequency can become high enough to produce a color change only as saturation of the available sites is approached. It is also shown that the destruction of color is a characteristic of metachromasy and that water molecules intercalated between approximated dye ions are responsible for the loss and change of color. A concept of metachromasy is proposed in which the interaction between water molecules and suitably approximated dye ions plays an essential role. The experimental studies are described against a background of the history and evolution of ideas on metachromasy. The literature is reviewed and reassessed particularly from the physicochemical viewpoint.  相似文献

We report the discovery, characterization and validation of 118 single nucleotide polymorphisms (SNPs) for poplar leaf rust Melampsora medusae f. sp. deltoidae identified using a gene‐targeted approach in an expressed sequence tag (EST) library. Although mature black spruce forests are a dominant cover type in the boreal forest of North America, it is not clear how their carbon (C) budgets vary across the continent. The installation of an eddy covariance flux tower on an Old Black Spruce (OBS) site in eastern Canada (EOBS, Québec) provided a first opportunity to compare and contrast its annual (2004) and seasonal C exchange with two other pre-existing OBS flux sites from different climatic regions located in Saskatchewan [Southern OBS (SOBS)] and Manitoba [Northern OBS (NOBS)]. Although there was a relatively uniform seasonal pattern of net ecosystem productivity (NEP) among sites, EOBS had a lower total annual NEP than the other two sites. This was primarily because warmer soil under a thicker snowpack at EOBS appeared to increase winter C losses and low light suppressed both NEP and gross ecosystem productivity (GEP) in June. Across sites, greater total annual GEP and ecosystem respiration ( R ) were associated with greater mean annual air temperatures and an earlier beginning of the growing season. Also, GEP at all three sites showed a stronger relationship with air temperature in spring and early summer compared with later in the growing season, highlighting the importance of springtime conditions to the C budget of these boreal ecosystems. The three sites had different parameter estimates describing the responses of R and GEP at the half hour time scale to near surface temperature and light, respectively. On the other hand, the responses of both R and GEP to temperature at the monthly scale did not differ among sites. These results suggest that a general parameterization could be sufficient at coarse time resolutions to model the response of C exchange to environmental factors of mature black spruce forests from different climatic regions.  相似文献

The influence of climatic and local nonclimatic factors on the fire regime of the eastern Canadian boreal forest over the last 8000 years is investigated by examining charred particles preserved in four lacustrine deposits. Herein, we compare the distribution of fire-free intervals (FFIs) and the synchronicity of fire events among sites, using Ripley's K -function to determine the extent of the role of local-scale vs. large-scale processes with respect to fire control. Between 8000 and 5800 cal. bp (calibrated years before present) the climatic and ecological conditions were less conducive to fire events than after this date. After 5800 cal. bp , the number of fires per 1000 years (fire frequency) progressively increased, reaching a maximum ca. 3400 cal. bp . There was a sharp decrease in fire frequency during the last 800 years. Between 8000 and 4000 cal. bp , comparable FFIs and synchronous fire episodes were determined for the study sites. During this period, the fire frequency was predominantly controlled by climate. After 4000 cal. bp , two sites displayed independent fire histories (different FFI distributions or asynchronous fire events), underlining the important influence of local factors, including short-term fuel wetness, characteristics of the watershed and landscape connectivity, in determining fire occurrence. We conclude that climatic changes occurred during the last 4000 years that induced a rise in the water table; this may explain the high spatial heterogeneity in fire history. Current and projected global climatic changes may cause similar spatial variability in fire frequency.  相似文献

To address the central question of how climate change influences tree growth within the context of global warming, we used dendroclimatological analysis to understand the reactions of four major boreal tree species –Populus tremuloides, Betula papyrifera, Picea mariana, and Pinus banksiana– to climatic variations along a broad latitudinal gradient from 46 to 54°N in the eastern Canadian boreal forest. Tree‐ring chronologies from 34 forested stands distributed at a 1° interval were built, transformed into principal components (PCs), and analyzed through bootstrapped correlation analysis over the period 1950–2003 to identify climate factors limiting the radial growth and the detailed radial growth–climate association along the gradient. All species taken together, previous summer temperature (negative influences), and current January and March–April temperatures (positive influences) showed the most consistent relationships with radial growth across the gradient. Combined with the identified species/site‐specific climate factors, our study suggested that moisture conditions during the year before radial growth played a dominant role in positively regulating P. tremuloides growth, whereas January temperature and growing season moisture conditions positively impacted growth of B. papyrifera. Both P. mariana and P. banksiana were positively affected by the current‐year winter and spring or whole growing season temperatures over the entire range of our corridor. Owing to the impacts of different climate factors on growth, these boreal species showed inconsistent responsiveness to recent warming at the transition zone, where B. papyrifera, P. mariana, and P. banksiana would be the most responsive species, whereas P. tremuloides might be the least. Under continued warming, B. papyrifera stands located north of 49°N, P. tremuloides at northern latitudes, and P. mariana and P. banksiana stands located north of 47°N might benefit from warming winter and spring temperatures to enhance their radial growth in the coming decades, whereas other southern stands might be decreasing in radial growth.  相似文献
