The effects on a cloned DNA fragment carrying an actinomycin resistance determinant on physiological processes in strains of streptomycetes with various potencies in producing this antibiotic, their inactive mutants, and the model strain ofStreptomyces lividans66 were studied. A study of the microflora of structural surfaces on board the russian segment of the international space station (RS ISS) has demonstrated that the species composition that is formed during the mission is similar to that on board the mir station. the microflora of the sampled sites was similar to that of residential and industrial buildings on earth. it was represented by 17 species of fungi, seven genera of bacteria, and three genera of actinomycetes. the degree of colonization of the surfaces of structural materials of the rs iss by destructive microorganisms was low and showed undulating dynamics. the mycelial fungi included “technophile” species, which caused biological corrosion of an aluminum-magnesium alloy in simulation experiments.  相似文献

Cell surface of leishmaniasis causal agent, a parasitic member of Protozoa of Leishmania genus, is covered by thick glycocalix consisting of various phosphatidylinositol-anchored molecules. This review deals with the structure and biosynthesis of the main phosphoglycans and glycoproteins of Leishmania cell surface, many of which incorporate the rare natural D-arabinopyranose, and the problem concerning the involvement of these molecules in support of Leishmania survival during their intricate life cycle is discussed.  相似文献

A collection of actinomycin-producing Streptomyces strains, their variants with different levels of antibiotic biosynthesis, and recombinant strains were screened in order to select new strains that produce polyketide antibiotics. Screening with the use of the cloned act gene encoding a component of actinorhodin polyketide synthase (PKS) multienzyme complex from Streptomyces coelicolor revealed that many strains tested can synthesize polyketide antibiotics along with actinomycins. A relationship between biosynthetic pathways of actinomycins and polyketides is discussed.  相似文献

Schistosomes are amongst the most important and neglected pathogens in the world, and schistosomiasis control relies almost exclusively on a single drug. The neuromuscular system of schistosomes is fertile ground for therapeutic intervention, yet the details of physiological events involved in neuromuscular function remain largely unknown. Short amidated neuropeptides, FMRFamide-like peptides (FLPs), are distributed abundantly throughout the nervous system of every flatworm examined and they produce potent myoexcitation. Our goal here was to determine the mechanism by which FLPs elicit contractions of schistosome muscle fibers. Contraction studies showed that the FLP Tyr-Ile-Arg-Phe-amide (YIRFamide) contracts the muscle fibers through a mechanism that requires Ca2+ influx through sarcolemmal voltage operated Ca2+ channels (VOCCs), as the contractions are inhibited by classical VOCC blockers nicardipine, verapamil and methoxyverapamil. Whole-cell patch-clamp experiments revealed that inward currents through VOCCs are significantly and reversibly enhanced by the application of 1 µM YIRFamide; the sustained inward currents were increased to 190% of controls and the peak currents were increased to 180%. In order to examine the biochemical link between the FLP receptor and the VOCCs, PKC inhibitors calphostin C, RO 31–8220 and chelerythrine were tested and all produced concentration dependent block of the contractions elicited by 1 µM YIRFamide. Taken together, the data show that FLPs elicit contractions by enhancing Ca2+ influx through VOCC currents using a PKC-dependent pathway.  相似文献

Interspecies variations in lipophosphoglycan (LPG) have been the focus of intense study over the years due its role in specificity during sand fly-Leishmania interaction. This cell surface glycoconjugate is highly polymorphic among species with variations in sugars that branch off the conserved Gal(β1,4)Man(α1)-PO(4) backbone of repeat units. However, the degree of intraspecies polymorphism in LPG of Leishmania infantum (syn. Leishmania chagasi) is not known. In this study, intraspecific variation in the repeat units of LPG was evaluated in 16 strains of L. infantum from Brazil, France, Algeria and Tunisia. The structural polymorphism in the L. infantum LPG repeat units was relatively slight and consisted of three types: type I does not have side chains; type II has one β-glucose residue that branches off the disaccharide-phosphate repeat units and type III has up to three glucose residues (oligo-glucosylated). The significance of these modifications was investigated during in vivo interaction of L. infantum with Lutzomyia longipalpis, and in vitro interaction of the parasites and respective LPGs with murine macrophages. There were no consequential differences in the parasite densities in sand fly midguts infected with Leishmania strains exhibiting type I, II and III LPGs. However, higher nitric oxide production was observed in macrophages exposed to glucosylated type II LPG.  相似文献

An attempt has been undertaken to evaluate interrelations of gymnospermous and dicotyledonous plants on the basis of immunochemical studies of seed proteins. For this purpose, 12 antisera were raised to proteins of taxa representing four gymnosperm classes: Ginkgoopsida, Cycadopsida, Coniferopsida, and Gnetopsida. Seed proteins of eight dicotyledonous subclasses (after Takhtadzhyan, 1987) were used. The representatives of all dicotyledonous subclasses gave immunochemical reactions with those of all gymnospermous classes. The data obtained suggest the presence of sufficiently close immunochemical relations between gymnosperms and dicotyledons. Samples were found among the representatives of subclasses Dilleniidae, Hamamelididae, and Rosidae, which gave satisfactory reactions with eight to ten antisera to proteins of dicotyledonous seeds. Analysis of the data we obtained suggests that gymnospermous and dicotyledonous plants took their origin from a common pragymnospermous ancestor and later evolved independently or that dicotyledons separated from gymnosperms at an early stage of their evolution before divergence of the latter into several phyletic lineages.  相似文献
