Phototaxis was measured in young, middle-aged, and oldDrosophila melanogaster flies of both sexes. The apparatus allowed us to measure the tendency to go toward light, independently of the time needed to do so; under such conditions, phototaxis is dissociated from locomotor activity. The percentage of photopositive flies decreased slightly with age (93.96, 80.17, and 78.97%, respectively, in young, middle-aged, and old flies). Results are discussed in connection with previous data for which the tendency to go toward light and the time to do so were not dissociated.  相似文献

Background:

There has been growing interest in the overdiagnosis of breast cancer as a result of mammography screening. We report incidence rates in British Columbia before and after the initiation of population screening and provide estimates of overdiagnosis.

Methods:

We obtained the numbers of breast cancer diagnoses from the BC Cancer Registry and screening histories from the Screening Mammography Program of BC for women aged 30–89 years between 1970 and 2009. We calculated age-specific rates of invasive breast cancer and ductal carcinoma in situ. We compared these rates by age, calendar period and screening participation. We obtained 2 estimates of overdiagnosis from cumulative cancer rates among women between the ages of 40 and 89 years: the first estimate compared participants with nonparticipants; the second estimate compared observed and predicted population rates.

Results:

We calculated participation-based estimates of overdiagnosis to be 5.4% for invasive disease alone and 17.3% when ductal carcinoma in situ was included. The corresponding population-based estimates were −0.7% and 6.7%. Participants had higher rates of invasive cancer and ductal carcinoma in situ than nonparticipants but lower rates after screening stopped. Population incidence rates for invasive cancer increased after 1980; by 2009, they had returned to levels similar to those of the 1970s among women under 60 years of age but remained elevated among women 60–79 years old. Rates of ductal carcinoma in situ increased in all age groups.

Interpretation:

The extent of overdiagnosis of invasive cancer in our study population was modest and primarily occurred among women over the age of 60 years. However, overdiagnosis of ductal carcinoma in situ was elevated for all age groups. The estimation of overdiagnosis from observational data is complex and subject to many influences. The use of mammography screening in older women has an increased risk of overdiagnosis, which should be considered in screening decisions.There is growing interest in the overdiagnosis of breast cancer resulting from mammography screening.1,2 It has been suggested that incidence rates after the introduction of mammography screening are higher than would be expected from the early detection of clinically significant disease alone. The clinical importance of ductal carcinoma in situ has long been unclear, and recent attention has also focused on the potential overdiagnosis of invasive cancer.1,3,4 Furthermore, more frequent screening, wider age ranges and higher false-positive rates in much of North America may result in increased overdiagnosis5 in comparison with Europe, the source of most reports.The Screening Mammography Program of BC provides bilateral 2-view mammography screening to female residents of British Columbia. The program began in 1988 at a single location and progressively expanded by adding centres in larger communities and extending mobile services; by 2000, geographic coverage was effectively 100%. Women aged 40–79 years are eligible to self-refer. Since 1997, women aged 40–49 years are reminded to return annually, and those over 50 years of age are reminded to return biennially. Women outside of this age-range can undergo screening with a physician’s referral. Rates of abnormal results on screening mammography in British Columbia are comparable to the community rates seen in the United States.6Our primary objective for this study was to estimate the extent of overdiagnosis of breast cancer due to screening. To do this, we examined cancer rates in the following subpopulations: all women 1970–1979, and women in 2005–2009 classified by active and nonactive participation in screening. From 1970 to 1979, screening was infrequent and no organized program existed. In the period 2005–2009, screening was well established and the use of hormone replacement therapy for menopause had declined.7 Our secondary objective was to determine the changes in population incidence of breast cancer, both invasive and ductal carcinoma in situ, following the implementation of a population-based screening program. Rates of psychopathology are elevated in marginalized and unstably housed persons, underscoring the need for applicable clinical measures for these populations. The Positive and Negative Syndrome Scale (PANSS) is a clinical instrument principally developed for use in schizophrenia to identify the presence and severity of psychopathology symptoms. The current study investigates whether a reliable and valid PANSS factor structure emerges in a marginally housed, heterogeneous sample recruited from the Downtown Eastside of Vancouver where substance use disorders and psychiatric illness are pervasive. Participants (n = 270) underwent structured clinical assessments including the PANSS and then were randomly assigned to either exploratory (EFA) or confirmatory factor analytic (CFA) subsamples. EFA pointed to a novel three factor PANSS. This solution was supported by CFA. All retained items (28 out of 30) load significantly upon hypothesized factors and model goodness of fit analyses are in the acceptable to good range. Each of the three first-order factor constructs, labeled Psychosis/Disorganized, Negative Symptoms/Hostility, and Insight/Awareness, contributed significantly to measurement of a higher-order psychopathology construct. Further, the latent structure of this 3-factor solution appears temporally consistent over one-year. This PANSS factor structure appears valid and reliable for use in persons with multimorbidity, including substance use disorders. The structure is somewhat distinct from existing solutions likely due to the unique characteristics of this marginally housed sample.  相似文献

Background

Many RNA viruses arise from animal reservoirs, namely bats, rodents and insectivores but mechanisms of virus maintenance and transmission still need to be addressed. The bicolored white-toothed shrew (Crocidura leucodon) has recently been identified as reservoir of the neurotropic Borna disease virus 1 (BoDV-1).

Principal Findings

Six out of eleven wild living bicoloured white-toothed shrews were trapped and revealed to be naturally infected with BoDV-1. All shrews were monitored in captivity in a long-term study over a time period up to 600 days that differed between the individual shrews. Interestingly, all six animals showed an asymptomatic course of infection despite virus shedding via various routes indicating a highly adapted host-pathogen interaction. Infectious virus and viral RNA were demonstrated in saliva, urine, skin swabs, lacrimal fluid and faeces, both during the first 8 weeks of the investigation period and for long time shedding after more than 250 days in captivity.

Conclusions

The various ways of shedding ensure successful virus maintenance in the reservoir population but also transmission to accidental hosts such as horses and sheep. Naturally BoDV-1-infected living shrews serve as excellent tool to unravel host and pathogen factors responsible for persistent viral co-existence in reservoir species while maintaining their physiological integrity despite high viral load in many organ systems.  相似文献

Question: How can the U.S. National Vegetation Classification (USNVC) serve as an effective tool for classifying and mapping vegetation, and inform assessments and monitoring? Location: Voyageurs National Park, northern Minnesota, U.S.A and environs. The park contains 54 243 ha of terrestrial habitat in the sub-boreal region of North America. Methods: We classified and mapped the natural vegetation using the USNVC, with ‘alliance’and ‘association’as base units. We compiled 259 classification plots and 1251 accuracy assessment test plots. Both plot and type ordinations were used to analyse vegetation and environmental patterns. Color infrared aerial photography (1:15840 scale) was used for mapping. Polygons were manually drawn, then transferred into digital form. Classification and mapping products are stored in publicly available databases. Past fire and logging events were used to assess distribution of forest types. Results and Discussion: Ordination and cluster analyses confirmed 49 associations and 42 alliances, with three associations ranked as globally vulnerable to extirpation. Ordination provided a useful summary of vegetation and ecological gradients. Overall map accuracy was 82.4%. Pinus banksiana - Picea mariana forests were less frequent in areas unburned since the 1930s. Conclusion: The USNVC provides a consistent ecological tool for summarizing and mapping vegetation. The products provide a baseline for assessing forests and wetlands, including fire management. The standardized classification and map units provide local to continental perspectives on park resources through linkages to state, provincial, and national classifications in the U.S. and Canada, and to NatureServe's Ecological Systems classification.  相似文献

Seven generations of selection for high and low spontaneous locomotor activity were made in the wild-laboratory strain Oregon of Drosophila melanogaster. Great care was taken to select for activity and not for reactivity. In opposition with the non totally unambiguous results obtained by another author, absolutely no response to selection could be obtained. Thus the Oregon strain of Drosophila melanogaster does not appear to possess any additive genetic variance for spontaneous locomotor activity. Yet before taking for granted that that conclusion is applicable to all strains of Drosophila melanogaster an experimental selection should be performed again using a freshly captured wild strain.  相似文献

Empirical models used to predict thermocline depths of lakes have typically been based on physical and morpho-metric variables. However, lakes with appreciable levels of dissolved organic material, including those found on the Canadian Precambrian Shield (DOC levels 1.4-12.41 mg/l), have seldom been included in these models. Our analysis suggests that for such lakes, thermocline depth is linked strongly to light penetration (Secchi depth r = 0.83, light extinction r = 0.85) which is strongly related to DOC concentration (Secchi depth r = 0.91, light extinction r = 0.97). A multivariate regression based on small Canadian Shield lakes suggests that DOC is the most important predictor of thermocline depth. Maximum effective length, maximum depth, and chlorophyll a contribute significantly to the prediction power of the regression model, but are of secondary importance in the presence of DOC.
