Many amphibian population declines have been associated with the introduction of alien aquatic predators. Here, we explore the vulnerability of tadpoles of two sympatric Japanese species [Pelophylax nigromaculatus (PN) and Rhacophorus schlegelii (RS)] to the invasive crayfish Procambarus clarkii. We first examined the behavioral responses of the tadpoles to the cues of caged, fed crayfish predator in a controlled laboratory experiment, and subsequently tested their survival when together in the presence of free-ranging predator in outdoor mesocosms that simulated natural ponds. Only PN reduced activity level to the cues of the predator, but this apparent behavioral defense recorded in the laboratory did not result into higher survival in outdoor mesocosms. In mesocosms, PN exhibited higher biomass increment but experienced higher mortality in predator environments. The mechanism mediating mortality remains unclear though. These results indicate that sympatric prey may differentially respond and be disproportionally vulnerable to novel predators. Our study illustrates the possible contribution of a life-history trait influencing risk of predation in newly invaded systems.  相似文献

Pinus are among the highly invasive species that have spread outside their plantation area after their introduction in the Southern Hemisphere. The case of Pinus kesiya invasion is observed in the high plateau of Madagascar, inside the sclerophyll Tapia woodland which is dominated by the endemic Uapaca bojeri tree species. The analysis of this invasion was carried out using 375 plots of 100 m2 each in Tapia woodland. Data on the vegetation structure, the plot characteristics and the propagule pressure were collected. We recorded a total of 740 pines distributed in 29.8% of the plots. The generalized linear model built on P. kesiya at the three different life stages allowed us to highlight a different explicative variable on the species’ presence and abundance separately. The factors explaining pine occurrence varied according to the pine life stage. In the seedling stage, the distance of the plot from the propagule source combined with the longitudinal position of the plot explained 18% of the pine presence. In the sapling and adult stages, the vegetation structure was the main important factor (22% and 11% of variation explained regarding presence and abundance). The frequency of U. bojeri and the degree of disturbance were the most important factors characterizing this vegetation structure. Based on these results, a strategy to control pine invasion in the Tapia woodland may focus on enrichment with U. bojeri and limitation of the plantation of P. kesiya in proximity.

Background and Aims

The oriental forest ecosystem in Madagascar has been seriously impacted by fragmentation. The pattern of genetic diversity was analysed on a tree species, Dalbergia monticola, which plays an important economic role in Madagascar and is one of the many endangered tree species in the eastern forest.

Methods

Leaves from 546 individuals belonging to 18 small populations affected by different levels of fragmentation were genotyped using eight nuclear (nuc) and three chloroplast (cp) microsatellite markers.

Key Results

For nuclear microsatellites, allelic richness (R) and heterozygosity (He,nuc) differed between types of forest: R = 7·36 and R = 9·55, He,nuc = 0·64 and He,nuc = 0·80 in fragmented and non-fragmented forest, respectively, but the differences were not significant. Only the mean number of alleles (Na,nuc) and the fixation index FIS differed significantly: Na,nuc = 9·41 and Na,nuc = 13·18, FIS = 0·06 and FIS = 0·15 in fragmented and non-fragmented forests, respectively. For chloroplast microsatellites, estimated genetic diversity was higher in non-fragmented forest, but the difference was not significant. No recent bottleneck effect was detected for either population. Overall differentiation was low for nuclear microsatellites (FST,nuc = 0·08) and moderate for chloroplast microsatellites (FST,cp = 0·49). A clear relationship was observed between genetic and geographic distance (r = 0·42 P < 0·01 and r = 0·42 P = 0·03 for nuclear and chloroplast microsatellites, respectively), suggesting a pattern of isolation by distance. Analysis of population structure using the neighbor-joining method or Bayesian models separated southern populations from central and northern populations with nuclear microsatellites, and grouped the population according to regions with chloroplast microsatellites, but did not separate the fragmented populations.

Conclusions

Residual diversity and genetic structure of populations of D. monticola in Madagascar suggest a limited impact of fragmentation on molecular genetic parameters.  相似文献

Editing is a general event in plant mitochondrial messenger RNAs, but has never been detected in a plant mitochondrial transfer RNA (tRNA). We demonstrate here the occurrence of a tRNA editing event in higher plant mitochondria: in both bean and potato, the C encoded at position 4 in the mitochondrial tRNA(Phe)(GAA) gene is converted into a U in the mature tRNA. This nucleotide change corrects the mismatched C4-A69 base-pair which appears when folding the gene sequence into the cloverleaf structure and it is consistent with the fact that C to U transitions constitute the common editing events affecting plant mitochondrial messenger RNAs. The tRNA(Phe)(GAA) gene is located upstream of the single copy tRNA(Pro)(UGG) gene in both the potato and the bean mitochondrial DNAs. The sequences of potato and bean tRNA(Pro)(UGG) genes are colinear with the sequence of the mature bean mitochondrial tRNA(Pro)(UGG), demonstrating that this tRNA is not edited. A single copy tRNA(Ser)(GCU) gene was found upstream of the tRNA(Phe) gene in the potato mitochondrial DNA. A U6-U67 mismatched base-pair appears in the cloverleaf folding of this gene and is maintained in the mature potato mitochondrial tRNA(Ser)(GCU), which argues in favour of the hypothesis that the editing system of plant mitochondria can only perform C to U or occasionally U to C changes.  相似文献

Introduction

Tuberculosis infection may remain latent, but the disease is nevertheless a serious public health issue. Various epidemiological studies on pulmonary tuberculosis have considered the spatial component and taken it into account, revealing the tendency of this disease to cluster in particular locations. The aim was to assess the contribution of Knowledge Attitude and Practice (KAP) to the distribution of tuberculosis and to provide information for the improvement of the National Tuberculosis Program.

Methods

We investigated the role of KAP to distribution patterns of pulmonary tuberculosis in Antananarivo. First, we performed spatial scanning of tuberculosis aggregation among permanent cases resident in Antananarivo Urban Township using the Kulldorff method, and then we carried out a quantitative study on KAP, involving TB patients. The KAP study in the population was based on qualitative methods with focus groups.

Results

The disease still clusters in the same districts identified in the previous study. The principal cluster covered 22 neighborhoods. Most of them are part of the first district. A secondary cluster was found, involving 18 neighborhoods in the sixth district and two neighborhoods in the fifth. The relative risk was respectively 1.7 (p<10−6) in the principal cluster and 1.6 (p<10−3) in the secondary cluster. Our study showed that more was known about TB symptoms than about the duration of the disease or free treatment. Knowledge about TB was limited to that acquired at school or from relatives with TB. The attitude and practices of patients and the population in general indicated that there is still a stigma attached to tuberculosis.

Conclusion

This type of survey can be conducted in remote zones where the tuberculosis-related KAP of the TB patients and the general population is less known or not documented; the findings could be used to adapt control measures to the local particularities.  相似文献

In forest tree genetic improvement, multi-trait genomic selection (GS) may have advantages in improving the accuracy of the genotype estimation and shortening selection cycles. For the breeding of Eucalyptus robusta, one of the most exotic planted species in Madagascar, volume at 49 months (V49), total lignin (TL), and holo-cellulose (Holo) were considered. For GS, 2919 single nucleotide polymorphisms (SNP) were used with the genomic best linear unbiased predictor (GBLUP) method, which was as efficient as the reproducing kernel Hilbert space (RKHS) and elastic net methods (EN), but more adapted to multi-trait modeling. The efficiency of individual I model, including the genomic data, was much higher than the provenance effect P model. For example, with V49, mean goodness-of-fit was: rI_Full =?0.79, rP_Full =?0.37 for I and P, respectively. The prediction accuracies using the cross-validation procedure were lower for V49: rI =?0.29 rP =?0.28. The genetic gains resulting from the indexes associating (V49, TL) and (V49, Holo) were higher using I than for the P model; for V49, the relative genetic gain was 37 and 20%, respectively, with 5% of selection intensity. The single-trait approach was as efficient as the multi-trait approach given the weak correlations between V49 and TL or Holo. The I model also brings greater diversity: for V49 the number of provenances represented in a selected population was two and three with the P model, and 6 and 16 with the I model.  相似文献

The assessment of liver lipid content and composition is needed in preclinical research to investigate steatosis and steatosis-related disorders. The purpose of this study was to quantify in vivo hepatic fatty acid content and composition using a method based on short echo time proton magnetic resonance spectroscopy (MRS) at 7 Tesla. A mouse model of glycogen storage disease type 1a with inducible liver-specific deletion of the glucose-6-phosphatase gene (L-G6pc−/−) mice and control mice were fed a standard diet or a high-fat/high-sucrose (HF/HS) diet for 9 months. In control mice, hepatic lipid content was found significantly higher with the HF/HS diet than with the standard diet. As expected, hepatic lipid content was already elevated in L-G6pc−/− mice fed a standard diet compared with control mice. L-G6pc−/− mice rapidly developed steatosis which was not modified by the HF/HS diet. On the standard diet, estimated amplitudes from olefinic protons were found significantly higher in L-G6pc−/− mice compared with that in control mice. L-G6pc−/− mice showed no noticeable polyunsaturation from diallylic protons. Total unsaturated fatty acid indexes measured by gas chromatography were in agreement with MRS measurements. These results showed the great potential of high magnetic field MRS to follow the diet impact and lipid alterations in mouse liver.  相似文献

Scattered populations of the same tree species in montane forests through Africa have led to speculations on the origins of distributions. Here, we inferred the colonization history of the Afromontane tree Prunus africana using seven chloroplast DNA loci to study 582 individuals from 32 populations sampled in a range-wide survey from across Africa, revealing 22 haplotypes. The predominant haplotype, HT1a, occurred in 13 populations of eastern and southern Africa, while a second common haplotype, HT1m, occurred in populations of western Uganda and western Africa. The high differentiation observed between populations in East Africa was unexpected, with stands in western Uganda belonging with the western African lineage. High genetic differentiation among populations revealed using ordered alleles (N(ST) = 0.840) compared with unordered alleles (G(ST) = 0.735), indicated a clear phylogeographic pattern. Bayesian coalescence modelling suggested that 'east' and 'west' African types likely split early during southward migration of the species, while further more recent splitting events occurred among populations in the East of the continent. The high genetic similarity found between western Uganda and west African populations indicates that a former Afromontane migration corridor may have existed through Equatorial Africa.  相似文献
