The reaction between the amino group and the carbonyl group is important in food quality control. Furthermore, advanced glycation end products from foods are considered to relate to aging and diabetes. Thus, it is important to control this reaction. In this study, we investigated the effects of salt concentration on the rates of browning reaction of amino acid, peptides, and proteins. A high concentration of sodium chloride retarded the reaction rate of Glc with amino acids as measured with the absorbance at 470 nm, but did not change the browning rate of Glc with peptides. On the other hand, sodium chloride retarded the browning reaction rate of proteins as measured with polymerization degree or by the loss of Lys. It is hoped that the results of this study will be applied in the control of amino-carbonyl reaction rates in the food industry.  相似文献

Simulation of X- and Q-band electron paramagnetic resonance (EPR) spectra of an unsymmetric dinuclear [Mn(2)(II,III)L(mu-OAc)(2)]ClO(4) complex (1), (L is the dianion of 2-{[N,N-bis(2-pyridylmethyl)amino]methyl}-6-{[N-(3,5-di-tert-butyl-2-hydroxybenzyl)-N-(2-pyridylmethyl)amino]methyl}-4-methylphenol) was performed using one consistent set of simulation parameters. Rhombic g-tensors and hyperfine tensors were necessary to obtain satisfactory simulation of the EPR spectra. The anisotropy of the effective hyperfine tensors of each individual (55)Mn ion was further analyzed in terms of intrinsic hyperfine tensors. Detailed analysis shows that the hyperfine anisotropy of the Mn(III) ion is a result of the Jahn-Teller effect and thus an inherent character. In contrast, the anomalous hyperfine anisotropy of the Mn(II) ion is attributed as being transferred from the Mn(III) ion through the spin exchange interaction. The anisotropy parameter for the Mn(II) is deduced as D(II)=-1.26+/-0.2cm(-1). This is the first reported D(II) value for a Mn(II) ion in a weakly exchange coupled mixed-valence Mn(2)(II,III) complex with a bis-mu-acetato-bridge. The [see text] electronic configuration of the Mn(III) ion in 1 is revealed by the negative sign of its intrinsic hyperfine tensor anisotropy, Deltaa(III)=a(z)-a(x,y)=-46cm(-1). Lower spectral resolution of the Q-band EPR spectrum as compared to the X-band EPR spectrum is associated to large line width broadening of the x- and y-components in contrast to the z-component. The origins of the unequal distribution of line width between the z- and x-, y-components are discussed.  相似文献

BackgroundVisceral leishmaniasis (VL) is a multifactorial disease, where the host genetics play a significant role in determining the disease outcome. The immunological role of anti-inflammatory cytokine, Interleukin 10 (IL10), has been well-documented in parasite infections and considered as a key regulatory cytokine for VL. Although VL patients in India display high level of IL10 in blood serum, no genetic study has been conducted to assess the VL susceptibility / resistance. Therefore, the aim of this study is to investigate the role of IL10 variations in Indian VL; and to estimate the distribution of disease associated allele in diverse Indian populations.MethodologyAll the exons and exon-intron boundaries of IL10 were sequenced in 184 VL patients along with 172 ethnically matched controls from VL endemic region of India.

Result and Discussion

Our analysis revealed four variations; rs1518111 (2195 A>G, intron), rs1554286 (2607 C>T, intron), rs3024496 (4976 T>C, 3’ UTR) and rs3024498 (5311 A>G, 3’ UTR). Of these, a variant g.5311A is significantly associated with VL (χ2=18.87; p =0.00001). In silico approaches have shown that a putative micro RNA binding site (miR-4321) is lost in rs3024498 mRNA. Further, analysis of the above four variations in 1138 individuals from 34 ethnic populations, representing different social and linguistic groups who are inhabited in different geographical regions of India, showed variable frequency. Interestingly, we have found, majority of the tribal populations have low frequency of VL (‘A’ of rs3024498); and high frequency of leprosy (‘T’ of rs1554286), and Behcet’s (‘A’ of rs1518111) associated alleles, whereas these were vice versa in castes. Our findings suggest that majority of tribal populations of India carry the protected / less severe allele against VL, while risk / more severe allele for leprosy and Behcet’s disease. This study has potential implications in counseling and management of VL and other infectious diseases.  相似文献

Limited proteolysis of buffalo plasma fibronectin (FN) by thermolysin yielded four gelatin-binding fragments of which, the major 59 kDa fragment, GBF1, was isolated by gelatin-Sepharose and heparin-Sepharose affinity columns. GBF1 appeared during early phase of thermolysin digestion and remained intact even after 4 hr of digestion. GBF1 may be similar to 56 kDa gelatin-binding fragment of FNs from human and hamster plasma. But, it is more resistant to thermolysin cleavage. The fragment binds to heparin with low affinity. On the basis of the structure of human plasma FN, the modular structure of GBF1 may be given as: 6Fn1 1Fn2 2Fn2 7Fn1 8Fn1 9Fn1 1Fn3. Biophysical properties of GBF1 suggest an expanded native conformation. The interaction of the fragment with gelatin is pH-dependent and independent of NaCl concentration.  相似文献

The current knowledge on biological protein acetylation is confined to acetyl CoA-dependent acetylation of protein catalyzed by specific acetyl transferases and the non-enzymatic acetylation of protein by acetylated xenobiotics such as aspirin. We have discovered a membrane-bound enzyme catalyzing the transfer of acetyl groups from the acetyl donor 7,8-diacetoxy-4-methyl coumarin (DAMC) to glutathione S-transferase 3-3 (GST3-3), termed DAMC:protein transacetylase (TAase). The purified enzyme was incubated with recombinant GST3-3 subunit and DAMC, the modified protein was isolated by sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE) in gel digested with trypsin and the tryptic digest was analyzed by mass spectrometry. The N-terminus and six lysines, Lys-51, -82, -124, -181, -191 and -210, were found to be acetylated. The acetylation of GST3-3 described above was not observed in the absence of either DAMC or TAase. These results clearly establish the phenomenon of protein acetylation independent of acetyl CoA catalyzed by a hitherto unknown enzyme (TAase) utilizing a certain xenobiotic acetate (DAMC) as the active acetyl donor.  相似文献

Biomass utilization has received much attention for production of high density solid fuels. Utilization of cheap and naturally available precursors through environmentally friendly and effective processes is an attractive and emerging research area. Pyrolysis and hydrothermal carbonization (HTC) are well-known technologies available for production of solid biofuel using conventional or microwave heating. Microwave heating is a simpler and more efficient heating method than conventional heating. This study presents a critical review on microwave pyrolysis and microwave HTC for solid fuel production in terms of yield and quality of products. Moreover, a brief summary of parameters of microwave pyrolysis and microwave HTC are discussed. The fuel, chemical, structural and thermal weight loss characteristics of solid fuels produced from different biomass are discussed and compared.  相似文献
