Upregulation of small heat-shock proteins (sHsps) in response to cellular stress is one mechanism to increase cell viability. We previously described that cultured rat hippocampal neurons express five of the 11 family members but only upregulate two of them (HspB1 and HspB5) at the protein level after heat stress. Since neurons have to cope with many other pathological conditions, we investigated in this study the expression of all five expressed sHsps on mRNA and protein level after sublethal sodium arsenite and oxidative and hyperosmotic stress. Under all three conditions, HspB1, HspB5, HspB6, and HspB8 but not HspB11 were consistently upregulated but showed differences in the time course of upregulation. The increase of sHsps always occurred earlier on mRNA level compared with protein levels. We conclude from our data that these four upregulated sHsps (HspB1, HspB5, HspB6, HspB8) act together in different proportions in the protection of neurons from various stress conditions.  相似文献

Antigenic similarities between Plasmodium and Babesia parasites of the phylum Apicomplexa have been previously demonstrated primarily by the serological cross reactivity observed in the indirect fluorescent antibody (IFA) test. We have now studied the antigenic relationship between the human malaria parasite, Plasmodium falciparum, and the hemoparasitic agent of cattle, Babesia bovis, using rabbit monospecific antibodies produced against individual culture-derived P. falciparum polypeptides and bovine polyspecific antibodies to B. bovis exoantigens. These respective antibodies were found to be distinctly cross reactive in the IFA test using infected erythrocytes (squirrel monkey--P. falciparum; bovine--B. bovis) as antigen substrates. Immunofluorescence was shown to be highly specific for parasite surfaces. Additionally, the degree of reactivity with soluble exoantigens contained in Plasmodium and Babesia culture supernatants was monitored by a two-site enzyme immunoassay employing the cross-reactive antibodies. Further evidence for antigenic cross reactivity between P. falciparum and B. bovis parasites was shown with the in vitro inhibition assay. Antibodies to P. falciparum and B. bovis were found to be highly inhibitory for the in vitro growth of P. falciparum in human erythrocytes.  相似文献

The in vitro and ex vivo effects of antidepressant drugs on membrane-bound phosphatidylinositol (PI) synthetase and PI: myo-inositol exchange enzyme activities were examined. In rat brain subcellular fractions, PI synthetase occurred exclusively in the microsomes. In comparison, the activity of CDP-diglyceride independent PI: myo-inositol exchange enzyme was low (3%). Of the various CDP-diglycerides tested for the activation of PI synthetase, CDP-dipalmitin was the most active. Addition of 1 mM of desipramine, amitriptyline, imipramine, iprindole, clomipramine and mianserin in vitro significantly inhibited (30–60%) PI synthetase activity, whereas the same concentration of zimelidine and fluoxetine had no effect. At low liponucleotide concentrations, PI synthetase activity was significantly enhanced by imipramine (1 mM), whereas the enzyme activity was inhibited at higher liponucleotide concentrations (>0.3 mM). In contrast, imipramine had no effect on the PI: myo-inositol exchange enzyme activity. No significant alteration in the PI synthetase activity was found following either acute (2 h) or chronic (21 d) treatment of rats with imipramine. The above results indicate that the de novo synthesis of PI is inhibited in vitro but not ex vivo by some antidepressant drugs. However, in view of the high concentration of the drugs required, the pharmacological significance of this inhibitory action with respect to their therapeutic effects is doubtful.  相似文献

Synthesis of heat shock proteins (HSPs) in the leaves of a drought- and heat-resistant (line ZPBL 1304), and a drought- and heat-sensitive (line ZPL 389) line of maize (Zea mays L.) was studied under two environmental stress treatments: (a) soil drying and high temperature and (b) high temperature. In the first treatment 13-day-old plants were exposed to 7-day soil drying followed by high temperature stress (45°C), and in the second treatment 20-day-old plants were exposed to high temperature stress (45°C). Second leaves were labeled with [35S]methionine. During the labeling period line ZPBL 1304 showed no signs of leaf dehydration under soil drying and high temperature stress conditions. In contrast, line ZPL 389 was dehydrated 23%, as determined by relative water content. Incorporation of [35S]methionine into protein was greater in the resistant than in the sensitive line in both treatments. The pattern of synthesis of HSPs in the two lines was similar in treatments 1 and 2. Both lines synthesized a high molecular mass set and a low molecular mass set of HSPs. Proteins from both sets from both lines of maize appeared similar to each other, with respect to the molecular mass. Heated plants of the drought- and heat-resistant line ZPBL 1304 synthesized a band of HSP(s) of approximately 45 kilodaltons which was not found in heated plants of the drought and heat sensitive line ZPL 389. This is the first report on qualitative intraspecific difference in the synthesis of HSPs in maize.  相似文献

Conclusion This outline of the instrumentality of the technology/technique of representation in postcommunist Serbia is but a brief attempt to address the modes of production of identity in postmodernism, its legitimation in/of dominant discourse, and their embedding in common sense language and practice. Postcommunist Serbia is obviously a special situation:23 a particular case of the social appropriation and construction of enjoyment and desire, intimately related to the desires and various enjoyments of the members of the community, precarious in its collective affirmation. Based on the idea of contingent collectivity, faction-fiction, in its meta-critique seems to signal the emergence of a new régime in the relation between historical reality and its representation in historical narratives.This new régime, obviously, cannot be exhaustively (or even extensively) discussed in a brief paper like this one. Nevertheless, some (three) points of departure for the general discussion of postmodern modes of representation are still possible. One such point of departure is the contingence of representation, another the apparent paradigmatic similarity between trash talk-shows and Echoes and Reactions. Summary We studied cell ultrastructure and carbohydrate levels in the leaf tissue ofArabidopsis thaliana L. (Heyn) cv. Columbia during rapid cold acclimation. Freezing tolerance of the leaves from 26 day old plants was determined after 48 h and 10 days at 4°C. Acclimation treatment of 48 h decreased the lethal freezing temperature from –5.7°C to –9.4°C. Freezing tolerance was not altered further by acclimation at 4 °C for 10 days. Ultrastructural changes in the parenchyma cells were evident after 6 to 24 h of cold acclimation. The plasma membrane showed signs of extensive turnover. Evidence of membrane invaginations and sequestering of membrane material was observed. In addition, numerous microvesicles, paramural bodies, and fragments of endoplasmic reticulum were noticed in the vicinity of plasma membrane. Modifications in the structure of cell membranes were evident after 5 days of exposure to low temperature. Small, darkly stained globules were seen on the plasma membrane, tonoplast, chloroplast envelope membrane, mitochondrion outer membrane, dictyosome cisternae membrane, and microvesicle membrane. As far as we are aware, this type of membrane modification has not been described previously in plant cells exposed to low temperature. We propose to call these structures membraglobuli. Acclimation treatment also increased the concentrations of soluble sugars and starch. These observations suggest that cold acclimation inA. thaliana induces changes in both plasma membrane properties and carbohydrate composition.  相似文献
