In contrast with hematopoietic cells and fibroblasts, which express mainly one form of protein tyrosine phosphatase (PTP) SHP-1 or SHP-2, epithelial cells like A431, HeLa, and 293 express both forms of PTP. These two PTP regulate NFkappaB activity differently; SHP-1 inhibits and SHP-2 stimulates NFkappaB activation. In epithelial cells the process of NFkappaB activation depends on the combination of two PTP activities. The activity of PTP SHP-1 dominates in this tandem according to our data. The signal regulatory protein (SIRPalpha) is the adapter and the substrate of PTP SHP-1 and SHP-2. We investigated the role of SIRPalpha and its dominant negative mutant in PTP activities in 293 cells. The overexpression of wild-type SIRPalpha suppresses the activities of both PTP, but has a stronger effect on PTP SHP-2, especially when this protein is overexpressed in 293 cells. In contrast with wild-type SIRPalpha, its dominant negative mutant acts predominantly against PTP SHP-1, and can be detected in the complex with PTP SHP-1. The expression of dominant negative mutant of SIRPalpha has an effect similar to the expression of dominant negative PTP SHP-1 in the process of NFkappaB activation.  相似文献

With the help of scanning and transmission electron microscopy, the morphology of spermatozoa of Icelus spiniger and egg envelopes of I. spiniger and I. spatula has been studied. Spermatozoa have an elongated head flattened on one side, an insignificant centriolar fossa, and numerous mitochondria in the middle section. The axoneme has a structure described by the formula 9 + 2. The envelopes of oocytes of both species have a follicular membrane, chorion, and zona radiata.  相似文献

Accessory cells were studied in early spermatogenesis of flatfishes Glyptocephalus stelleri and Pleuronectes pinnifasciatus using transmission electron microscopy. The morphological organization of accessory cells in G. stelleri was similar to that of Sertoli cells. The relationships between germinal bodies and mitochondria were studied in the holothurian Apostichopus japonicus and the flounder Pleuronectes asper using TEM. In the gonial cells of both species the mitochondria are arranged around germinal bodies and are in contact with the latter. A gradual disappearance of the outer membrane is found in the mitochondria that interact with the germinal substance. Later on, dispersion of the globules of the mitochondrial matrix containing mitochondrial cristae occurs. It is supposed that the substance of the mitochondrial matrix takes part in the development and functioning of the germinal plasm in both invertebrates and vertebrates.  相似文献

Spermatogenesis in silver salmon Oncorhynchus kisutch and cherry salmon O. masou was studied by transmission electron microscopy. The spermatogenic cells of both species have been shown to have no considerable differences in their dimensional and ultrastructural parameters. The characteristic peculiarity of spermatogonia was the presence of a germ determinant substance, which is the cytoplasmic marker of sex line cells. The primary and secondary spermatocytes, as well as early spermatids, were arranged in gonads as cell clusters at the same stage of development. In spermiogenesis, the regular appearance of the electron-lucent vesicle was observed, which forms a structural complex with the apical condensation of the nuclear membrane. This complex is morphologically similar to the not-yet-formed spermatid acrosomes in many Metazoa and is referred to by the authors as an acrosome-like structure (AS). At the final stage of spermiogenesis, the disappearance of an AS allows one to considering it to be a temporarily existing structure that recapitulates the ancestral morphotype of salmonid sperm. Spermatozoa of the both species are typical, primitive, acrosome-lacking cells, which are characteristic of many representatives of Teleostei that have external insemination.  相似文献

The process of the destruction of nonreleased spermatozoa was studied using TEM in the postspawning gonads of the chum salmon Oncorhynchus keta and the pointhead flounder Cleisthenes herzensteini, as representatives of monocyclic and polycyclic fishes respectively. It was shown that in both species residual sperm cells are destroyed through fragmentation of chromatin and the entire cell. Flavonoid quercetin and its derivative, methylquercetin, inhibit the replication of poliovirus in several cell lines. Here, we show that replication of poliovirus is inhibited by quercetin and that the extent of this inhibition depends on the intracellular content of pirin, a quercetinase. HeLa cells contain higher content of pirin protein than normal kidney human epithelial (NKE) or 293 cells do. Poliovirus replication in HeLa cells is significantly more resistant to quercetin than its replication in NKE and 293 cells. Overexpression of pirin reduced antiviral inhibitory effect of quercetin, while siRNA-induced suppression of pirin level made poliovirus replication more sensitive to the flavonoid. The results suggest that quercetinase activity of pirin determines the resistance of poliovirus infection to quercetin.  相似文献

Ultrastructural aspects of sperm destruction patterns were offered as additional cytological parameters for evaluation of the genus affinity of flatfishes Hippoglossoides dubius and Cleisthenes herzensteini. At the beginning of spermatozoan destruction, cell membrane in both species was found swollen, besides, discontinuity of membranes was observed, and membraneous parts were seen separating from sparmatozoa. We observed the ability of separated membraneous parts to aggregate to twisting conglomerates that wind round the objects of destruction. In H. dubius the membraneous conglomerates wound round spermatozoa, and after that such spermatozoa were phagocyted by follicular cells. In C. herzensteini, the membraneous conglomerates grasped the particles of destructed spermatozoa: the formed residual bodies were collected in the gonad lumen but not phagocyted by follicular cells. The expressiveness of the differences found in the pattern of sperm destruction is so considerable that, in the authors' opinion, these data are to supplement a list of criteria making reasonable reconsideration of the taxonomic status of C. herzensteini: its belonging to the genus Hippoglossoides, and establishing of the genus Cleisthenes as an independent rank.  相似文献
