Careful review of the literature demonstrates conflicting results concerning the ability of the deafferented medial basal hypothalamus to support gonadotropin release in the rat and thus one may question the existence of LH-RH neurons in the medial basal hypothalamus. The direct search for the LH-RH perikarya in the rat hypothalamus has not settled the question of whether these releasing hormone neurons are located in the medial basal hypothalamus. Most investigators do agree that following complete hypothalamic deafferentation there is a reduction of the immunoassayable LH-RH in the medial basal hypothalamus; however, these results do not necessarily prove that LH-RH originates outside the hypothalamus. It is argued that the completely deafferented medial basal hypothalamus may be so altered by the deafferentation procedure that it may be inadequate as a means to study neuroendocrine function. 相似文献

Visual species identification of cetacean strandings is difficult, especially when dead specimens are degraded and/or species are morphologically similar. The two recognised pilot whale species (Globicephala melas and Globicephala macrorhynchus) are sympatric in the North Atlantic Ocean. These species are very similar in external appearance and their morphometric characteristics partially overlap; thus visual identification is not always reliable. Genetic species identification ensures correct identification of specimens. Here we have employed one mitochondrial (D-Loop region) and eight nuclear loci (microsatellites) as genetic markers to identify six stranded pilot whales found in Galicia (Northwest Spain), one of them of ambiguous phenotype. DNA analyses yielded positive amplification of all loci and enabled species identification. Nuclear microsatellite DNA genotypes revealed mixed ancestry for one individual, identified as a post-F1 interspecific hybrid employing two different Bayesian methods. From the mitochondrial sequence the maternal species was Globicephala melas. This is the first hybrid documented between Globicephala melas and G. macrorhynchus, and the first post-F1 hybrid genetically identified between cetaceans, revealing interspecific genetic introgression in marine mammals. The growth inhibitory activity in conditioned medium of African green monkey kidney epithelial (BSC-1) cells that has been shown to arise, at least in part, from transforming growth factor beta 2 (TGF-beta 2) [Hanks, S. K., Armour, R., Baldwin, J. H., Maldonado, F., Spiess, J., & Holley, R. W. (1988) Proc. Natl. Acad. Sci. U.S.A. 85, 79-82] was tested for growth inhibitory activity prior to and following acidification. Similar to TGF-beta 1 from human platelets, the inhibitory activity from BSC-1 cells demonstrated an 8-10-fold stimulation following acidification, showing that the activity was secreted from the cells in latent form. Conditioned medium from BSC-1 cells was collected, acidified, and fractionated by procedures that separate TGF-beta 1 and -2. Biological activity was assayed by using the BSC-1 cell proliferation assay. Two active proteins with properties similar to known TGF-beta 1 and TGF-beta 2 were identified. Identity was confirmed by using immunological and amino acid sequencing techniques. These results were consistent with Northern blot analysis of total BSC-1 RNA, using cDNA probes for TGF-beta 1 and TGF-beta 2, which demonstrated strong signals for both mRNAs. Metabolic labeling in conjunction with two-dimensional gel electrophoresis revealed that the cells secrete approximately 10% TGF-beta 1 and 90% TGF-beta 2. 相似文献

An enzyme system prepared from maize chloroplasts catalyzes the synthesis of DNA from maize chloroplast DNA sequences cloned in bacterial plasmids. Cloned maize chloroplast DNA fragments Bam HI 17' (2470 bp) and Eco RI x (1368 bp) have been shown to be preferred templates for in vitro DNA synthesis catalyzed by pea chloroplast DNA polymerase preparations [Gold et al. (1987) Proc. Natl. Acad. Sci. USA 84, 194-198]. Analysis of replicative intermediates indicates that although the template activity of the recombinant plasmid pZmcBam 17' is substantially greater than that of the pZmcEco x, replication in both cases originates from within a 455 bp region which overlaps the two plasmids. The remaining approximately 1500 basepair portion of maize chloroplast BamHI fragment 17' is not more active because it contains additional origins for replication. The overlapping region shows sequence homology with a portion of the Chlamydomonas reinhardtii chloroplast chromosome that contains a replication origin. Replication is shown to proceed bidirectionally within the 455 bp origin region. Recombinant plasmid pZmc 427, which is also active in the in vitro DNA synthesis assay, promoted localized replication initiation within a 1 kbp Bg1II-Eco RI fragment of the chloroplast DNA insert, a region that includes the 3' terminal part of the psbA gene. 相似文献

Daily s.c. injection of (-)deprenyl (2.0 mg/kg/day) for three weeks in young male rats caused a threefold increase in superoxide dismutase (SOD) activity in the striatum of the brain compared with the value in saline-injected control rats. Furthermore, the activity of catalase (but not of glutathione peroxidase) was also increased significantly by deprenyl treatment. The results confirmed the previous findings of Knoll on SOD activity and furthermore provided evidence that the activity of catalase is also significantly induced by the drug, which was not found in the previous study. 相似文献

Two unicellular marine algae cultured in media containing sodium selenite were examined for glutathione peroxidase activity. The 400 g supernatant from disrupted cells of both the green alga Dunaliella primolecta and the red alga Porphyridium cruentum were able to enhance both the H2O2 and the tert-butyl hydroperoxide dependent oxidation of glutathione. The glutathione peroxidation activity of D. primolecta was reduced only slightly by heating the 400 g supernatant, a 30% decrease in the rate with H2O2 and 10% decrease in the rate with t-BuOOH being observed. Heating caused the H2O2 dependent activity in P. cruentum to be reduced by only 30%, but the activity with t-BuOOH was reduced by 90%. Freezing decreased the t-BuOOH dependent activity of P. cruentum by 90%, but did not lower the t-BuOOH dependent activity of D. primolecta or the H2O2 dependent activity of either alga. It was concluded that the heat and cold stable, glutathione peroxidation was non-enzymatic in nature. A variety of small molecules (ascorbate, Cu(NO3)2, selenocystine, dimethyldiselenide and selenomethionine) were shown to be able to enhance the hydroperoxide dependent oxidation of glutathione in the assay system employed in this study. Such compounds could be responsible for the activity observed in algae. The heat and cold labile t-BuOOH reductase activity of P. cruentumwas possibly enzymatic, but was not attributable to the presence of glutathione-S-transferase. Both algae, when cultured in the presence of added selenite, displayed an approximate doubling of the non-enzymatic H2O2 and t-BuOOH dependent glutathione oxidase activities. The heat and cold labile t-BuOOH reductase activity of P. cruentum was unaltered when the alga was grown in the presence of added selenite. These observations are consistent with the hypothesis that selenium compounds present in the algae are responsible for the selenium induced glutathione peroxidation. 相似文献

Chemical modification of Rhodospirillum rubrum chromatophores by 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) results in inactivation of photophosphorylation, Mg2+-ATPase, oxidative phosphorylation and ATP-driven transhydrogenase, with apparent first-order kinetics. Other energy-linked reactions such as light-driven transhydrogenase and light-dependent proton uptake were insensitive to NBD-Cl. The Ca2+-ATPase activity of the soluble coupling factor from chromatophores (R. rubrum F1) was inactivated by NBD-Cl with kinetics resembling those described for Mg2+-ATPase and photophosphorylation activities of chromatophores. Both NBD-chromatophores and NBD-R. rubrum F1 fully recovered their activities when subjected to thiolysis by dithioerythritol. Phosphoryl transfer reactions of chromatophores and Ca2+-ATPase activity of R. rubrum F1 were fully protected by 5 mM Pi against modification by NBD-Cl. ADP or ATP afforded partial protection. Analysis of the protection of Ca2+-ATPase activity by Pi indicated that NBD-Cl and Pi are mutually exclusive ligands. Spectroscopic studies revealed that tyrosine and sulfhydryl residues in R. rubrum F1 underwent modification by NBD-Cl. Certain neuropeptides are known to cause a hypotensive response, thought to be due to mast cell degranulation. The effects of five antagonists of luteinizing hormone-releasing hormone on blood pressure and heart rate were compared in the anesthetized rat. When given intravenously, all five compounds induced hypotensive and bradycardiac effects. The order of potency for these effects was Nal-Arg Antagonist approximately detirelix [( N-Ac-D-Nal(2)1, D-pCl-Phe2,D-Trp3,D-hArg(Et2)6,D-Ala10]LHRH) greater than [N-Ac-D-Nal(2)1, D-pCl-Phe2,D-Pal(3)3,D-hArg(Et2)6,L-hArg (Et2)8,D-Ala10]LHRH (RS-26306) approximately antide greater than [N-Ac-D-Nal(2)1, D-pCl-Phe2,D-Pal(3)3,6, L-hArg(Et2)8,D-Ala10]LHRH (RS-15378) and did not parallel the order of antiovulatory potencies of these compounds. The hypotensive activity of LHRH antagonists, therefore, appeared dissociable from their antiovulatory activity. RS-26306 and RS-15378 appeared to have the greatest therapeutic ratios. 相似文献

Summary Rye secalins, telomeric C-bands, and telocentric chromosomes were used as markers in the progeny of a test-cross in order to determine the position of seed storage-protein genes Glu-R1 and Gli-R1 with respect to the centromere and both telomeres of chromosome 1 R in rye. These genes were linked to the centromere (32.35±3.28% and 36.27±3.37% recombination, respectively). Glu-R1 was loosely linked to the telomere of the long arm (43.63±3.47% recombination), while Gli-R1 was closely linked to the telomere of the short arm (9.80±2.08% recombination). This finding supports the possibility that rye - and -secalin genes may be located on the satellites, as has been described in wheat for genes that code similar proteins. The importance of metaphase-I pairing failure and its consequences for the estimation of the recombination fraction are also discussed. 相似文献
