The primary structure of one of the bovine brain substance P precursors has shown the existence of a second mammalian tachykinin sequence, named substance K, that is remarkably homologous to that of the amphibian peptide kassinin. In this study, three substance K sequences were chemically synthesized and were submitted to parallel bioassays with kassinin, substance P and physalaemin. The results show that the three substance K peptides all possess biological activities characteristic of the tachykinin family and that their biological activities more closely resemble those of kassinin than those of substance P or physalaemin. This suggests that substance K may have a physiological role which is related to but different from that of substance P in mammalian organisms.  相似文献

Rapid increases in activities and components of mitochondrial particles isolated from cotyledons of Pisum sativum var. Alaska during the early stage of germination are described. Respiratory rate of the cotyledons increased rapidly as hydration proceeded. A similar but slightly delayed increase in respiratory activity of the isolated mitochondrial fraction was observed. The respiratory control ratio and adenosine 5′-pyrophosphate/oxygen ratio rose during imbibition. Cytochrome oxidase and malate dehydrogenase activities in the mitochondrial fraction increased during the initial phase of imbibition. The increase seemed to precede that in respiratory activity. A significant activity of cytochrome oxidase and most of the malate dehydrogenase activity in the cotyledons were present in the postmitochondrial fraction in the case of the dry seeds. Mitochondrial protein and phospholipid also increased during imbibition, and the rise in the components seemed to concur with that in respiratory activity. The mechanism of mitochondrial development during imbibition is discussed.  相似文献

Summary 3H-thymidine incorporation into mitochondrial DNA of the liver and the kidney cells of chick embryos and newborn mice in tissue culture was shown by means of electron microscope radioautography with accurate localization. In these cells, about 20% of all the mitochondria were labeled at their matrices between the cristae within 4 hours in contact with the radioisotope, which were removed by DN'ase.From the results, it is clear that the mitochondria of avian and mammalian cells in tissue culture synthesize DNA.  相似文献

Excretory and secretory (ES) products collected from adult worms of Strongyloides ratti stimulated interleukin-3 (IL-3) production with mesenteric lymph node cells from infected C57BL/6 mice, but not with normal mesenteric lymph node cells. The IL-3 stimulating components were not major IgG binding antigens. Activity of the IL-3 stimulating components was stable by treatment with protease, although reduced by heating in boiling water.  相似文献

To analyze whether prolactin (PRL) in human follicular fluid (FF) is synthesized locally or derived from the circulation, PRL concentrations of plasma and FF were determined in the patients after ovarian stimulations. The amounts of PRL messenger ribonucleic acid (mRNA) in the follicular tissues during different menstrual phases were also determined. The FF PRL concentration was correlated positively with plasma PRL and highest estradiol levels during the stimulatory cycle. No PRL mRNA sequence was detected in the RNAs extracted from follicles at any stage in the menstrual cycle, although beta-actin mRNA was detected in all samples. In a comparison with pituitary RNA, the PRL mRNA concentration in ovarian follicular tissues seemed to be 10,000 times less than that in the pituitary. These results suggest that FF PRL may not be synthesized locally, but derived from the pituitary via the circulation through passive diffusion, and thus regulated by estrogen.  相似文献

The intestinal mast cells (IMC) were examined in normal and adoptively immunized rats infected with Nippostrongylus brasiliensis. An increase in the numbers of IMC was observed in infected recipients of thoracic duct lymphocytes (TDL) obtained from donor rats which had themselves been infected 10 days previously (Day 10 TDL). The increase in the number of IMC in the mucosa was related to the number of Day 10 TDL transferred into infected recipients. When TDL were fractionated into populations of cells either bearing (sIg+) or lacking (sIg?) surface immunoglobulin, only sIg? cells were able to confer the IMC response. Antigenic stimulation was necessary for the differentiation of intestinal mast cells. There was a marked difference between different strains of rats with regard to worm burden and intestinal mast cell kinetics although the increase in intestinal mast cells was always closely related to the final stage of the rapid phase of worm expulsion. These results are compatible with the concept that intestinal mast cells are derived from T cells and suggest that sIg+ cells do not influence IMC differentiation. Alternatively, the possibility that the transferred TDL regulate the differentiation of cells of host origin into IMC cannot be excluded.  相似文献

Thoracic duct lymphocytes (TDL) obtained from rats either on the tenth day of a primary infection (Day 10 TDL) or 1 or 5 weeks after a tertiary infection (hyperimmune TDL) with Nippostrongylus brasiliensis were fractionated into cells lacking (sIg?) or bearing (sIg+) surface immunoglobulin by a rosetting procedure. The abilities of unfractionated TDL, of the two subpopulations, and of the reconstituted cells to confer protection against the parasite were examined. The effector cells which cause worm expulsion were found only in (sIg?) cells from Day 10 TDL and also predominantly in (sIg?) cells from hyperimmune TDL. However, a small but significant degree of protection was conferred by (sIg+) cells from hyperimmune TDL. These results suggest that the mechanisms involved in worm expulsion are regulated by (sIg?) cells but that (sIg+) cells from hyperimmune rats can also contribute to the mechanisms of worm expulsion.  相似文献

The protective capacities of different sources of immune lymphocytes against Nippostrongylus brasiliensis infection were examined. Thoracic duct lymphocytes (TDL) drained from donors on the tenth day of a primary infection (Day 10 TDL) conferred greater protection against adult worms established by larval infection than either mesenteric lymph node cells (MLNC) or TDL drained from hyperimmune donors. Day 10 TDL also conferred a high degree of protection against intraduodenally implanted “normal” and “damaged” worms. These results suggest that the different susceptibilities of “normal” and “damaged” worms to adoptive protection is a quantitative rather than a qualitative phenomenon. The results also emphasise that kinetic and dose-response experiments are important in evaluating the protective capacities of transferred cells.  相似文献
