In vitro incubation of thymocytes with [2-14C]acetate results in a more intensive label incorporation into chromatin lipids as compared to nuclear lipids. The deleterious effect of gamma-irradiation leads to specific changes in [2-14C]acetate incorporation into the total fraction of phosphatidylcholine-phosphatidylserine and chromatin sphingomyelin. This is paralleled with an increase in the ratio of specific radioactivities of chromatin cardiolipin and nuclear cardiolipin. The changes in specific radioactivities of sphingomyelin, phosphatidylcholine-phosphatidylserine and cardiolipin suggest a role of phospholipids in the mechanisms of regulating the activity of intranuclear structures. 相似文献

Introduction
Ultrasonography (US) might have an added value to clinical examination in diagnosing early rheumatoid arthritis (RA) and assessing remission of RA. We aimed to clarify the added value of US in RA in these situations performing a systematic review.Methods
A systematic literature search was performed for RA, US, diagnosis and remission. Methodological quality was assessed; the wide variability in the design of studies prohibited pooling of results.Results
Six papers on the added value of US diagnosing early RA were found, in which at least bilateral metacarpophalangeal (MCP), wrists and metatarsophalangeal (MTP) joints were scanned. Compared to clinical examination, US was superior with regard to detecting synovitis and predicting progression to persistent arthritis or RA. Eleven papers on assessing remission were identified, in which at least the wrist and the MCP joints of the dominant hand were scanned. Often US detected inflammation in patients clinically in remission, irrespective of the remission criteria used. Power Doppler signs of synovitis predicted X-ray progression and future flare in patients clinically in remission.Conclusions
US appears to have added value to clinical examination for diagnosing of RA when scanning at least MCP, wrist and MTP joints, and, when evaluating remission of RA, scanning at least wrist and MCP joints of the dominant hand. The aim of this study was to analyze quantitatively cellular respiration in intraoperational tissue samples taken from human breast cancer (BC) patients. We used oxygraphy and the permeabilized cell techniques in combination with Metabolic Control Analysis (MCA) to measure a corresponding flux control coefficient (FCC). The activity of all components of ATP synthasome, and respiratory chain complexes was found to be significantly increased in human BC cells in situ as compared to the adjacent control tissue. FCC(s) were determined upon direct activation of respiration with exogenously-added ADP and by titrating the complexes with their specific inhibitors to stepwise decrease their activity. MCA showed very high sensitivity of all complexes and carriers studied in human BC cells to inhibition as compared to mitochondria in normal oxidative tissues. The sum of FCC(s) for all ATP synthasome and respiratory chain components was found to be around 4, and the value exceeded significantly that for normal tissue (close to 1). In BC cells, the key sites of the regulation of respiration are Complex IV (FCC?=?0.74), ATP synthase (FCC?=?0.61), and phosphate carrier (FCC?=?0.60); these FCC(s) exceed considerably (~10-fold) those for normal oxidative tissues. In human BC cells, the outer mitochondrial membrane is characterized by an increased permeability towards adenine nucleotides, the mean value of the apparent K(m) for ADP being equal to 114.8?±?13.6?μM. Our data support the two-compartment hypothesis of tumor metabolism, the high sum of FCC(s) showing structural and functional organization of mitochondrial respiratory chain and ATP synthasome as supercomplexes in human BC. 相似文献

The functional geometry of the reconstructed dendritic arborization of Purkinje neurons is the object of this work. The combined effects of the local geometry of the dendritic branches and of the membrane mechanisms are computed in passive configuration to obtain the electrotonic structure of the arborization. Steady-currents applied to the soma and expressed as a function of the path distance from the soma form different clusters of profiles in which dendritic branches are similar in voltages and current transfer effectiveness. The locations of the different clusters are mapped on the dendrograms and 3D representations of the arborization. It reveals the presence of different spatial dendritic sectors clearly separated in 3D space that shape the arborization in ordered electrical domains, each with similar passive charge transfer efficiencies. Further simulations are performed in active configuration with a realistic cocktail of conductances to find out whether similar spatial domains found in the passive model also characterize the active dendritic arborization. During tonic activation of excitatory synaptic inputs homogeneously distributed over the whole arborization, the Purkinje cell generates regular oscillatory potentials. The temporal patterns of the electrical oscillations induce similar spatial sectors in the arborization as those observed in the passive electrotonic structure. By taking a video of the dendritic maps of the membrane potentials during a single oscillation, we demonstrate that the functional dendritic field of a Purkinje neuron displays dynamic changes which occur in the spatial distribution of membrane potentials in the course of the oscillation. Learning and memory disorders accompanying epileptogenesis were studied in rats with the use of two experimental models of epilepsy, picrotoxin kindling and kainic treatment. Rise of exploratory activity and decrease in animal's capability for experimental extinction of a response were characteristic of the initial stage of epileptogenesis. It was suggested that a dysfunction of brain hippocampal system can be responsible for cognitive disorders. To reveal their mechanisms, lipid contents were determined in the neocortex and hippocampus in appropriate periods after exposure to epileptogenic factors. Long-term changes in hippocampal lipid spectrum were found five days after the exposure to kainic acid. In particular, after sodium valproate treatment (the compensation of kainic effects), the total content of phospholipids in hippocampus was decreased. The hippocampal sphingomyelin level dropped as a result of picrotoxin kindling. The sphingomyelin changes suggest some recovery processes in hippocampal cells and point to an adaptive role of membrane lipids in the mechanisms of the damaging epiptogenous effects. 相似文献

Cultured neuronal networks, which have the capacity to respond to a wide range of neuroactive compounds, have been suggested to be useful for both screening known analytes and unknown compounds for acute neuropharmacologic effects. Extracellular recording from cultured neuronal networks provides a means for extracting physiologically relevant activity, i.e. action potential firing, in a noninvasive manner conducive for long-term measurements. Previous work from our laboratory described prototype portable systems capable of high signal-to-noise extracellular recordings from cardiac myocytes. The present work describes a portable system tailored to monitoring neuronal extracellular potentials that readily incorporates standardized microelectrode arrays developed by and in use at the University of North Texas. This system utilizes low noise amplifier and filter boards, a two-stage thermal control system with integrated fluidics and a graphical user interface for data acquisition and control implemented on a personal computer. Wherever possible, off-the-shelf components have been utilized for system design and fabrication. During use with cultured neuronal networks, the system typically exhibits input referred noise levels of only 4-6 microVRMS, such that extracellular potentials exceeding 40 microV can be readily resolved. A flow rate of up to 1 ml/min was achieved while the cell recording chamber temperature was maintained within a range of 36-37 degrees C. To demonstrate the capability of this system to resolve small extracellular potentials, pharmacological experiments with cultured neuronal networks have been performed using ion channel blockers, tetrodotoxin and tityustoxin. The implications of the experiments for neurotoxin detection are discussed. 相似文献

The effect of dietary supplementation of old rats (26–33 months) with hydrogenated peanut oil on the activity of mitochondrial
enzymes in skeletal muscles has been studied. The activities of NADH-coenzyme Q1 oxidoreductase, cytochrome c oxidase, and citrate synthase were determined spectrophotometrically in muscle homogenates. The activities of respiratory
complexes I and IV were shown to significantly decrease with the age compared to the activity of the same enzymes in young
animals, while the activity of citrate synthase was virtually unchanged. The fatty acid composition of muscle homogenates
of old rats differed from that of young animals by a reduced content of myristic, oleic, linoleic, and α-linolenic acids and
enhanced content of dihomo-γ-linolenic, arachidonic, and docosahexaenoic acids. Per oral supple-mentation of the old rats
with hydrogenated peanut oil completely restored the activity of complex IV and increased the activity of complex I to 80%
of the value observed in muscles of young animals, reducing the content of stearic, dihomo-γ-linolenic, arachidonic, eicosapentaenoic,
docosapentaenoic, and docosahexaenoic acids relative to that in the groups of old and young rats. The content of oleic and
linoleic acids increased relatively to that in the group of the old rats, as well as young animals. The possible mechanisms
of the restoration of the activity of the respiratory enzymes under the administration of hydrogenated peanut oil are discussed. 相似文献
