The early colonization of Scots pine (Pinus sylvestris L.) sapwood exposed above ground (staple bed) was studied. Two different types of exposures were used, one in an open field and the other in a shaded field. Decay type and degree of degradation due to soft rot, and mass and strength loss of wood were correlated. Fungal species in Scots pine sapwood were identified by sequencing, using the fungal nuclear ribosomal DNA (nrDNA) after 24 months.The most abundant decay type found was soft rot, which also agreed with the mass loss (7–8%). Pine sapwood did not differ significantly between the two sites regarding the average mass loss during the time of exposure. The early colonization of wood by soft rot fungi together with mass loss indicates that this fungal type might be more common in above-ground conditions than recognized earlier.  相似文献

The objective of this study was to evaluate the natural durability of five lesser-utilized wood species from Mozambique. Both laboratory methods and field tests were applied for assessing wood decay of muanga (Pericopsis angolensis), metil (Sterculia appendiculata), namuno (Acacia nigrescens), ncurri (Icuria dunensis), and ntholo (Pseudolachnostylis maprounaefolia). Laboratory tests involved soft-, brown-, and white-rot fungi and termites. Heart- and sapwood of ncurri and ntholo were exposed in above-ground field tests; additionally, all species were exposed to in-ground contact tests. The results indicated that namuno, muanga, ncurri, and ntholo are resistant to soft-, brown- and white-rot fungi and the termite species Reticulitermes grassea and Mastotermes darwiniensis. Comparatively, soft-rot caused more severe decay on the studied wood species than did basidiomycete fungi. The brown-rot fungi Coniophora puteana, Gloeophiyllum trabeum, and Postia placenta caused less decay on the tested species than did the white-rot Trametes versicolor. Metil was not resistant to any of the mentioned hazards. Therefore, this species is not recommendable for exterior use if untreated.  相似文献

In this study, the effect of two boric acid concentrations (1% and 2%) and four derivates of tall oil with varying chemical composition were tested separately and in combination. The tall oil derivates were chosen in a way that they consist of different amounts of free fatty, resin acids and neutral compounds. Decay tests using two brown rot fungi (Postia placenta and Coniophora puteana) were performed on both unleached and leached test samples. Boric acid showed a low weight loss in test samples when exposed to fungal decay before leaching, but no effect after leaching. The tall oil derivates gave better efficacy against decay fungi compared to control, but are not within the range of the efficacy needed for a wood preservative. Double impregnation with boric acid and tall oil derivates gave synergistic effects for several of the double treatments both in unleached and leached samples. In the unleached samples the double treatment gave a better efficacy against decay fungi than tall oil alone. In leached samples a better efficacy against brown rot fungi were achieved than in samples with boron alone and a nearly similar or better efficacy than for tall oil alone. Boric acid at 2% concentration combined with the tall oil derivate consisting of 90% free resin acids (TO-III) showed the best performance against the two decay fungi with a weight loss less than 3% after a modified pure culture test.  相似文献

A modification of the Freundlich and Avdalovic method (J. Immunol. Methods 62, 31 (1983] is reported. Buffy coats, separated and pooled together, are used for isolation of monocytes (70% yield, 100% purity). Cell density of working suspension is increased up to 0.65 X 10(9) cells/75 cm2 surface by multiplication of the active fibronectin sites. For the purpose, cryoprecipitate is used instead of plasma for coating the glass-gelatin surface. Monocytes, isolated by that procedure, could be successfully cryopreserved with dimethyl sulfoxide cryoprotective solution.  相似文献

Two types of silanes, namely aklylalkoxysilanes and arylalkoxysilanes, were used for wood impregnation aiming at hydrophobisation of the ligno-cellulose material. The suggested mixture of alkyltriethoxysilanes and dialkyldiethoxysilanes improved water repellence when compared to conventionally used silanes. An attempt to immobilise boric acid with the siloxane matrix was also done. Decay resistance of the treated samples was tested by soil-jar test using the brown rot fungus Postia placenta.Features of the impregnated wood such as moisture content, swelling, anti-swelling efficacy and weight percentage gain were not significantly related to the mass loss of samples caused by the fungus. Samples impregnated with EtPhSi(OEt)2 silanes showed improved durability (3.5–11.5% mass loss). Addition of boric acid to two of the silanols showed the best result for durability of wood, i.e. the lowest mass loss of 2.3 and 1.1%. These treatments upgraded the wood to “very durable”, a significant improvement from its natural status, i.e. slightly durable. The studied alkoxysilanes have moderate ability to penetrate the wood cell wall and hence to improve its hydrophobicity. These silanes are promising although more research should be carried out on the degree of oligomer polymerisation in the wood cell wall.  相似文献

One consistent finding among studies using shotgun metagenomics to analyze whole viral communities is that most viral sequences show no significant homology to known sequences. Thus, bioinformatic analyses based on sequence collections such as GenBank nr, which are largely comprised of sequences from known organisms, tend to ignore a majority of sequences within most shotgun viral metagenome libraries. Here we describe a bioinformatic pipeline, the Viral Informatics Resource for Metagenome Exploration (VIROME), that emphasizes the classification of viral metagenome sequences (predicted open-reading frames) based on homology search results against both known and environmental sequences. Functional and taxonomic information is derived from five annotated sequence databases which are linked to the UniRef 100 database. Environmental classifications are obtained from hits against a custom database, MetaGenomes On-Line, which contains 49 million predicted environmental peptides. Each predicted viral metagenomic ORF run through the VIROME pipeline is placed into one of seven ORF classes, thus, every sequence receives a meaningful annotation. Additionally, the pipeline includes quality control measures to remove contaminating and poor quality sequence and assesses the potential amount of cellular DNA contamination in a viral metagenome library by screening for rRNA genes. Access to the VIROME pipeline and analysis results are provided through a web-application interface that is dynamically linked to a relational back-end database. The VIROME web-application interface is designed to allow users flexibility in retrieving sequences (reads, ORFs, predicted peptides) and search results for focused secondary analyses.  相似文献
