A note on antibiotic-resistant Escherichia coli in adult man, raw sewage and sewage-polluted River Tigris in Mosul, Nineva

A total of 600 isolates of Escherichia coli were isolated, over a 9 month period during 1984, from healthy human adults, raw sewage and the sewage-polluted River Tigris in Nineva. Over 90% of these organisms were E. coli type 1, but only 8.3% could be serogrouped as enteropathogenic E. coli . Resistance of these organisms to 11 antimicrobial drugs was assessed. Over 40% were antibiotic-resistant and of these 77.1% were resistant to more than one antibiotic. The minimal inhibitory concentration of ampicillin for 193 selected strains from the various sources was determined and ranged from <0.625- > 160 μg/ml. The high incidence of antibiotic-resistant E. coli in this locality and the possible implications to human health are discussed.     

Transport Properties of the Tomato Fruit Tonoplast : I. Identification and Characterization of an Anion-Sensitive H-ATPase

An anion-sensitive H⁺-translocating ATPase was identified in membrane vesicles isolated from mature green tomato (Lycopersicon esculentum) fruit. The H⁺-ATPase was associated with a low density membrane population having a peak density of 1.11 grams per cubic centimeter, and its activity was inhibited by NO₃⁻, N,N′-dicyclohexylcarbodiimide and diethylstilbestrol but not by vanadate, azide, molybdate, or oligomycin. This H⁺-ATPase has an unusual pH dependence indicating both a slightly acidic and a near neutral peak of activity. Chloride was found to be a potent stimulator of ATPase activity. The K_m for the H⁺-ATPase was approximately 0.8 millimolar ATP. The characteristics of this H⁺-ATPase are very similar to those described for a number of plant cell tonoplast H⁺-ATPases suggesting that the activity identified in tomato fruit membranes is tonoplast-associated. This report demonstrates the feasibility of isolating tonoplast vesicles from acidic fruit tissues for studies of transport activities associated with fruit development and maturation.     

Synthesis and evaluation of the antidepressant activity of the enantiomers of bupropion

The synthesis of the enantiomers of bupropion, (rac)-2-tert-butylamino-3′-chloropropiophenone 1 (Wellbutrin®) is described. The enantiomers were compared with the racemate in both the tetrabenazine-induced sedation model and the inhibition of uptake of biogenic amine assay. No significant differences were found in their potencies to reverse tetrabenazine-induced sedation in mice or in their IC₅₀ values as inhibitors of biogenic amine uptake into nerve endings obtained from mouse brain. © 1993 Wiley-Liss, Inc.     

Synthesis of New Bis(pyrazolo[1,5-a]pyrimidines) Linked to Different Spacers as Potential MurB Inhibitors

An efficient protocol was adopted to efficiently prepare three new series of bis(pyrazolo[1,5-a]pyrimidines) linked to different spacers. The new bis(pyrazolo[1,5-a]pyrimidines) were prepared in 80–90 % yields by reacting the respective bis(enaminones) and 4-(4-substituted benzyl)-1H-pyrazole-3,5-diamines in pyridine at reflux temperature for 5–7 h. The new products showed a wide spectrum of antibacterial activity against six different bacterial strains. In general, propane- and butane-linked bis(pyrazolo[1,5-a]pyrimidines), which are attached to 3-(4-methyl- or 4-methoxybenzyl) units, had the best antibacterial activity with minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values up to 2.5 and 5.1 μM, respectively. Additionally, the previous products demonstrated promising MurB inhibitory activity with IC₅₀ values up to 7.2 μM.     

Haemoglobin Arya: alpha 2-47 (CD5), aspartic acid yields asparagine.

A new haemoglobin variant (haemoglobin Arya), is described from an Iranian female. The substitution is at residue 47 (CD5) of the alpha chain in which aspartic acid has been substituted by asparagine. The presence of haemoglobin Arya was not associated with clinical symptoms. This variant has normal stability at 50 degrees C, but is slightly unstable when tested at 55 degrees C.

Role of N-Cadherin cis and trans Interfaces in?the Dynamics of Adherens Junctions in Living Cells

Cadherins, Ca²⁺-dependent adhesion molecules, are crucial for cell-cell junctions and remodeling. Cadherins form inter-junctional lattices by the formation of both cis and trans dimers. Here, we directly visualize and quantify the spatiotemporal dynamics of wild-type and dimer mutant N-cadherin interactions using time-lapse imaging of junction assembly, disassembly and a FRET reporter to assess Ca²⁺-dependent interactions. A trans dimer mutant (W2A) and a cis mutant (V81D/V174D) exhibited an increased Ca²⁺-sensitivity for the disassembly of trans dimers compared to the WT, while another mutant (R14E) was insensitive to Ca²⁺-chelation. Time-lapse imaging of junction assembly and disassembly, monitored in 2D and 3D (using cellular spheroids), revealed kinetic differences in the different mutants as well as different behaviors in the 2D and 3D environment. Taken together, these data provide new insights into the role that the cis and trans dimers play in the dynamic interactions of cadherins.     

Efficacy of pesticides on the functional response on larval ectoparasitoid,Habrobracon hebetor Say (Hymenoptera: Braconidae)

Exposing to sub-lethal and low lethal doses of pesticides may cause changes in natural enemy behavioural, such as functional, response. In this study, the effects of chlorpyrifos, carbaryl, abamectin and spinosad were evaluated on the functional response of the Habrobracon hebetor to different densities of last instar larvae of Anagasta kuehniella Zeller. Young adult females of the parasitoid were exposed to LC₃₀ of chlorpyrifos, carbaryl, abamectin and spinosad that were 0.32, 4.03, 3.05 and 17.51?mg a.i./l for 24?h, respectively. Host densities of 2, 4, 8, 16, 32 and 64 were offered to treated young females for 2?h in 10-cm Petri dishes and then the parasitism data were recorded. Experiments were conducted in eight replications. Functional response type was determined using logistic regression and the parameters were appraised by non-linear regression using statistical analysis software. Functional response was type Ш in control and insecticide treatments. Searching efficiency in control, chlorpyrifos-, carbaryl-, abamectin- and spinosad-treated wasps were 0.008?±?0.002, 0.002?±?0.0009, 0.0034?±?0.0013, 0.0076?±?0.002 and 0.0073?±?0.002?h^?1and handling times were 1.38?±?0.1, 7.64?±?1.01, 3.3?±?0.315, 1.55?±?0.1 and 1.46?±?0.11?h, respectively. Chlorpyrifos and carbaryl had the highest effect on searching efficiency of H. hebetor. Spinosad and abamectin showed less adverse effect on the functional response parameters. Finally, after conducting the advanced field studies, spinosad and abamectin may be used as a compatible chemical material with biological control agent in integrated pest management programmes.
