Requirements for seed germination (emergence of radicle) andseedling formation (emergence of both radicle and cotyledons)of a hemi-root parasite Sopubia delphinifolia were studied inthe absence of any host stimulus, in Petri dish and asepticcultures. Both water washing in trickling tap water as wellas cold treatment were effective in inducing/stimulating germinationand seedling formation in the light. Although ethrel, an ethylenereleasing compound, stimulated radicle emergence it inhibitedthe emergence of the cotyledons. Light was found to be essentialfor germination; none of the growth substances could replacethe light requirement. Light responses seem to be mediated throughthe phytochrome system. The results indicate that the emergenceof the radicle, its further growth into the root and the emergenceof cotyledons are controlled by different factors. The present report is a part of our study on the reproductivebiology of a traditional Indian medicinal plant,Commiphora wightii,a source of guggul steroids. Field examination showed a predominantlylarge number of isolated and groups of female individuals. Onlyone andromonoecious and two exclusively male plants were recorded.Female plants set seed irrespective of the presence or absenceof pollen. Hand-pollination experiments and embryological studieshave confirmed the occurrence of non-pseudogamous apomixis,nucellar polyembryony and autonomous endosperm formation forthe first time in this plant, which is presently threatenedby over-exploitation. Detailed studies were carried out on the phenology, floral biology, pollination ecology and breeding system of Boswellia serrata Roxb. (Burseraceae) the source of 'salai guggul'. The trees remain leafless during the entire period of flowering and fruiting. The inflorescence is a terminal raceme and produces up to 90 bisexual, actinomorphic flowers. On average a flower produces 10 044 ± 1259 starch-filled pollen grains. About 85% of the fresh pollen grains are viable; the pollen to ovule ratio is 3348 : 1. The stigma is of the wet papillate type. The style is hollow with three flattened stylar canals filled with a secretion product. The stylar canals are bordered by a layer of glandular canal cells. The inner tangential wall of the canal cells shows cellulose thickenings. The ovary is trilocular and bears three ovules, one in each locule. Flowers offer nectar and pollen as rewards to floral visitors. The giant Asian honey bee ( Apis dorsata ) and A. cerana var. indica (Indian honey bee) are the effective pollinators. The species is self-incompatible and the selfed pollen tubes are inhibited soon after their entry into the stigma. Self-pollen tubes develop a characteristic 'isthmus' as a result of enlargement of the tube soon after emergence through the narrow germ pore. Cross-pollinated flowers allowed normal pollen germination and pollen tube growth, and resulted in fruit- and seed-set. Under open pollination fruit-set was only about 10%. Although manual cross-pollinations increased fruit set, it was only up to about 20%. Strobilanthes kunthianus (Acanthaceae) is a semelparous species with synchronized flowering and mast seeding once in 12 years. As semelparous plants have only one chance to reproduce, they are expected to develop effective strategies to prevent reproductive failure. The reproductive strategies of S. kunthianus were investigated by studying the floral traits, pollination biology, and breeding system that are critical for reproductive success. The species exhibits a series of floral traits: (1) gregarious flowering attracts a large number of Apis cerana indica , the major pollinator; (2) the stigma is sensitive to touch by the pollinator; in fresh flowers, the receptive surface faces the entry path of the incoming bee, facilitating pollen deposition; as an immediate response, the stigma curves backwards moving the receptive surface away from the path of the exiting bee, thus preventing autogamy and interference in pollen transfer; (3) flowers remain fresh for 2 days with receptive stigma and nectar and pollen reward. These traits render the species 100% pollination efficient to ensure a high seed set. As the species is self-compatible, the prevailing high degree of geitonogamous pollinations does not interfere with fruit set. The stimulation of adenosine monophosphate-activated protein kinase (AMPK) is a prime target to decrease the hyperglycemic condition, hence it is a lutein (L) and oxidised lutein (OXL) is a target molecule for the treatment of type II diabetes. In the current study, a plausible interaction of L and OXL with AMPK was investigated by molecular docking. In addition, the effect of L and OXL for the activation of AMPK that triggers the downstream regulator peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α), TFAM expression, mitochondrial DNA (mtDNA), mitochondrial biogenesis and superoxide dismutase 2 (SOD2) in high glucose treated HepG2 cells were investigated by quantitative polymerase chain reaction and Western blot analysis. Molecular docking reveals higher binding affinity of L (ΔG = −6.3 kcal/mol) and OXL (ΔG = −15.5 kcal/mol) with AMPK, compared with metformin (ΔG = −5.0 kcal/mol). The phosphorylation of AMPK increased by 1.3- and 1.5-fold with L and OXL treatment, respectively, in high glucose induced HepG2 cells. The activation of PGC-1α is significant (P < 0.05) in OXL group than L. Similarly, TFAM expression is increased with L and OXL compared with the high glucose group. Further increase in SOD2 and mtDNA, confirms the efficacy of L and OXL in restoring the mitochondrial biogenesis in high glucose induced cells through AMPK, PGC-1α, and TFAM.  相似文献

Developmental morphometry, qualitative and quantitative analysis of the accessory gland secretory proteins, fecundity and productivity in relation to protein ejected during subsequent (first to fourth-time) matings have been studied in Drosophila ananassae Doleschall and Drosophila varians Bock. In both species, size and secretion of accessory glands increases from 1 to 8 days and the stored secretion ejected from males to the female genital tract during subsequent mating varies. The maximum number of eggs and flies are produced from the females mated with bachelor males and it is a minimum when virgin females are mated with fourth-time mated males. Sodium dodecylsulfate–polyacrylamide gel electrophoresis analysis of accessory gland secretory protein patterns and their glycosylation differs in both the species. Correlation coefficient analysis between gland size and quantity of secretion, percentage of secretory protein transferred per mating, and eggs and flies that emerged showed a highly significant, positive relationship. Among different matings, the number of eggs laid and flies that emerged per female between subsequent (first to fourth-time) matings of males was found to be highly significant and the difference between fecundity and productivity between the two species was highly significant.  相似文献

Nucellus cultures of Luffa cylindrica and Trichosanthes anguina(Cucurbitaceae) have been established. Tracheary differentiationoccurred in cultures of both taxa. Supplementing the culturemedium with 2,4-D caused limited and diffuse differentiationof tracheary elements. In the absence of 2,4-D, tracheary differentiationwas profuse and the tracheary elements formed compact nodules;the differentiation of tracheary nodules could be traced towell-developed meristemoids. Our results show that the formationof tracheary nodules is an ultimate event in morphogenesis andbears no causal relation to organogenesis. The suitability ofthe nucellus as a system for studies on tracheary differentiationis emphasized.  相似文献

Pollen grains of Brassica juncea (L) Czern. were exposed tohigh temperature (45, 60 and 75°C, for 4–24 h) andtheir viability [using the fluorochromatic reaction (FCR) test],germination, and ability to set fruits and seeds were investigated.High temperature up to 60°C did not affect pollen viability;these pollen samples germinated on the stigma and produced fruitsand seeds. There was no correlation between viability and seedset in pollen samples exposed to 75°C; they showed onlyabout 25% reduction in viability but failed to germinate onthe stigma and, consequently, to set seeds. However, these pollensamples induced parthenocarpy. A clear relationship betweenviability and seed-set was observed when the extent of retentionof fluorescence in pollen over a 2-h period was used to assessviability. The fluorescence was retained even after 2 h in pollensamples which induced seed-set, while in those which failedto induce seed-set the fluorescence was lost in about 60 mindue to the leakage of fluorescein. Thus, in stressed pollen,the extent of retention of fluorescence may give a better indicationof pollen function than initial fluorescence, as is being doneroutinely in FCR test. Treatment of stigma with a lectin (Con A/PHA) before pollinationwas effective in overcoming self-incompatibility in Petuniahybrida, a gametophytic self-incompatible system, and Erucasativa, a sporophytic self-incompatible system. Treatment ofpollen with glucose/N-acetyl-D-galactosamine (tested only withPetunia) was also effective. These results suggest the involvementof pollen lectins and specific sugar components of the pistilin self-incompatibility recognition.
