Nuclear Receptors in Mosquito Vitellogenesis   总被引:1,自引:0,他引:1  
Vitellogenesis in insects involves the coordinated activityof the fat body, which produces large amounts of yolk proteinprecursors (YP), and oocytes, which specifically accumulatethese proteins. The expression of YP genes is achieved throughstrict sex-, tissue-, and hormone-specific control in the femalefat body. In mosquitoes, expression of YP genes is controlledby 20-hydroxyecdysone (20E). To elucidate the role of 20E inmosquito vitellogenesis, we cloned cDNAs encoding the Aedesaegypti ecdysteroid receptor (AaEcR) and two isoforms of itsheterodimeric partner, the Ultraspiracle homologue (AaUSP).The two AaUSP isoforms differ in their A/B domains and havedistinct expression patterns. The ecdysone regulation of YPgenes likely involves products of early genes. We cloned thegene of the mosquito homologue to the Drosophila early geneE75 (AaE75) belonging to the nuclear receptor superfamily. Kineticsof AaE75 expression correlate with the expression of YP genes,suggesting that AaE75 may have a regulatory role in YP geneexpression. A second nuclear receptor superfamily member, theNGFI-B homologue AaHR38 is implicated in repression of the ecdysone-signalingpathway in the fat body of the previtellogenic female mosquitoat the state-of-arrest. Finally, three isoforms of the hepatocytenuclear factor 4 (HNF-4) homologue AaHNF-4 are differentiallyexpressed in the mosquito fat body during vitellogenesis, suggestingtheir involvement in regulating vitellogenic events in thistissue.  相似文献

An important question for the conservation of species dwelling in fragmented habitats is whether changes to the intervening landscape create a barrier to gene flow. Here, we make use of the spatial distribution of the granite outcrop‐dwelling lizard, Ctenophorus ornatus, to compare inferred levels of gene flow between outcrops in a nature reserve with that between outcrops in the adjacent agricultural land. Genetic variation, relatedness and subdivision were compared within groups of individuals from different outcrops similar in size and distance apart at each site. In the agricultural land, we found significantly lower genetic variation within outcrops and greater genetic differentiation between outcrops than in the reserve. Further, the rate at which genetic divergence between outcrops increased over geographical distance was significantly greater in the agricultural land than in the reserve. We also found that individuals were more closely related within outcrops but more distantly related between outcrops in the cleared land. These effects occur over a small spatial scale with an average distance between outcrops of less than five kilometres. Thus, even though land clearing around the outcrops leaves outcrop size unchanged, it restricts gene flow, reducing genetic variation and increasing population structure, with potentially negative consequences for the long‐term persistence of the lizards on these outcrops.  相似文献

Microsatellites were identified from three fully sequenced Y chromosome‐specific bacterial artificial chromosome clones from the tammar wallaby, Macropus eugenii. Ten microsatellites were genotyped in male tammar wallabies. Four loci were polymorphic with between two and six alleles per locus. Eleven different haplotypes were identified from 22 male tammar wallabies. No amplifications were obtained from female samples. Each microsatellite was also shown to amplify reliably in at least one other macropod species. These markers may therefore prove useful as some of the first male‐specific genetic markers for marsupials, with potential application to studies of male‐biased dispersal and mating systems.  相似文献

Abstract. Morphological and electrophoretic variation is evaluated in 272 female and 424 male specimens of Aphidius reared from known hosts representing seventeen aphid-host species. Much of this material was reared under experimental conditions in order to examine the effects of environmental conditions such as temperature and different host species, and to induce as much variation as is possible in known single populations. Multivariate analysis and electrophoresis are used to demonstrate the degree to which entities defined by morphological characters and electrophoretic bands correspond to collection from a common host.
Many characters commonly used in the identification of this group, e.g. sculpturing on the propodeum, are shown to be very variable. Despite this, several morphologically and electrophoretically definable groups of Aphidius are demonstrated, many of them corresponding to individual aphid-hosts, while others, especially those parasitizing pea and cereal aphids were found to occur on more than one host. The possible nominal species associated with each defined group are discussed and morphological keys given to these complexes, and to the species of of Aphidius known to parasitize pea and cereal aphids.  相似文献

In theory, one factor determining the rate and nature of the assembly of island biotas is the presence or absence of stepping stone islands, yet no field studies have demonstrated stepping stone function in practice. Krakatau, in Sunda Strait, is about equidistant from Java and Sumatra. Sebesi lies about half way between Krakatau and Sumatra, but no island intervenes between Krakatau and the nearest coast of Java. We assess the evidence that Sebesi has acted as an important stepping stone for Krakatau's recolonization since the devastating 1883 volcanic eruption. About a quarter of Krakatau's resident land birds, two-fifths of its reptiles, bats and land molluscs, and about two-thirds of its termites, pteridophytes, butterflies and spermatophytes are unknown on Sebesi, evidently having colonized without stepping stone involvement. Identifiable Sumatran taxa do not outnumber identifiable Javan ones on Krakatau, nor do historical distribution records indicate movement from Sebesi to Krakatau in animal groups. Krakatau's biota is not a subset of Sebesi's in predominantly anemochorous or thallassochorous plant groups, butterflies, reptiles or bats, and is only marginally so in termites. It is a subset in predominantly zoochorous spermatophyte groups, except Ficus species, and in birds and land molluscs. Comparison with a weaker stepping stone candidate, Panaitan, provides no evidence for a stepping stone role for Sebesi in butterflies or termites. We discuss the dispersal and establishment constraints on colonization by the groups involved, and conclude that, overall, Sebesi had little impact as a stepping stone. 1. The spread of Argentine ants, Linepithema humile (Mayr), in introduced areas is mainly through the displacement of native ant species owing to high inter‐specific competition. In South Africa, L. humile has not established in the climatically suitable eastern and northern escarpments dominated by the African big headed ant, Pheidole megacephala (Fabricius), probably owing to local biotic resistance. 2. Inter‐specific aggression, at the individual and colony level, and competition for a shared resource were evaluated in the laboratory. 3. Aggression between the two ant species was very high in all of the assays. Both species suffered similar mortality rates during one‐on‐one aggression assays, however, during symmetrical group confrontations, L. humile workers showed significantly higher mortality rates than P. megacephala workers. During asymmetrical group confrontations both species killed more of the other ant species when they had numeric advantage. Both ant species located the shared resource at the same time; however, once P. megacephala discovered the bait, they displaced L. humile from the bait through high inter‐specific aggression, thereafter dominating the bait for the remainder of the trial. 4. The results demonstrate the potential of P. megacephala to prevent the establishment and survival of incipient L. humile colonies through enhanced resource competition and high inter‐specific aggression. This is the first study to indicate potential biotic resistance to the spread of L. humile in South Africa.  相似文献

A blood meal stimulates the ovaries of the mosquito, Aedes aegypti , to internalize vitellogenin from the haemolymph for storage as yolk. The nature of the stimulation is biphasic, with a pulsed factor from the head released within the first 5 min of feeding stimulating the first phase of uptake, and the continuous release of another head factor initiating the second phase. Using an in vitro assay of [35S]-vitellogenin uptake by ovaries, we sought to determine the intracellular signalling pathways utilized by the ovaries during stimulation of both phases of uptake. The adenylate cyclase activators forskolin and ethanol produced a statistically significant increase in rate of uptake in previtellogenic ovaries to levels characteristic of first-phase activated ovaries. Forskolin also induced the second phase of uptake in ovaries that had been arrested in the first phase of uptake by decapitation of the mosquito within 5 min of ingesting blood. Initiation and up-regulation of the rate of vitellogenin uptake does not depend on new protein synthesis.  相似文献

Stages of mitosis of the micronuclei of Stentor coeruleus were described as seen by transmission electron microscopy. Cells in division and those regenerating new oral membranelles were studied. Microtubules were found in early prophase in the karyoplasm and interspersed between the condensing chromatin. A monaxial intranuclear spindle is formed by early metaphase, with kinetochore microtubule attachment sites on the chromosomes. The spindle elongates, separating the daughter nuclei at anaphase. A new nuclear envelope, consisting of two unit membranes, begins to form at late anaphase. Small segments of membrane found in the space between the newly forming and the old micronuclear envelopes appear to fuse to form the new nuclear envelope. No ultrastructural differences were found in the mitotic nuclei of cells in division or regeneration.  相似文献
