Many herbs and spices are known to modulate the immune system and have been shown to restore the immunity in immuno-compromised individuals. Spices generally used to increase the taste and flavor of food also has the history of usage as an ayurvedic medicine. Therefore to explore the health modulating effects of Cuminum cyminum and to identify the active compound, immunomodulatory properties were evaluated using flowcytometry and ELISA in normal and immune-suppressed animals. C. cyminum and compound 1 stimulated the T cells and Th1 cytokines expression in normal animals. Swiss albino mice subjected to Cyclosporine-A induced immune-suppression were dosed orally with C. cyminum (25, 50, 100 and 200 mg/kg) on consecutive days. The results showed that administration significantly increased T cells (CD4 and CD8) count and Th1 predominant immune response in a dose dependent manner thereby suggesting immunomodulatory activity through modulation of T lymphocytes expression. In restraint stress induced immune-suppressed animals, compound 1 countered the depleted T lymphocytes, decreased the elevated corticosterone levels and size of adrenal glands and increased the weight of thymus and spleen. Based on the data we may conclude that C. cyminum is a potent immunomodulator and may develop as a lead to recover the immunity of immuno-compromised individuals.  相似文献

A new steroidal sapogenin molecule 1 having unique characteristics, 21-nor and unusual C19 carboxylic acid has been isolated from the roots of Asparagus racemosus. On the basis of chemical evidence, extensive spectroscopic analysis including two dimensional (2D) NMR and X-ray studies of single crystal, the structure of 1 was determined as (1S,2R,3S,8S,9S,10S,13S,14S,16S,17R,22R,25R)-21-nor-18β,27α-dimethyl-1β,2β,3β-trihydroxy-25-spirost-4-en-19β-oic acid. 1 crystallizes in monoclinic space group P21 with a = 9.295(2), b = 11.238(2), c = 11.376(2) Å; β = 91.993(4)°, Z = 2, Dcal = 1.344 Mg/m3. The structure was solved by direct methods and refined by full-matrix least-squares procedure to a final R-value of 0.0561 for 4064 observed reflections. 1 was tested against the type of immune responses generated during treatment in normal and immune-suppressed animals and detailed biological activity evaluation suggests it to be a potent immunostimulator.  相似文献

A sipholane triterpenoid, named sipholenone A, with anti‐cancer properties was isolated from the Red Sea sponge Siphonochalina siphonella and characterized by proton and carbon‐13 nuclear magnetic resonance (1H NMR and 13C NMR) spectroscopies. The goal of this study was to visualize the binding of this triterpenoid with human serum albumin (HSA) and to determine its binding site on the biomacromolecule. The interaction was visualized using fluorescence quenching, synchronous fluorescence, far‐ and near‐UV circular dichroism (CD), UV–visible and Fourier transform‐infrared (FT‐IR) spectroscopies. UV–visible spectroscopy indicated the formation of a ground‐state complex as a result of the interaction. Sipholenone A quenches the fluorescence of HSA via a static quenching mechanism. A small blue shift in the fluorescence quenching profiles suggested the involvement of hydrophobic forces in the interaction. Sipholenone A binding takes place at site I of subdomain II A with a 1:1 binding ratio, as revealed by displacement binding studies using warfarin, ibuprofen and digitoxin. Far‐UV CD and FT‐IR studies showed that the binding of sipholenone A to HSA also had a small effect on the protein's secondary structure with a slight decrease in the α‐helical content. Several thermodynamic parameters were calculated, along with Forster's radiative energy transfer analysis.  相似文献

The targeting of a cellular co-factor, rather than the HIV-1-specific RNAs, by small interfering RNAs holds promise as the rapid mutational ability of the HIV-1 genome may obviate the potential clinical use of RNAi against this virus. The DEAD-box RNA helicase DDX3 is an essential Rev co-factor in the CRM1-Rev-RRE complex that promotes the export of unspliced and single-spliced HIV-1 RNAs from the nucleus to cytoplasm. In this report, human DDX3 was targeted by specific short hairpin RNAs, and the down-regulation of cell's endogenous DDX3 suppressed the nuclear export of unspliced HIV-1 RNAs but did not affect the cell viability. We further showed that the knockdown of cellular DDX3 could effectively inhibit the replication of HIV-1. Therefore, the current results suggest that the RNA helicase DDX3 may become a potential target by RNAi for future genetic therapy of HIV/AIDS.  相似文献

Atmospheric gas plasmas (AGPs) are able to selectively induce apoptosis in cancer cells, offering a promising alternative to conventional therapies that have unwanted side effects such as drug resistance and toxicity. However, the mechanism of AGP-induced cancer cell death is unknown. In this study, AGP is shown to up-regulate intracellular reactive oxygen species (ROS) levels and induce apoptosis in melanoma but not normal melanocyte cells. By screening genes involved in apoptosis, we identify tumor necrosis factor (TNF)–family members as the most differentially expressed cellular genes upon AGP treatment of melanoma cells. TNF receptor 1 (TNFR1) antagonist–neutralizing antibody specifically inhibits AGP-induced apoptosis signal, regulating apoptosis signal–regulating kinase 1 (ASK1) activity and subsequent ASK1-dependent apoptosis. Treatment of cells with intracellular ROS scavenger N-acetyl-l-cysteine also inhibits AGP-induced activation of ASK1, as well as apoptosis. Moreover, depletion of intracellular ASK1 reduces the level of AGP-induced oxidative stress and apoptosis. The evidence for TNF-signaling dependence of ASK1-mediated apoptosis suggests possible mechanisms for AGP activation and regulation of apoptosis-signaling pathways in tumor cells.  相似文献

Purpose

Buildings consume a large amount of energy for space cooling during the summer season, creating an overall sustainability concern. The upfront cost associated with sustainability repels the decision-makers to often end up adopting solutions that have huge operations and maintenance costs. Therefore, the purpose of this study is to assess the lifecycle cost (LCC) implications of optimum configurations of active and passive strategies for reducing the cooling load in buildings.

Methods

Several green building active and passive strategies and technologies were assimilated and their thermal performance in a hot semi-arid climate of Lahore in Pakistan using DesignBuilder V6.1 was simulated to obtain the most optimum cooling load configuration. Furthermore, LCC is estimated, and overall efficiency is evaluated to identify the most effective space cooling configuration.

Results and discussion

The results suggest that a configuration of EPS for external wall insulation, vertical louvers for external shading, 6 mm blue HRG (low-E soft coated)?+?12 mm air space?+?6 mm clear glass for windows, polystyrene as roof insulation, cross ventilation through windows, and LED lighting system has the best performance. This is the first-of-its-kind study in the hot semi-arid climate of South Asia with the city of Lahore in Pakistan as the test case and can be generalized for places with similar conditions. The findings will help the decision-makers in selecting the most load-efficient and cost-effective green building technologies to help improve overall sustainability.

Conclusion

The implementation of the proposed strategies not only aids in providing user-friendly and effective decision-making but also promotes the adoption of sustainability in buildings by leveraging the existing green building technologies to enhance the environmental and economic aspects. This is a promising approach to facilitate the spread of green building construction in developing countries. It is recommended to utilize the strategies grouped in Scenario 8 to achieve a reduced cooling load and LCC of a residential building throughout its lifecycle.

Blood represents a convenient diagnostic specimen for clinical analysis. Serum metabolites and peptides have the potential to serve as reliable indicators of progression from a normal to a diseased state. However, the presence of salts, lipids and high concentrations of protein in blood serum can adversely affect its mass spectrometric profiling, as all of these moieties can hinder the ionisation and detection of diagnostic biomarkers. Several pre-fractionation strategies using chromatographic adsorbents have been employed to desalt samples and remove abundant proteins such as albumin and immunoglobulin. As an alternative to multi-stage fractionation chromatography, we describe in this practical review the adaptation and validation of a simple and fast solid-phase extraction technique using ZipTips. The protocol allows for the purification and concentration of peptides in a few, mostly automated steps, prior to spectral biomarker pattern diagnostics using MALDI MS and MS/MS. It has the added advantages of being suitable for use in a high-throughput and potentially clinical environment, only requiring a few microlitres of serum. We have evaluated, optimised, and standardised a number of analytical parameters ranging from serum storage and handling to automated peptide extraction, crystallisation, spectral acquisition, and signal processing. Using standardised protocols the average CV of serum profiles within- and between-run replicates has been evaluated and found to be around 10 % based on the variability of all detected peaks (more than 100 peaks per profile). The results show that this easy and fast method of using ZipTips, tested over a whole year, is more reproducible and more suitable for serum profile screening than magnetic bead-based methodologies which show higher variability and higher rates of rejected, low-quality spectra upon applying strict data quality control filters.  相似文献
