Atrial natriuretic factor is biologically active also in the absence of vasopressin: studies on Brattleboro-strain diabetes insipidus rats

Since atrial natriuretic factor (ANF) has been shown to inhibit vasopressin secretion, the role of this effect in the acute biological actions of ANF was investigated using Brattleboro-strain diabetes insipidus (DI) rats. Under thiobarbital anesthesia, synthetic rat ANF of a 25 amino acid sequence was administered intravenously as a bolus (8 micrograms/kg) into the jugular vein. The urine volume, urinary sodium and potassium concentration, blood pressure, and heart rate were determined. It was found that ANF administered exogenously can exhibit its diuretic, natriuretic and vasorelaxant activities even in the absence of vasopressin. This indicates that the inhibition of vasopressin secretion is not an indispensible mechanism for acute biological effects of ANF.     

Atrial natriuretic factor inhibits vasopressin secretion from rat posterior pituitary

The effects of synthetic atrial natriuretic factor (ANF) were studied in superfused rat posterior pituitary gland. ANF (10(-6)M, 10(-10)M) significantly inhibited basal as well as KC1 (50 mM) or angiotensin II-stimulated immunoreactive arginine vasopressin secretion. The magnitude of inhibition was greater at 10(-6)M than at 10(-10)M. ANF also decreased cAMP secretion and increased cGMP secretion from the posterior pituitary. These results suggest that ANF directly acts on the posterior pituitary to inhibit arginine vasopressin secretion and that this effect is, at least, partly mediated by the changes in cyclic nucleotide production.     

On the constancy of the evolutionary rate of cistrons

Summary The variations of evolutionary rates in hemoglobins and cytochrome c among various lines of vertebrates are analysed by estimating the variance. The observed variances appear to be larger than expected purely by chance.If the amino acid substitutions in evolution are the result of random fixation of selectively neutral or nearly neutral mutations, the evolutionary rate of cistrons can be represented by the integral of the product of mutation rate and fixation probability in terms of selective values around the neutral point. This integral is called the effective neutral mutation rate.The influence of effective population number and generation time on the effective neutral mutation rate is discussed. It is concluded that the uniformity of the rate of amino acid substitutions over diverse lines is compatible with random fixation of neutral or very slightly deleterious mutations which have some chance of being selected against during the course of substitution. On the other hand, definitely advantageous mutations will introduce significant variation in the substitution rate among lines. Approximately 10% of the amino acid substitutions of average cistrons might be adaptive and create slight but significant variations in evolutionary rate among vertebrate lines, although the uniformity of evolutionary rate is still valid as a first approximation.Contribution No. 813 from the National Institute of Genetics, Mishima, Shizuokaken 411 Japan. Induction of differentiation of cultured human promyelocytic leukemia cells by retinoids

Human promyelocytic leukemia cells (HL-60) were induced to phagocytize, reduce NBT dye(nitroblue tetrazolium), and change into forms that were morphologically similar to mature granulocytes by retinoic acid and related retinoids, but not by the pyridyl analog of retinoic acid. Induction of differentiation could be detected after 4 days of treatment of the cells with retinoic acid at as low a dose as 4 × 10^?8 M. Thus, retinoids may be used in studies on the control of cell differentiation and malignancy of human myeloid leukemia cells.     

Was globin evolution very rapid in its early stages?: A dubious case against the rate-constancy hypothesis

Summary Goodman et al's (1975) claim of accelerated evolution in the early stages of globin evolution is based on an erroneous assignment of the time of divergence of vertebrate myoglobin and hemoglobin. When this is corrected, there is no basis for their claim. The data are much more consistent with the nearly constant rate expected on the neutral mutation-random drift hypothesis than with the uneven rates expected if most amino acid changes were caused by substitution of favorable mutants through Darwinian selection. In addition, the majority of the codons determined by their maximum parsimony method have turned out to be wrong when compared to the actual nucleotide sequences of rabbit and human hemoglobins determined by direct sequencing.Contribution No. 1314 from the National Institute of Genetics, Mishima, 411 Japan.
