Hard conditions of long-term manned spaceflight can affect functions of many biological systems including a system of drug metabolism. The cytochrome P450 (CYP) superfamily plays a key role in the drug metabolism. In this study we examined the hepatic content of some P450 isoforms in mice exposed to 30 days of space flight and microgravity. The CYP content was established by the mass-spectrometric method of selected reaction monitoring (SRM). Significant changes in the CYP2C29, CYP2E1 and CYP1A2 contents were detected in mice of the flight group compared to the ground control group. Within seven days after landing and corresponding recovery period changes in the content of CYP2C29 and CYP1A2 returned to the control level, while the CYP2E1 level remained elevated. The induction of enzyme observed in the mice in the conditions of the spaceflight could lead to an accelerated biotransformation and change in efficiency of pharmacological agents, metabolizing by corresponding CYP isoforms. Such possibility of an individual pharmacological response to medication during long-term spaceflights and early period of postflight adaptation should be taken into account in space medicine.  相似文献

A negative correlation has been established when comparing the data on sister chromatid exchange frequency and DNA repair capabilities of lymphocytes in patients with non-specific inflammatory pulmonary diseases. The role of DNA repair mechanisms in the alteration of sister chromatid exchange frequency in lymphocytes is discussed.  相似文献

Experiments on the transfection of cultured SKOV3 tumor cells of human ovarian adenocarcinoma and HeLa cells of human cervical carcinoma with gene Bax have demonstrated that SKOV3 cells are highly sensitive to the protein product of this gene, whereas the sensitivity of HeLa cells is substantially lower. HeLa cells obtained as a result of Bax transfection and subsequent selection are characterized by an extremely high Bax protein content and a hypersensitivity to doxorubicin. All Bax-transfected SKOV3 cells with an increased Bax content have died. In the SKOV3 cell line, a Bax exon 3 mutation has been found that corresponds to genotype G7/G9, whereas the native type of the Bax gene corresponds to genotype G8/G8. Comparative analysis of the inhibitory effect of modified DNA and RNA oligonucleotides on telomerase activity and tumor cell growth in vitro has been carried out. The study was performed with MCF-7, HeLa, and Mel-10 human cell line extracts. It was shown that PS-TelP5 and PS-TM024 phosphorothioate DNA oligonucleotides inhibited telomerase at nanomolar concentrations and suppressed the growth of tumor cells in vitro.  相似文献

Mouse cytochrome P450 subfamily 1A, 3A, 1E, 2C, 2D isoenzyme activities and corresponding contents were measured by means of triple quadrupole mass spectrometer in Multiple Reaction Monitoring mode (MRM). The technique was developed and tested on microsomes from control mouse and after induction with phenobarbital or methylcholanthrene. MRM allowed us to measure the content of individual P450 isoforms without using isotopic-labeled peptides or derivatization reagent. The results of modifying the content of certain P450 isoforms correlated with the change of enzymatic activity, defined by marker substrates.  相似文献

Mesenchymal stem cells (MSCs) are found in virtually all organs and tissues. These cells can presumably be transformed into tumor stem cells by genotoxic factors and, subsequently, initiate tumor growth. The aim of the present work consisted in analysis of the possibility of malignant transformation of cultured MSCs from the bone marrow (BM) of mice after in vitro exposure to γ-radiation and in the characterization of biochemical and histological features of tumors that developed after the transplantation of BM MSCs to syngenic mice. Two of five mice developed tumors 3 to 4 months after the subcutaneous injection of BM MSCs irradiated at a dose of 1 Gy, five of five animals developed tumors after the administration of BM MSCs irradiated at a dose of 6 Gy, and only one of five mice injected with nonirradiated BM MSCs developed a tumor 6 months after cell transplantation. Telomerase activity in a tumor that developed from BM MSCs irradiated at a dose of 6 Gy was twice as high as that in the tumor that developed from BM MSCs irradiated at a dose of 1 Gy. The histological structure of the neoplasms corresponded to that of multicomponent mesenchymoma, a malignant tumor also termed “a mix of sarcomas.” The tumors consisted of tissue fragments of different histological types. Thus, BM MSCs exposed to 1 or 6 Gy of radiation can be transformed into tumor cells and give rise to multicomponent mesenchymomas, whereas malignant transformation of control BM MSCs occurs much less often.  相似文献

Heat shock protein Hsp70 is known to play an important role in cell protection against a variety of harmful factors. This property, at least in part, is due to Hsp70 ability to restore the native conformation of newly synthetized or damaged proteins. In this activity Hsp70 is accompanied by two proteins, Hdj1 and Bag1, that enable Hsp70 to peform cycles of binding-release of target proteins. The aim of this study was to investigate interactions of Hdj1 and Bag1 co-chaperones with Hsp70 in vivo. The accumulation of Hsp70 was stimulated by heat stress, and later, at certain periods following the stress, cell probes were collected for biochemical and microscopic analysis. The data of Western blotting showed that within 24 h after heat shock amounts of Hsp70 and Hdj1 raised to remain at the elevated level for nearly 48 h. Several time points within this period were chosen for analysis of the complexes between Hsp70 and co-chaperones. The data of reciprocal immunoprecipitation/immunoblotting and confocal microscopy showed that Hsp70-Hdj1 complexes were detected primarily at early stage after heat shock, then Hsp70 was preferably bound to Bag1. The dynamics of chaperone complex formation and changes in their intracellular localization are discussed in terms of cell reaction to stress.  相似文献

The Russian Federation prohibited the distribution of herbal mixtures with synthetic aminoalkylindoles JWH-018 and JWH-073, agonist cannabinoid receptors, on January 22, 2010. The lack or low content of their native compounds in urine requires detailed identification of their metabolites, which are excreted with urine and are present in blood. Using gas and liquid chromatography-mass spectrometry, we identified a series of metabolites in urine samples from humans and rats that were products of the following reactions: (a) mono- and dihydroxylation of the parent compounds with hydroxyl groups located at aromatic and aliphatic residues, (b) carboxylation, (c) N-dealkylation and (d) N-dealkylation and hydroxylation. The prevailing urinary metabolites in humans are monohydroxylated forms, while N-dealkylated and N-dealkyl monohydroxylated forms are found in rats. Twenty-six samples of herbal smoking mixtures with JWH-018, purchased in Russia, were analysed.  相似文献
